The overall goal of dentistry is to have a healthy and functional dentition for a lifetime.
Throughout our lifetime, we have two sets of teeth the primary dentition (Total of 20) and the
permanent dentition (Total of 32)
On the basis of form and function the teeth of the human dentition are divided in to four types.

Components of Preventive dentistry


1. Patient education
The role of the dental professional in patient education is to motivate patients to value their oral health.
2. Fluoride
Most dentists consider fluoride to be the most reliable therapy for the prevention of dental caries.
3. Sealants
Sealants are most frequently applied to the difficult to clean occlusal surfaces of the teeth, Decay causing bacteria are
prevented from reaching occlusal pits and fissures.
4. Plaque Control
Dental Plaque is the primary cause of dental caries and periodontal disease.
Patients cannot remove all of the plaque everyday but they can keep it under control by brushing, Flossing, using inter dental
cleaning aids.

Tooth Brushing technique


1. Place the head of a soft brush parallel with the occlusal plane with the brush head covering three to four teeth ,
beginning at the most distal tooth in the arch.
2. Place the bristle at the gingival margin pointing at a 450 angle to the long axis of the teeth.
3. Exert gentile vibratory pressure, using short back and forth motions without dislodging the tips of the bristles , this force of
bristles ends into the salci , as well as in to interproximal embrassares and should produce precipitate blanching of the gingiva.
4. Complete several strokes (20) in the same position.The repetitive motion cleans the tooth surfaces , concentrating on the apical
1/3 of clinical crowns.
T he gingival sulci and as far as on the proximal surfaces as the bristles can reach.
5. Lift the brush, move in to the adjacent teeth and repeat the process for the next three or four teeth.
6. Continue around the arch brushing about three teeth at time. Then use the same method to brush the lingual surface.
7. If the brush is too large to reach the lingual surfaces of the anterior teeth, It should be turned vertically to press the end of the
brush into the gingival sulcus area.
8. After the completing the maxillary arch, move the brush to the mandibular arch, and brush in the same organized way to reach the
all teeth.
9. Brush the occlusal surfaces of three or four at time by pressing the bristle firmly into the pits and fissures and brushing with several
short, back and forth strokes.
10. Brush the tongue.

Ployacrylamide

gel electrophoresis is an
analytical technique used to separate proteins on
their molecular weight basis or net charge. In this
particular method protein molecules move
through a semi-solid matrix of acry-amide and
bis-acrylamide within electric field. Molecular
mobility is inversely proportional to the friction
caused by matrix.

1. SDS PAGE - Used to separate molecules
based on their molecular weight.
2. Native PAGE - Used to separate molecules
based on their net charge.

There are two main types pf gels


1. Continuous gel
2. Discontinues gel

The continuous gel contains a gel slab with equal


composition. Proteins move though the gel and
get separated. The discontinuous gel contains
two parts known as stacking gel and resolution
gel. The stacking gel is less cross linked (4-5%)
and protein molecules move with a less
resistance. Purpose of this stacking layer is to
maximize protein resolution by concentrating
samples in a ultra thin layer (1-100 mm) before
entering to the resolution layer. Cross linking of
resolution layer change from 6% to 15%. This
layer resolves protein molecules, When moving
through thee matrix.

In SDS PAGE protein samples are mixed with


SDS (Sodium Dodecyl Sulphate) to denature the
protein without breaking its peptide bonds. SDS
binds to and increase the proteins charge/ mass
ratio giving the proteins a net negative charge.
In Native PAGE structure of the protein molecule
is not denatured.

The gel is casted according to the requirements

Finally the gel tank is filled with running buffer


(Native or SDS - PAGE) and correct current and
voltage is provided through the electrophoresis
power supply unit.

The sample start moving downwards and the


movement can be observed by the dye front.
When the dye front reaches the end of the glass
plates power is disconnected and the gel is
removed from glass plates to stain.
Usually 45 minutes shaking in CBR - 250
solution and destaining overnight is sufficient to
visualize protein bands.
Alternately microwave staining and destaining
can be done and protein bands can be visualize
within 15 minutes.

Protein samples are mixed with sample buffer in


2 : 1 ratio. In SDS-PAGE, the samples are
denatured by immersing sample tubes in a water
bath at 95 C for 3 minutes. Samples are then
loaded to the wells in the gel.

former General Secretary and committee member of Standing committees I have
rendered my efforts and services to the SLUTOU during the past years in various ways
for the upgrading of the union activities.
The journey from the inception of our union, was not a flowery path. Many obstacles
and difficulties constantly barred us from achieving what we have achieved today.
However all these achievements were possible due to the untiring efforts of our
pioneer members. Our members contribute their valuable technical skills and talents to the university
teaching, education, research, health sector and also to the public. There is no doubt that the service of the
technical officer plays a vital role in the university laboratory system. Hence the technical officers are
recognized in their fields both locally and internationally.
Further the university technical officers contribute to enhance the quality of university education through
their services. I am assured that the SLUTOU News Bulletin provides a pathway to all our members to reach
the whole world through their skilled performances.

Angulugaha gamage Punyalatha studied


at Meepe Amathayana school and
Southland College Galle. in her primary
and secondary education. She Joined to
the Kelaniya university at 1978 as a clerk
grade 11 and promoted to the trainee
Technical officer seg B in 1980 . On the date of. 01 03
1984
she was transfered to he department of
Pharmacology university of Ruhuna. as a Technical
officer she joined to the SLUTOU in 1986 with the first
batch of members of the faculty of Medicine Galle.
She dedicated her valuable service to the SLUTOU as a
faculty representative for five years and as a University
secretary for one year.
She has rendered her service the faculty of Medicine as a
member of Student Support Services and the Drugs for
diseases committee of the medical exhibition2006 . She
has served in the department of Physiology in the two
years and assisted to the final M B B S examination in the
department of Gyn/ Obstratic
as addition to her
dutiesand participated to several work shops.
Mrs P. Gamage is a friendly and active trade
union activates and much interested in house keeping and
social affairs.
The end of her service is a great lost to the union activities
and to the University of Ruhuna

Arumapperuma Somasiri studied at
Telijjawilla central and Godapitiya Maha
Vidyalaya Matara. He entered to the
Ruhuna university service on 17th June
1985 as a trainee Technician. Since then
he served at the department of the
Agricultural economics at the Faculty of Agriculture of
the Ruhuna university. He obtained the requied training in
the field of Audio visual from the department of the
Agriculture at Gannoruwa Peradeniya.
He was the member of SLUTOU from 1986 with the first
batch of members of the faculty of Agriculture and served
as the Faculty representative in several occasions. He had
dedicated his long service of twenty eight years at the
some institution and received awards for the efficient
service and duties at the Faculty annual symposiums. He

got returned as a staff Technical officer (grade1) on 04
November 2013.
Mr. A Somasiri is a friendly and active trade union
activates and much interested in farming and social

hospital peradeniya in 5th November 2000 as a
trainee Technical officer and during this period he
gained remarkable experience regarding work
associated with Medical Laboratory.

Also he has an experience as a visiting lecturer in


the Faculty of Allied Health Sciences, University of
peradeniya since 2008 and has acquired abilities in
both teaching and practical in fields of Laboratory
Management, Biochemistry, Haematology and
Clinical Pathology.

Simultaneously he registered for Bachelor of


Science at the Open University Sri Lanka and
completedhis first degree in 2005 with a Second
Class Lower Division. After the first degree Dr.
Jayasekara enrolled in research related to the
chronic kidney disease under the supervision of
Professor Dhammika Menike Dissanayake in
addition to his routing duties. He extended
research activities by enrolling for a research based
M Phil at Faculty of Medicine, University of
Peradeniya in early 2009 and the Faculty extended
M Phil to a direct PhD according to the progress
gained through hisresearch carrier. Accordingly Dr.
Jayasekara completed PhD in the field of
Biomedical Sciences (affective date 06 Feb 2014).

We wish him all the very best for his future


endeavours.

The title of the PhD thesis is Epidemiology, early


kidney injury markers and the role of G6PD
deficiency in the pathogenesis of chronic kidney
disease in North Central Region of Sri Lanka The
findings of the above studies were published in
environmental toxicological symposiums in India,
China, Vietnam and Malaysia in 2008, 2009, 2010,
2011 respectively.

MESSAGE FROM THE GENERAL SECRETARY / SLUTOU


I have the great pleasure in sending this message to the News Bulletin of the Sri Lanka Universities'
Technical Officers' Union (SLUTOU) being the General Secretary of the SLUTOU for the year 2013/2014.
It has been necessary to reconsider and think about the function and the duties of the University Grants
Commission (UGC) since there are some officials who do not want to settle the problems and issues related to us.
Such officials create new problems instead of settling the remaining grievances. Therefore, we should work in an
understandable way for minimizing issues.
During this year, we were able to have a two fruitful discussion with the Chairperson of the UGC, and
she has agreed and promised to solve many issues related to the SLUTOU. I take this opportunity to extend my
sincere thanks for Prof. (Mrs.). Kshanika Hirimburegama, Chairperson/UGC and Mr. Samantha
Wickramarachchi, Personal Secretary of Chairperson for the provision of a discussion. Therefore, it would be the
responsibility of the new Committee of Management to negotiate with the UGC for early implementation of such
promises for the betterment of the members of the SLUTOU.
This year too we were able to organize a valuable workshop for our members enabling the update of their new knowledge and
skills. The services of Mrs. Amara Wijeratne, Vice President/SLUTOU and Mr. K.S. Wijenayake were exemplary for the organization of the
aforementioned workshop.
My sincere thanks are extended to the President, Vice President, Treasurer, Editor and all members of the Committee of
Management who helped me during my tenure as the General Secretary for the year 2013/2014. I would like to extend my special thanks to
Mr. W.V. Lakshman Kumara, Assistant Registrar of the University of Peradeniya for his valuable clarifications on various issues and regular
guidance for handling and settling the issues of the members in a balanced way. Further, I would like to extend my sincere thanks for Mr.

Vietnamese time bomb


At the end of the Vietnam War, American soldiers returned to the
USA thinking that they were safe. But some of the soldiers were
carrying in their bodies a bacterium that would suddenly emerge,
years or even decades later, causing disease and even death. As
the illness developed long after the initial exposure, with no
advance warning, it was nicknamed the Vietnamese time bomb.
What is this mystery disease and is it found in Sri Lanka?
Burkholderia pseudomallei is a saprophytic bacterium found in
soil in tropical countries. It infects humans accidentally, during
contact with soil or water, via inoculation through the skin or by
inhalation and ingestion. Occupational exposure to mud and
surface water is a risk factor. The infection caused by B.
pseudomallei is called melioidosis. Melioidosis, also known as the
"Vietnamese time bomb", received wide attention largely after the
Vietnam War. As helicopters deposited troops throughout the
tropical nation, their blades kicked up dirt, exposing soldiers and
pilots to hidden pathogens in the soil. In modern times it is found
mainly in people with occupational exposure to soil such as rice
farmers and construction workers.
Melioidosis is unusual in many ways. It can have a very long
incubation period, up to even 50 years after exposure. Also,
melioidosis tends to cause a wide range of symptoms that
resemble many other diseases, leading some scientists to give it
a second nickname "the Great Mimicker.
Melioidosis commonly infects the lungs causing pneumonia. The
next common presentation is abscess (cavity of pus) formation.
More severe disease, even sepsis causing death, is seen among
patient with diabetes, chronic kidney disease or liver disease. The
antibiotic of choice is ceftazidime or meropenem. The mortality of
melioidosis can be 20 to 50% even with treatment.
Melioidosis is endemic in the tropical and sub tropical regions of
South East Asia and Northern Australia. The first cases of
melioidosis were documented in Burma by Alfred Whitmore and C
S Krishnaswami in 1912. The first case in Sri Lanka was reported
in a European tea broker in 1927 but since there were very few
cases reported until 2006 Sri Lanka was considered as non
endemic for melioidosis. Since 2006, however, more than 50
cases have been reported from all parts of the country and it is
now recognized as an emerging infection in Sri Lanka.

How to Identify this bacterium


B. pseudomallei is not fastidious and will grow on a large variety of
culture media (blood agar, MacConkey agar, CLED medium etc.). It
can be isolated in pure growth from sterile sites and in mixed
growth from sputum. Ashdown's medium is a selective medium.
The organism grows more slowly than other common bacteria.
On blood agar, colonies are pin point with no
haemolysis at 24 hours. After 48 hours haemolytic chalky
coloured colonies with silvery metallic appearance in the well can
be seen. After one week the colonies appear umbonate (raised
center) and gradually develop wrinkling. The isolate has a
characteristic earthy smell.

On MacConkey agar colonies are pin point and have a nonlactose fermenting appearance at 24 hours. After 48 hours they
become the bright pink colour of a lactose fermenter!

Gram stain
B. pseudomallei is a Gram-negative bacillus with a characteristic
"safety pin" appearance (bipolar staining. It is motile, gives a
positive oxidase test and does not form spores .On sensitivity
testing, the organism appears highly resistant. It is innately
resistant to a large number of antibiotics including colistin,
polymyxin and gentamicin but, unlike other pseudomonads, it is
sensitive to co-amoxyclav.

Laboratory identification of B. pseudomallei can be difficult as few
technicians are familiar with the isolate. Even if they identify it as
an oxidase positive, non-fermenting bacterium they may report it
as Pseudomonas sp.
Many laboratories fail to perform the oxidase test on isolates that
appear lactose fermenting on MacConkey agar. In addition, the
oxidase test may be weakly positive when performed on a single
colony. It is more reliable if performed from the well.
Pellicle formation in broth, safety pin appearance on microscopy
and wrinkling may lead to misidentification as environmental
contaminants. Colony variants such as serous and mucoid
colonies are seen and a single strain may display multiple colony
types, so inexperienced laboratory staff may mistakenly believe
the growth is not pure and it may be discarded as a mixed or
contaminated culture.
Commercial identification kits such as API NE or Rapid NF are
unable to identify this bacterium accurately calling it B.cepacia or
P comomonas.
Therefore it is important that microbiology laboratories should
obtain a known culture and familiarize all the technical staff with
the cultural and microscopic appearance of this bacterium.

Honorary Award for Outstanding Services to the SLUTOU - 2014

Sunil Ananda Gurugamage


Sunil Ananda Gurugamage, known to many of us simply as Gura. His Alma mater is prestigious
Mahinda College. After schooling he attended Twikenham Polytechnical Institute of London,
UK. Then he joined Almulla Construction Company limited in Dubai to construct a multi storey
building, and to put up air port run a way and the terminal of Sharjah in UAE. He gained
professional experiences during this period. Finally he came back to Sri Lanka and joined a huge construction
company named Ballast Nedam, at the capacity of an electrical engineer.

On 17 September 1984, he was appointed as the Superintendent of Works in the University of Ruhuna and
by 1996 he was promoted to the post of special grade that was categorized under the university executive grades (A04).
By 1990, he was serving to the Ceylon University Clerical And Technical Assistants' Association
(CUCTAA) by now the Sri Lanka Universities Technical Officers' Union (SLUTOU) as a committee
member. In 1991, when I was appointed as the University Secretary of Ruhuna for SLUTOU, I unilaterally decided
to get enrolled him as a member of SLUTOU.
In 1994, he was the chief organizer of AGT and AGM held at the Auditorium of Faculty of Medicine, Galle.
He served as the Vice President of SLUTOU in 1996 and 1997 and was very active and instrumental in the trade
union action SLUTOU launched at which UGC issued circular no 605.
Mr. Gurugamage was unanimously nominated by IUTUF as the convener & caller for a newly empowered
committee of four persons representing all non-academic unions to be conferred with the honorable Minister of
Education & Higher Education, Dr. Richard Pathirana.
As a result of the discussion, the SLUTOU committee in which Guru was as member, had with the Minister,
Deputy Minister of Higher Education (Prof. Vishwa Warnapala), Secretaries of Ministries of Higher Education,
Labour & Finance, Auditor General, Attorney General, Chairman & Secretary of UGC, issued another circular No.
712 by granting salary increase.
During last 09 years he held the responsibilities as University Secretary of Ruhuna for 2005 / 06 and 2011
/12, Agriculture Faculty Representative of Ruhuna for 2009 / 10 respectively. In 2006, Mr. Gurugamage was called
th
upon to conduct the entire program as the commentator at the ceremony held for commemorating 50 anniversary of
SLUTOU.
During his tenure he molded and guided many young professionals. He did his duty with many distinctions
for approximately twenty years.
Dear Mr. Gurugamage, the difference you make is nothing short of a legend. Finally, let me congratulate and
wish you all success!!
