1. (30 points) An enzyme C3P0 is being tested for its heat stability as an additive in a laundry detergent formulation to remove hard-to-remove stains especially oils and fats from food and grease. This test can also predict the integrity of the enzyme during manufacturing of the detergent in which process temperatures may degrade the enzyme leading to deactivation and makes the formulation ineffective. The reaction is described as;

fats fattyacids + glycerol

k d = 30 .e R.T , [min 1 ]; Ed = 5 Kcal / mole
Km of the enzyme is 5 mM (M = gmol/L). At 600C, lipase is subjected to deactivation with a half-life of 10 min. Fat hydrolysis is carried out in a batch reactor which simulates a top-loading washing machine. Initial fat concentration is 150 gmol/m3. a. b. c. (10 points) How long does it take for the enzyme to hydrolyze 80% of the fat present? (Hint: solving differential equations can be either to get general or particular solution) Ans. _______ min.
Ed

(10 points) From (a), what is the reaction velocity? Ans.__________ gmol/m3-min. (10 points) Name the enzyme according to proper naming and classification. Ans.________

2. (30 points) You are to screen 3 different strains of the yeast, Saccharomyces Achillesiae with different yield coefficients using the general stoichiometric reaction below;

a.C6 H 12O6 + b.O2 + c.NH 3 C6 H 10O3 N18 + d .CO2 + e.H 2O + f .C2 H 5OH
BTI-393 BTI-847 BTI-456 a. b. c. YX/S = 0.7 g/g YX/S = 0.11 g/g YX/S = 0.05 g/g

(10 points) Determine which strain produces most ethanol and calculate the ethanol concentration produced using 50g/L of glucose. Ans._______________ , ______(g/L) (10 points) As an objective to produce the chosen strain of the same microorganism, calculate the oxygen concentration to supply if desired dry cell yeast concentration is 25 g/L. Ans.________(g/L) (10 points) From (b), calculate the respiratory quotient. Ans.________

3. (40 points) The chemostat growth of a microorganism on glucose-limiting substrate gave a cell growth rate of:

1 dX 4.S . = , where its unit is g cells/m3-hr; S in [g/L] and with YX/S = 0.5 g/g, X dt 5.(S + 5)
d. e. f. g.

So = 25 g/L.

(10 points) For a recycle ratio of 0.15 and with recycle cell concentration of 90 g/L, calculate the cell concentration in reactor if dilution rate is 0.1 hr-1. Ans.________(g/L) (10 points) From (a), determine the fraction of cells returned to the reactor. Ans._______ (10 points) From (a) calculate the productivity. Ans.__________(g/L-hr) (10 points) Calculate the cell concentration for an optimum setting without recycle to match the productivity in (a) Ans.______(g/L)

"I pledge on my honour that I have honestly accomplished this examination without the aid of anyone nor have given information to others to violate the examination code of conduct of the UST faculty of engineering. I believe that doing my best effort with the right mindset and attitude will fulfill my highest ideals as a student of ChE513 Biochemical Engineering and of this beloved royal and pontifical university."
