Abstract Background: Diabetes melitus (DM) is one of predisposition factor for vulvovaginal candidosis (VVC). The diagnosis were based on clinical examination, potasium hydroxide and
Gram stained. 30-50% of these examinations gave negative result. This condition because of non C.albicans. The media of Candida spp using Sabouraud dextrose agar (SDA) cannot differentiate C.albicans and non C.albicans. Now, media CHROMagarCandida is available to differentiate the Candida spp. There is still argument about species of Candida causing VVC in DM and non

DM. Recently there was thought that C. glabrata was more prominent found in VVC with DM compared non DM.. Objective: To compare Candida spesies causing VVC in DM and non DM. and compare the
result of CVV using potasium hydroxide, Gram stained and CHROMagarCandida.

Methode: In 46 patients who were diagnosed clinically VVC with DM and 46 VVC non DM attempted to Outpatient Dermato Venereology Department of Dr M Djamil Hospital Padang undergoing laboratory examination by potassium hydroxide, Gram stained and culture examination using CHROMAgar Candida medium. Result: In 46 patients VVC with DM, we find C. albicans species 69,9 % and C. glabrata 28,3%. In VVC non DM C albicans 97,8% dan C. glabrata 2,2 %. Statistically there were differences between Candida species causing VVC in DM and non DM (p < 0,05). The age
between 22-70 yo, positive of pseudohypha by potasium hydroxide were 72, Gram stained were 74, and CHROMagarCandida were 88.

Conclusion: Candida glabrata was more prominent in VVC with DM compared non DM. Candida albicans more prominent in VVC non DM compared DM. The media of
CHROMagarCandida is better than potasium hydroxide, Gram stained and SDA media. The advantages of CHROMagarCandida, quicker to identify Candida spp, easier, simpler, cheaper and no need trained person. Key word: vulvovaginal candidosis, species, DM. KOH, Gram, CHROMagarCANDIDA
