Isomerase Project

No economic evaluations of the various phases of the glucose isomerase
project are required at this time. More research information is required
on the various cell lysis procedures (Manton-Gaulin and mold lysis) and the
enzyme binding data (IRA-904 and Monsanto system). Once this data is obtained,
an economic evaluation should be made of each portion of the project and the
alternatives. This evaluation could be based on enzyme reuse, flow rates and
the engineering scaleup parameters.
The recent reports (RDM, 1970, No. 99 and ROR, 1970, No. 46) are a
summary of the pilot plant data on sonification and DEAE-cellulose as an
enzyme carrier. The sonification procedure is not commercially feasible
(Memo: C. G, Pheil to Mr, S, A. Angotti, October 6, 1970) due to the low
flow rate through the unit. However, it is still used as a control system
to evaluate other lytic systems.
The DEAE-cellulose effort was terminated upon depletion of the enzyme
activity in the pilot plant colums. This support is held in reserve as an
alternate method if no other support is economically feasible. However, the
comercial use of this support is dependent on the development of another
type of process operation other than a column operation. This support material
is also used as a control system to evaluate the binding levels and enzyme
stability of the alternate methods.
