Principio
Los Antgenos Febriles son suspensiones bacterianas especialmente preparadas
para realizar la seroaglutinacin en porta, con el n de detectar la presencia de
aglutininas relacionadas con Brucellosis y Salmonellosis.

Principle
Febrile antigens are bacterial suspensions specially prepared for use in slide
agglutination procedure for detecting specic agglutinins implicated in certain febrile
diseases such as Brucellosis and Salmonellosis.

Negative results can be obtained with patients at the early stages of the disease or
with those under antibiotic treatment.
In some cases, prozone phenomena can be found; thus, when suspecting a high titre,
it is advisable to dilute the sample to 1:20 with physiological saline (NaCl 0.9 %) and
perform the test once again.
Quality control
It is recommended that positive and negative controls be run with the samples to be
processed to check the right performance of the test.
C.- Tube agglutination. Quantitative determination
It is recommended to conrm each positive reaction by the Tube technique.

Conservacin y estabilidad
Los reactivos mantenidos a 2 - 8 C, son estables hasta la fecha de caducidad indicada
en la etiqueta. No congelar.
Muestra
Suero. Puede conservarse a 2 - 8 C durante una semana.
Precauciones
Los antgenos agelares contienen 0,5% de formol y los antgenos somticos 0,5%
de fenol para su estabilizacin, por ello deben manipularse con la debida precaucin.
La eliminacin de los residuos debe hacerse segn la normativa legal vigente.
A. Aglutinacin en porta. Determinacin rpida.Interpretacin de los resultados:
Tcnica
1.- Llevar los reactivos, muestras sricas y controles a temperatura ambiente.
2.- Homogeneizar la suspensin bacteriana con suavidad, incluso el volumen
contenido en la pipeta del cuentagotas.
3.- Dispensar 20 l de suero y controles en los crculos de reaccin. Aadir a
cada uno, una gota de la correspondiente solucin bacteriana.
4.- Mezclar con la ayuda de un agitador desechable, balancear por espacio de
1 minuto y observar la presencia o ausencia de aglutinacin.
Resultados
POSITIVO: Aglutinacin. Equivale aproximadamente
a un ttulo de 1:80 de la prueba de aglutinacin en tubo.
NEGATIVO: No aglutinacin. Ttulo inferior a 1:80.
Tcnica
Segn se esquematiza en el cuadro adjunto, para
cada antgeno disponer, en una gradilla, una serie
de 7 tubos de tipo serolgico (Kahn).
Tcnica
1.- Llevar los reactivos, muestras sricas y controles temperatura ambiente.
2.- Homogeneizar la suspensin bacteriana con suavidad, incluso el
volumen contenido en la pipeta del cuentagotas.
3.- Dispensar 80, 40, 20, 10 y 5 l de suero en cada uno de los crculos de
reaccin. Aadir a cada uno una gota de la suspensin bacteriana.
4.- Mezclar con ayuda de un agitador desechable, balancear con suavidad
por espacio de 1 min. y observar la presencia o ausencia de aglutinacin.
Resultados
Los volmenes de suero usados corresponden aproximadamente a ttulos
1:20, 1:40, 1:80, 1:160 y 1:320 en la prueba de aglutinacin en tubo.
Lectura
Sirvindose de una buena fuente
de luz, observar los tubos,
preferiblemente sobre un fondo
oscuro.

La intensidad y el tono del color de la suspensin bacteriana, puede variar lote a lote.
Esta variacin no afecta en modo alguno los resultados.

Procedure
For each antigen prepare, as indicated in the
following scheme 7 Kahn tubes and place them
in a rack.

Procedure
1.- Bring sample, reagent and controls to room temperature.
2.- Softly homogenize the bacterial suspension including the volume in the
dropper.
3.- Dispense 20 l of serum and each control on different zones of the slide.
Add one drop of the bacterial suspension to each zone and mix with the
aid of a disposable stirrer.
4.- Tilt during 1 min. and observe with the aid of a proper light, for the presence
or absence of agglutination.
Results
POSITIVE: Agglutination. Equivalent to a titre of 1:80 (aprox) by the Tube
technique.
Reading
Using a proper lamp and reading
against a black background,
record the degree of agglutination
as indicated:
++++ 100% of the
microorganisms are agglutinated.
Supernatant is clear.
+++ Approximately 75% of the
microrganisms are agglutinated.
Supernatant is clear
++ Aproximately 50% of the
microrganisms are agglutinated.

NOTE
The tone and colour intensity of the antigen suspension can vary from lot to lot.
This variation will not affect the final results.
Results
The serum volumes dispensed above correspond approximately to titres of
1:20, 1:40, 1:80, 1:160 and 1:320 by the Tube technique.
Quantitative determination
QCA has also available the following reagents, diluted and ready for use:
POSITIVE: Agglutination.
NEGATIVE: No agglutination.
Interpretation of results
Titres equal or higher than 1:80 have probable diagnostic signicance. However, the
precise diagnosis of the disease depends not only on the analytical results but also
on a whole clinical evaluation.
Titre rises considerably between the acute phase and the convalescence.
Performance characteristics
The antigenicity of all febrile antigens have been standardized with control antiserum
of established titer
Interferences: There is no interference by RF at titers lower than 300 IU/ml.
Hemoglobin (10 gL/L); Lipids (10 g/L) or Bilirubin (20 mg/dl) do not interfere the
test
Traces of detergent in the labware may give falsely positive results, as well as the use
of an old or contaminated saline solution.

On peut observer des faux ngatifs au dbut de la maladie ou chez des patients
traits avec des antibiotiques.
Dans certains cas, des phnomnes de prozone peuvent se produire ; si un titre trs
lev est suspect, il est conseill de diluer lchantillon 1:20 dans une solution saline
(NaCl 0,9 %) et de rpter lessai.

Si vous le souhaitez, la dtermination peut tre effectue avec les mmes ractifs
concentrs utiliss lors de la dtermination sur lame selon la mthode indique cidessous.
Conservation et stabilit
Conservs une temprature comprise entre 2 et 8 C, les ractifs sont stables
jusqu la date de premption indique sur ltiquette. Ne pas congeler.

Srum. Il peut tre conserv entre 2 et 8 C pendant une semaine.
Prcautions demploi
Pour garantir leur stabilisation, les antignes agellaires contiennent 0,5 % de
formol et les antignes somatiques 0,5 % de phnol. Ils doivent par consquent tre
manipuls avec prcaution.
Llimination des dchets doit tre effectue conformment aux normes en vigueur.
Technique
Pour chaque antigne, disposer comme indiqu
sur le tableau ci-joint, une srie de 7 tubes de type
srologique (Kahn) sur un porte-tubes.

Technique
1. Laisser les ractifs, les chantillons sriques et les contrles atteindre la
temprature ambiante.
2. Homogniser dlicatement la suspension ractive ainsi que le volume
contenu lintrieur de la pipette du compte-gouttes.
3. Dposer 20 l de srum et de contrles sur les cercles de raction.
Ajouter sur chacun dentre eux une goutte de la solution bactrienne
correspondante.
4. Mlanger laide dun agitateur jetable, agiter pendant une minute puis
observer la prsence ou labsence dagglutination.
Rsultats
POSITIF: agglutination. quivaut un titre denviron 1:80 de lessai
dagglutination en tube.
NGATIF: absence dagglutination. Titre infrieur 1:80.

Rsultats
Les volumes de srum utiliss correspondent approximativement aux titres
1:20, 1:40, 1:80, 1:160 et 1:320 lors de lessai dagglutination en tube.
POSITIF : agglutination.
NGATIF : absence dagglutination.
Interprtation des rsultats
Les titres gaux ou suprieurs 1:80 sont probablement signicatifs. Le diagnostic
prcis de la maladie dpend de lvaluation des donnes cliniques et des rsultats
analytiques.
Le titre augmente considrablement entre la phase aigu et la convalescence.
Fonctionnement et caractristiques de performance du dispositif
La ractivit des antignes a t tablie au moyen dun srum contrle de titre connu
pour chacun dentre eux, car il nexiste pas de matriel de rfrence permettant de
dterminer la sensibilit du produit.
Interfrences: aucune interfrence due la prsence de facteurs rhumatodes na
t observe jusqu des concentrations de 300 UI/ml. La prsence dhmoglobine
( 10 g/l), de lipides ( 10 g/l) ou de bilirubine ( 20 mg/dl) ninterfre pas avec lessai.
Des restes de dtergent sur le matriel peuvent produire des ractions faussement
positives, ainsi que lemploi dune solution saline ancienne ou contamine.

Contrle de qualit
Il est recommand dutiliser un contrle positif et un contrle ngatif avec chaque
groupe dchantillons analyser an de vrier le bon fonctionnement de lessai.
REMARQUES
Lintensit et la tonalit de la couleur de la suspension bactrienne peuvent
varier dun lot lautre.
Cette variation naffecte en aucun cas les rsultats.
