C. FUNGI 1.1 Wet Mount


Introduction: Introduction The wet mount is a preparation of a culture to observe a structure of microorganism. Some samples can be place directly under the microscope. However, many samples batter when placed in a drop of water on the microscope slide. The water helps support the sample and it fill the space between the cover slip and the slide allowing light to pass easily through the slide, the sample, and the cover slip. Objective: Objective To introduce the technique of preparation a wet mount for a microscopical examination. To examine three fungi cultures. Materials and Methods Page: Materials and Methods Simple microscope, stain (lactophenol cotton blue), cover slides, cover slips, wire inoculating loop, spirit lamp, fungi cultures ( Rhizopus sp., Aspergillus sp., Trichoderma sp.) Place a drop of stain on a cover slide. Sterilize the inoculating loop, let it cool and use it to gather a thin piece of mycelium with spore from one of the fungi cultures. Cover the drop with a clean cover slip, by placing it at approximately 45 0 angle, with one edge touching the drop, and let go. Observe the preparation under a microscope starting with the low power objective lens 10X followed by he 40X objective lens. Results: Results and Discussion

From the observation under the microscope, we can identify the fungi in the culture. Rhizopus has a rhizoid form, raised elevation and filiform margin. While Aspergillus have a circular shaped, raised elevation and entire margin. The specimen of Trichoderma has circular form, raised elevation and entire margin. We were unidentified the size of specimen because the ocuilar micrometer are not to provide. Conclusion: Conclusion Wet Mount is the way to observe the structure of fungi under the microscope using the specific technique. Its more easily to examine the type of fungi was given. In this experiment, we are unable to observe the sample are given correctly because the sample are culture over than one month, and a lot of spore at surrounding. Question 5: Identify the following structure and draw: (i) hypha (ii) conidiophore / sporangiophore (iii) sterigma (iv) spores - coenocytic or septim - branced or unbranched - present or absent - type, shape and colour
