INHIBITION
Mostly enzyme inhibitors act reversibly , they do not cause any permanent changes in the enzyme. However, there are also irreversible inhibitors that permanently modify the target enzyme.
INHIBITION
The mechanism of action of an ihibitor its inhibiton type can be determined by comparing the kinetics of the inhibited and uninhibited reactions. Based of the different kinetics , inhibition can typed to :
Competitive inhibitors Uncompetitive inhibitors Noncompetitive inhitors
TYPES OF INHIBITION
TYPES OF INHIBITION
Competitive inhibition
There is competition between the substrate S , and the inhibitors I for the enzyme.
If an inhibiting substance can bind at enzyme active site. Enzyme that binded inhibitors can not bind the substrate or the other way
KINETICS
Non-competitive inhibition
The inhibitors is persumed not to bind to an active site on the enzyme.
But rather to bind at some other site. This complex formation involve some changes confromation of the enzyme. Makes it impossible for the substrate to bind in active site.
TYPES OF INHIBITION
Uncompetitive inhibition
Inhibitors combines reversibly with the enzyme-substrate complex after it forms. Its assumed that this complex is so stable that it does not lead to formation of the expected product. If this were not so, this case would reduce to a special case of noncompetitive inhibition.
KINETICS
COMPETITIVE
NONCOMPETITIVE
UNCOMPETITIVE
Conclusion
This leads to a decrease in the rate of the reaction not only because some substrate is consumed, but also because of the decrease in the effective concentration of the enzyme
HOMEWORK
Sketch a Lineweaver Burke plot for this enzyme in the presence and absence of inhibitor and report values for the table below given the following equations : Vmax app Kmax app V max Km
No inhibitor inhibitor y = 1.52x + 1.51 y = 1.58x + 4.27 0.662 1.006 0.233 0.370
What the type of inhibitor, justify your respon and explain what about the mode of inhibitor binding.
