Detection: The L-2400 UV
and L-2420 UV-Visible
Detectors
Both detector types are designed for highest sen-
sitivity by utilising the double beam photometric
detector principle in combination with a grating
monochromator. To improve light intensity and
detection sensitivity in the visible range, the
L-2420 UV-Visible detector contains a tungsten
lamp in addition to the deuterium lamp. Detection
wavelength is time-programmable for optimum
detection sensitivity at the absorbance maximum
of each analyte. If negative peaks should be
recorded and integrated, e.g. applying indirect UV
detection for ion chromatography, the polarity
of the detector signal can be reversed. In
stopped-flow mode spectra of the analytes
can be recorded and background-subtracted for
proper substance identification and selection of
the optimum detection wavelength. Both detec-
tors can be operated either by integration into
the LaChrom Elite system with control through
the EZChrom Elite chromatography data
system or as stand-alone modules via the
optional keypad. In stand-alone mode, several con-
trol functions using contact signals are available.
Highest Sensitivity
and Resolution
Advanced optics and innovative electronics com-
bined in the LaChrom Elite UV-Visible detectors
ensure minimised RI effects, a large linear
dynamic range and superior signal to noise
even with the fastest response times. So lowest
trace level detection limits and better quantitative
reproducibility are achieved. The low baseline
drift makes signal detection insensitive to varia-
tions in ambient conditions and leads to optimum
peak integration. A small optical bandwidth ensures
high spectral resolution of the detectors.
Fit for Fast Chromatography
and Micro Applications
The active noise filter combined with a wide
range of response times, enables optimum
peak data acquisition even in high-speed chro-
matography, e.g. using Chromolith columns.
Narrow, rapidly eluting peaks can be recorded
with the highest signal to noise ratio whilst main-
taining peak shape. For all application ranges, from
micro up to semi-preparative chromatography,
different optional pre-aligned and easy-to-install
flow cells are available.

The LaChrom Elite Diode Array Detector is
designed as a single beam photometric detector
with a grating monochromator. It is equipped
with a 1024 bit diode array element to sup-
ply the highest spectral resolution over the
complete wavelength range. For light sources the
detector contains both a deuterium and a
tungsten lamp to ensure maximum sensitivity
even in the visible range. The selectable optical
slit width and electronic diode bunching opti-
mise spectral resolution and sensitivity as
required for each specific analysis. The setting of
the electronic noise filter is coupled to the data
acquisition period. This effectively suppresses
noise whilst at the same time accurately
records peak shapes, particularly fast eluting
peaks in high-speed chromatography.
3-dimensional chromatographic data is recorded
and processed in combination with the EZChrom
Elite chromatography data system.
The following advanced 3D functions

Convenient Maintenance
All maintenance parts are accessible from the
front of the instruments. Therefore, the user can
easily carry out exchange of the detector lamps and
cleaning or exchange of the flow cell.
FDA, GMP, GLP functions included
All detector types are equipped with a mercury lamp for automatic wavelength calibra-
tion. Using the different spectral lines of deuterium and mercury lamps, wavelength calibration
can be performed over the whole spectral range of the detectors. In contrast to calibration
filters (e.g. Holmium oxide filters), which have to be recalibrated themselves periodically, the
spectral lines of the lamps remain completely constant with time and need not to be recali-
brated. On the other hand, for wavelength and absorbance calibration with standard solutions
according to the European Pharmacopoeia, an optional cuvette holder can be supplied.
The detectors contain a maintenance logbook, which records the exchange date of the
lamps, the lamp energy, the total lamp on time and the number of lamp on switchings.
This facilitates regular maintenance and essential documentation. In combination with the
EZChrom Elite chromatography data system, the complete range of GxP-related
functions becomes available e.g., recording of the instrument serial number, hardware
logbooks and audit trails, automatic documentation of the instrument parameters, etc..
When connected to EZChrom Elite software, full compliance with the FDA rule 21CFR Part
11 is achieved. Moreover, module qualification (OQ) can be substantially facilitated using
the optional Auto-Validation software.
