Catalog Number: Introduction All ustrious pmTFP1-clathrin is a mammalian expression vector that encodes a fusion of mTFP1 N-terminal to human clathrin light chain LCB. It can be a great tool for your research on endocytosis. The properties of All ustrious mTFP1 are superior to those of the commonly used Aequorea victoria variant ECFP and which can be easily detected in the mitotic spindle of mammalian cells using standard CFP filter sets. In addition, the fluorescent signal from mTFP1 is significantly brighter than that from EGFP using standard GFP filter sets. mTFP1 also makes an excellent fluorescence resonance energy transfer (FRET) donor to yellow or orange fluorescent proteins. Clathrin is the main structural component of the lattice covering the cytoplasmic face of the coated pits and coated vesicles in which specific macromolecules are entrapped in the process of receptor-mediated endocytosis. It is a large, soluble protein composed of heavy chains (molecular size about 192 kD) and light chains (molecular size about 32-38 kD). There are two major classes of clathrin light chains identified, LCA and LCB. They are coded by distinct genes and expressed in tissue-specific patterns. Sequence analysis shows an overall homology of 60% between LCA and LCB and the presence of brain-specific insertion sequences (In the brain, tissue-specific mRNA splicing yields larger forms of LCA and LCB, containing additional insertion sequences of 30 and 18 amino acids, respectively). Therefore at least 4 distinct forms of clathrin light chains are found in mammalian cells. Current All ustious Fluorescent Protein Family Members: The founding member is mTFP1. mTFPG3 is a green FP with 3 amino difference from mTFP1. It has a slightly red-shifted emission spectrum and is 1.5 fold brighter compared to EGFP. While being very bright, mTFPG3 can be photobleached within ~5 sec, about 30 times faster than EGFP, suitable for certain cell-based assays that require a bright FP with very short half-life. mTFP0.7 is a precursor during the evolution of mTFP1. It has photo-switchable properties like Dronpa that cycles between fluorescent and nonfluorescent states. It may be developed into components in PALM/SIM applications.
