MONERA’S KINGDOM

Basic Competence

3.5 To description about the characteristic of Monera’s Kingdom

and to communication the role of Monera’s Kingdom in the
live.

OPENING

Items

Monera’s Kingdom is organism’s member of prokaryotic, that

are organism which don’t have membrane of nucleus cell. In general,
this organism have unicellular characteristic with nucleus item which
dispersed inside protoplasm cell.

The organisms which include the member of Monera are

bacteria and Cyanophyta ( Green-Blue algae ). The main different of
them is the ability on photosynthesis. Cyanophyta can for
photosynthesis, while of bacteria, in general can’t for
photosynthesis, except several kinds of bacteria, not all.

Green-Blue algae is kind of Monera’s Kingdom groups because

Green-Blue algae have prokaryotic characteristic, one cell, the
others have many cells with thread’s form and colony’s form, have
chlorophyll. Have fikobilin’s pigment ( the bundle of fikoeretrin and
fikosianin ). The reproduction with splitting itself, fermentation, or
heterocyst.

THE FIRST ( 1st ) EXPERIMENT

Purpose

Perceiving bacterium forms which there in the rotten food-

stuff.

Materials and Appliances

• The Materials

1. The rotten food

a. Rotten tomato ( in 3 days rotten )

b. Rotten Potato ( in 3 days rotten )

c. Rotten of Coconut’s water ( in 2 days rotten )

2. Blue’s Metilen

• The Appliances

1. Microscope 5. Desk mop

2. Cover slip 6. Thick clothes mop

3. Glass side 7. Pipette

4. Toothpick

Steps or Procedure

1. Prepare the glass side, cover slip and the microscope.

Page10

2. Cleaning the glass side with sterile’s mop until clean.

3. Take the thick water ( lendir ) of rotten’s food with toothpick, oily or
spread on the glass side as flimsy as possible.

4. Give blue’s metilen on the top of object ( the thick water on glass
side ) and covering them with cover slip.

5. Take the glass side ( with object ) on the stage for observation.

6. Perceiving by microscope with low magnification, then changes it

into strong magnification for clear result.

7. Arrange the coarse adjustment knob and fine adjustment knob for
clear result.

8. Perceiving the form and draw it.

9. Answer the questions relation with that form.

THE SECOND ( 2nd ) EXPERIMENT

Purpose

Perceiving bacterium forms which there in the crust’s water

( basic’s water from aquarium ).

Materials and Appliances

• The Material

1. The crust’s water ( basic’s water from aquarium )

• The Appliances

1. Microscope 5. Desk mop

2. Cover slip 6. Thick clothes mop

3. Glass side 7. Pipette

4. Toothpick 8. Water

Steps or Procedure

1. Prepare the glass side, cover slip and the microscope.

2. Cleaning the glass side with sterile’s mop until clean.

3. Take the crust’s water from basic’s aquarium with toothpick, oily or
spread on the glass side as flimsy as possible.

4. Give water on the top of object ( the crust’s water on glass side ) and
covering them with cover slip.

THE RESULT FROM THE FIRST ( 1st ) EXPERIMENT

In the first’s experiment, we get some picture from any object which
we perceiving with microscope.

• FROM ROTTEN TOMATO

Magnification :

* Ocular’s lens = 12,5

* Objective lens= 10

12,5 x 10
= 125 kali

• FROM ROTTEN POTATO

Magnification :

* Ocular’s lens = 12,5

* Objective lens= 10

12,5 x 10
= 125 kali

• FROM ROTTEN OF COCONUT’S WATER

Magnification :

* Objective lens= 10

= 125 kali

THE RESULT FROM THE SECOND ( 2nd )

EXPERIMENT

In the second’s experiment, we get some picture and event from any
object which we perceiving with microscope.

The Result of Observation

Magnification :

* Ocular’s lens = 12,5 12,5 x 10

* Objective lens = 10 = 125 kali

Question

1. How the form of the algae which you found ?

The form of algae is Oscillatoria’s form

2. How the characteristic ?

a. Active for moving

b. Have purple’s color in the surface’s skin

c. Have blue’s color inside of the bacteria

d. Oscillatoria’s form

e. Long and dull

CLOSING

Bacterium, of Latin word of bacterium ( plural, bacteria), is giant

group of life organism. They very small ( microscopic) and most unicellular
have single cell, with cellular structure which relative modestly without
nucleus / core of cell, cytoskeleton, and other organelle like and
mitochondria of chloroplast. Their cellular structure is explained
furthermore in article concerning prokaryote, because bacterium represent
prokaryotes, to differentiate them with organism owning cell more
complex, referred
as by eukaryote.
Term " bacterium"
has been applied by
for all prokaryote or
for big group of
them, depends on
idea concerning
their [relation/link].

Bacteria have many

forms. We get some
form from the
experiment using
microscope.

The Crust’s water form basic aquarium has Oscillatoria’s form.
