Jose Gurrola Period 4 Biotechnology DNA Extraction Lab Purpose: To extract DNA from the wheat germ, observe

its qualities and also, to gain experience and knowledge in understanding the principles and properties of DNA Protocol (Flowchart)
To a 50ml tube with 20ml water add 2.5-3 grams of raw wheat germ

Start off the lab Gently rock the tube or mix of a coffee stirrer or small Popsicle stick for 1 minute

Why: To Re-hydrate the wheat germ Using a graduated cylinder transfer pipette add 3 ml of 20% Palmolive soap to the tube with water and wheat germ. Leave the pipette in soap. (Only use half the pipette in this step) Why: To coat each cell with soap. Loosen the cap to allow steam to escape Let it sit for at least 5 minutes. (At 65C as a recommendation)

Why: To let the soap mix better with the wheat germ and separate the DNA Before removing the tube, fill a pipette with 3ml of alcohol. (This should only take a minute or less) Why: You will be using the 3ml of alcohol for the next step, make sure you check that you have the right amount. Take out the tube from bath and then gently add 3ml of alcohol

Why: the reason for this is to filter the DNA allowing parts of it that are hydrophobic a place to escape. Adding gently also prevents the solution from creating mixing together. Watch DNA rise up and then remove the DNA with an inoculation loop and place in microfuge tube. After all this store it in the foam rack. FINNAL STEP: Label your 3.2-3.7ml-microfuge tube and label it to not confuse it with the other groups samples. You have now successfully extracted DNA from wheat germ to test with however you wish.
