DETECTING E.COL1-0157 WITH FIBER-OPTIC BIOSENSOR

INTRODUCTION
Escherichia coli is a kind of bacteria, and it will cause the enterohemorrhagic E.coli infection. Though the types of E.coli that cause this kind of infection are many, most of them have no other dangerous complications. But there is one type E.coli-0157: H7, will not only cause the

enterohemorrhagic E.coli infection, but also bring about the dangerous hemolytic uremic syndrome that might cause human death. So, it is important to build up a system for rapid identification of Escherichia coli in clinical.

METHODS AND MATERIALS


We use a 488nm argon guide laser and guide into an objective to match the numerical aperture of the fiber and let the light propagate by total internal reflection. A 5cm length region of fiber had been cladding by ethyl acetate and the <antibody-antigen-FITC labeled antibody> sandwich structure were immobilized on this region and will be excited by the evanescent wave, then release 520nm fluorescence. The fluorescence signal will be detected by the PMT. PMT will send the signal to the lock-in amplifier and analyze them.

RESULTS A N D DISCUSSION
Here we prepare four different kinds of E.coli subtypes, and the results show that only E.coli-0157: H7 will react with the fiber-optic biosensor and cause the signal intensity change. That means our system has the specific binding affinity with the target sample. Except the quantitative and qualitative analysis, we also calculate the kinetics. We have four different concentrations of IgG samples, and the results show that the and 4.4 x lo" s-'.
