BIO 206 DOWNSTREAM PROCESSING IN BIOTECHNOLOGY- I

ASSIGNMENT - II
1. Calculate specific resistance of cake and resistance of filter medium on basis of the following
data for a constant pressure filtration. The mass of solid deposited per unit volume of filtrate
was 30 kg/m
3
on a filter medium of area 1 m
2
. The pressure drop was 2 bar and viscosity of
filtrate was 1.1 10

2. Time-pressure drop data for a constant rate cake filtration process is given below
Time (s) 0 10 20 30 40 50
P (kPa) 5 10 15 20 25 30
If we are to carry out the filtration at twice the filtration rate, predict the pressure drop
required after 2 hours of filtration. Assume that the cake is incompressible.

3. A lab scale filtration experiment of slurry with a solids content of 24.6 g/L on a filter of area
of 5 cm
2
at a pressure drop of 500 mm of Hg gave the following data. The filtrate had a
viscosity of 1 cP.
Time (s) 6 10 16 23 31
V (cm

Calculate the time required for filtration of 2 m

of the same feed slurry on a plate and frame
filter press consisting of 15 frames , each of area 30 cm

.

4. You are filtering beer containing 6% citric acid on a continuous rotary vacuum filter. The
filter has an area of 18.1 m
2
, a negligible medium resistance, a cycle time of 75 sec, and a
pressure difference of close to 1 atm. The cake which forms has a washing efficiency of only
60%, but it is incompressible and permeable:

The cake retains 7% of the filtrate leaving and should be washed until the cake contains only
10% citic acid originally entered. Calculate the filtration time and the washing time required
to process 3000 L/hr of beer.

5. Many animal cells can be cultivated on the external surface of dextran beads. These cell-
laden beads or microcarriers have a density of 1.02 g/cm
3
and a diameter of 150 mm. A 50-
liter stirred tank is used to cultivate cells grown on microcarriers to produce a viral vaccine.
After growth, the stirring is stopped and the microcarriers are allowed to settle. The
microcarrier-free fluid is then withdrawn to isolate the vaccine. The tank has a liquid height
to diameter ratio of 1.5; the carrier-free fluid has a density of 1.00 g/cm
3
and a viscosity of
1.1 cP. Estimate the settling time by assuming that these beads quickly reach their maximum
terminal velocity.
