Preparing Medium for Culturing Algae: 1).

Pipette 132 uL of F/2-a and 132 uL of F/2-b per one liter of FSW. *Best to prepare two liters of medium at one time. Medium can be made in two-1 liter flasks and then combined and thoroughly mixed in one gallon jars. 2). Once the medium is thoroughly mixed, pour medium into smaller (250ml or 500ml) flasks for autoclaving. Put ~ 100ml of medium in 250 ml flasks and ~200ml of medium into 500ml flasks. Cover the flasks with aluminum foil. 3). Autoclave medium. Follow instructions on the autoclave. Be sure to choose slow exhaust for cooling phase. 4). Flasks should be removed from autoclave and allowed to cool overnight before inoculation with current algal culture. 5). Pour a small amount of the current algal culture into the newly made algal medium and maintain under fluorescent light to encourage growth. Be sure to label the flasks with the genus of algae and date of inoculation (day/month).
