Goal:

To evaluate the water quality on the basis of microbiology.

Location:

quality control laboratory.

Time:

When the specimens are received in the lab.

Equipment:Incubator, microscope, Grinder, sterile air hood and autoclave.

Material:

Water, media, glass air, Bunsen burner.

Responsibility:

Procedure

Sampling Bottles
Sterilise the bottles in the autoclave after carefully washing and rinsing to
remove all traces of detergent.
Collect the samples in glass bottles with wide opening and close them with a
round glass stopper or a screw cap.
For dechlorination add 0.1 ml of 10 % sodium thiosulphate solution to each
bottle before sterilisation.
If sample to be taken from a tap open it and allow the water to flow for about
5 minutes before collecting the sample.
Examine for physical changes such as colour, odor, turbidity and smell.

Examine 0.1 ml of water sample. If high bacterial count is expected dilute

the sample.
Place the sample in petri dish and pour the medium cooled to 45 C.
Alternately place the 0.1-ml of sample on the surface of the medium and
spread with sterilises spreader.
Routine media employed are Total Plate count, blood agar, MacConkey,
pseudomonous selective agar .
Incubate the plates at 37 C. Monitor the growth after 24 and 48 hours. Final
counts are to be taken after 48 hours.
