This document provides instructions for viewing specimens under a microscope using either transmitted or fluorescence microscopy. For transmitted microscopy, the user moves the reflective slider, rotates the condenser and polarizer, selects transmitted light mode, and adjusts the light intensity before focusing on and aligning the sample. For fluorescence microscopy, the user moves the reflective slider to select emission wavelength, selects fluorescence mode, and focuses on the sample.

CONVENTIONAL MICROSCOPY NOTES
Viewing a Specimen (Transmitted) 1. Move reflective slider to left until at last of three positions. 2. Rotate condenser to either position 'DIC .5-1.4' or 'DIC . 1-.3' corresponding to the numerical aperture of the selected objective. 3. Rotate top polarizer out of light path. 4. Click on 'T' under Conv in Control Panel window. 5. Turn on halogen light source next to microscope and adjust intensity. 6. Focus on sample. 7. Align the condenser. (see transmitted light notes)
Viewing a Specimen (Single Fluorescence)
1. Move reflective slider to left to either middle position or rightmost position. (middle position for red emission / rightmost for green emission) 2. Click on 'FL' under Conv in Control Panel window. 3. Focus on sample.
