Otra forma mediante la cual CMV combate el estrs celular y evita la apoptosis es
mediante el ARN de 2.7 kb (2.7), el cual es transcrito pero no codifica para
productos traducidos funcionales20. El efecto anti-apopttico de ARN 2.7 est
dado por la interaccin de ste con el complejo 1 mitocondrial (NAD oxidoreductasa), el que es capaz de inducir apoptosis bajo ciertas condiciones como
enfermedades mitocondriales y factores ambientales 21. El ARN 2.7 interacciona
especficamente con la subunidad GRIM-19 (genes asociados con la mortalidad
inducida por retinoides/interferon) esencial para el ensamblaje y funcin del
complejo 1. El complejo 1 activo soporta la formacin del gradiente electroqumico
necesario para la produccin de ATP, por lo que su interaccin con el ARN 2.7
sugiere que el virus estabiliza el gradiente manteniendo la produccin de
energa22. Por otra parte, es bien sabido que vMIA igualmente promueve la
estabilidad de la membrana mitocondrial, siendo activo predominantemente en
etapas tardas de la infeccin 23; sin embargo, el ARN 2.7 es expresado de forma
abundante en etapas mucho ms tempranas de la infeccin (12 a 24 h) 22.
Inhibicin de la va extrnseca de la apoptosis: participacin de UL36
Como se mencion anteriormente, la apoptosis puede ser tambin inducida por la
unin de los receptores de la muerte con su ligando. Dichos receptores son
protenas transmembranales que pertenecen a la familia del receptor del factor de
necrosis tumoral (TNFR). Se incluyen los receptores de Fas (CD95), TNFR y los
receptores de TRAIL (ligando inductor de apoptosis relacionado a TNF), que
contienen un dominio de muerte (DD) en la cola citoplsmica. La sealizacin de
dichos receptores puede ser simplificada como la unin del receptor con su
respectivo ligando que estimula el reclutamiento de una molcula adaptadora
FADD (dominio de la muerte asociado a Fas) o TRADD (dominio de la muerte
asociado a TNFR)11,24. Dichos adaptadores poseen un dominio DED y uno DD, lo
cual le permite unirse al receptor de la muerte por un lado, y por otro a una
caspasa iniciadora que contiene un homlogo DED. El complejo formado recibe el
nombre de complejo de sealizacin inductor de la muerte (DISC) y permite la
agregacin y autoactivacin de procaspasas-8, que a su vez activarn a una
caspasa efectora como la caspasa-313. En este proceso pueden intervenir
protenas anti-apoptticas que son denominadas FLIP (protenas inhibidoras de
FLICE:enzima convertidora de interleucina-1 tipo FADD), y cuya funcin es la
inhibicin de la activacin de la procaspasa-8 al competir por FADD, ya que estas
protenas tambin contienen un dominio DED 19.
La protena codificada por el gen de CMV humano UL36 denominada inhibidor de
la apoptosis inducida por caspasa-8 (vICA) previene la muerte celular mediada por
receptores a travs de un mecanismo semejante a los reguladores FLIP, al unirse
a la procaspasa-8 evitando su interaccin y activacin con la protena adaptadora
FADD. Es de notar que vICA no posee homologa en su secuencia aminoacdica
con los FLIPs y de forma ms relevante, que no contiene el dominio DED 25. Todos
los CMV de los cuales se cuenta con su secuencia contienen homlogos de vICA
lo que hace suponer que este es un mecanismo anti-apopttico importante en la
patognesis viral15. De manera interesante, en el modelo murino se ha encontrado
que el homlogo de vICA de CMV murino codificado por el gen M36 es importante
para la capacidad infectiva del virus. Empleando lneas celulares y cultivos
