Ph.D. CCMB, India. Doctoral thesis Molecular status of Detoxification genes


and functional proteomics in recurrent early pregnancy loss (REPL)
Supervisor: Dr. Lalji Singh, Director, Centre For Cellular and Molecular
Biology, Hyderabad.
Description: REPL is a heterogeneous condition experienced by about 20%
couples and in half of the cases the reasons are unknown. Present study is aimed at
dissecting out the molecular genetic markers responsible for REPL in South Indian
women. Mutational screening of genes involved in detoxification, estrogen
metabolism, blood homeostasis, vascular activity etc provided us with exciting
results with a strong association between the risk of recurrent miscarriages and the
polymorphic markers studied was found. In addition, a number of novel
polymorphisms that were not reported earlier had been discovered. This data is
valuable for appropriate model development to assess the risk associated with a
particular or combination of alleles.
Another part of the study is targeted at the analysis of blood platelet proteome and
a comparison between normal and REPL females to fish out the candidate proteins
that can be used as diagnostic markers. Platelet proteome showed consistent
differences between cases & controls and the identification of the proteins by
MALDI-TOF and peptide mass fingerprinting was done.
Techniques Used: DNA isolation from blood, PCR primer design, synthesis and
amplification of DNA, RFLP, DNA Sequencing using ABI-3730 automated
sequencer, Sequence analysis using Gene tools, Mutant phenotype prediction using
Transcription factor binding site search, phylogenetic footprinting analysis,
comparative modelling. Statistical analyses include Significance tests, Correlation
and regression, power calculations. Protein analytical techniques included Blood
cell separation in a dextran gradient, platelet protein preparation, quantitation,
Isoelectric Focussing in pre-cast as well as manually developed pH gradient gels,
SDS-PAGE, staining to visualize the protein spots, image analysis using PDquest
software from BIORAD, preparation of peptides from gel spots for MALDI-TOF

MS analysis, analysis of MALDI spectra, database searching to find out the


peptide mass fingerprints.

M.Tech Dissertation: carried out for a period of six months at Centre for
Biotechnology, Anna University, Chennai
Title:Multivariable optimization of poly--hydroxy butyrate production by
Azotobacter beizerinckii by the use of response surface methods.
Supervisor: Prof. Kuntala jayaraman and Dr. R.Sachidanandham
Description: Azotobacter beijerinckii is known to grow and produce the
biodegradable plastic, poly--hydroxy butyrate (PHB) using defined medium. In
the present study, we developed and optimised a process for the production of PHB
from molasses and Corn steep liquor (CSL) as the carbon and nitrogen sources.
The growth profile of A.beijerinckii was monitored and the product harvesting time
was optimised. Experiments were designed using the full factorial design and the
results were analysed by the 2D and 3D response surfaces. A mathematical model
was developed in terms of total Carbohydrate and Nitrogen content to predict the
yield of PHB. This was used to calculate the optimum molasses and CSL
concentrations that maximize the yield.
Techniques Used: Basic microbiological methods such as culture maintenance,
revival, media preparation, sterilisation, staining, microscopic examination;
Bioreactor operation (1lit, 2lit), shake flask batch experiments, PHB extraction
using a solvent extraction method, using STATGRAPHICS plus for WINDOWS
for experimental design, regression analysis, Design Expert software for studying
the response surfaces.

WORK EXPERIENCE

As a Scientist at Dr.Surapanenis Genomic Solutions Pvt Ltd since September 2007 till
date. Job responsibilities include gene isolation, cloning, and expression of protein in
prokaryotic system, protein purification and modification (fluorescent tagging, enzyme

coupling).
TEACHING EXPERIENCE

An experience of two years in Biotechnology and Biochemistry for postgraduate and under
graduate students at V.S.Lakshmi Womens College, Kakinada, an affiliated centre to Andhra
University. Subjects taught were Intermediary Metabolism, Enzymology, Clinical
Biochemistry, Molecular Biology, Industrial biotechnology, Immunology.

RESEARCH EXPERTISE

Biochemistry: Protein purification utilising the affinity chromatography technique, protein


estimations, Western Blotting, ELISA, Polyacrylamide Gel Electrophoresis, 2-D gel
electrophoresis, image analysis using soft wares like PDQUEST, Proteomic analysis using
techniques like MALDI-TOF. Qualitative and quantitative analysis of various classes of bio
molecules, enzyme assays and kinetic studies. Evaluation of clinical parameters with special
emphasis on biological fluids.
Molecular biology: Isolation of DNA and RNA from cells and tissues, PCR, RT-PCR,
agarose and polyacrylamide gel electrophoresis, DNA sequencing and Gene scan.
Microbiology: Culturing of bacterial cells, Staining, basic Bioreactor operations (up to 20L
level). Downstream operations involved in product purification.
Computational Skills: Working knowledge of computational tools such as SPSS, Epistat,
Statgraphics, MATLAB, Arlequin, GOLD, Swiss PDB modeller, Clustal, Sequence
assembling using Auto assembler, Gene tool. Programming knowledge in computer
languages such as C and PERL with orientation towards biological applications.
