Method / Procedure

4.1 Preparation of Meatloaf Emulsion


A mixture of 500 grams of minced beef with minced onions, one whole egg, 70
grams of breadcrumbs, one celery stick, along with approximately 2.5 grams of
salt and black pepper is made. All ingredients measured with analytical balance.
The emulsified ingredients are then placed into a big bowl and covered with an
aluminium foil. It is being refrigerated overnight in order to freeze the meatloaf
emulsion. After 24 hours, the temperature of frozen meatloaf emulsion is reduced
by thawing it in a chiller. The thawed meatloaf emulsion is now ready to be baked
traditionally using baking oven then same formula used to be cooked using ohmic
cooking unit and to be analyzed later on.
4.2 Designing The Meatloaf Ohmic Cooker Unit and Cooking Procedure
The ohmic cooker unit for the meatloaf consists of a transformer, a thermocouple,
multimeter and also an alternating current as its power supply.

It is made up of a plastic encased in stainless steel whereby the plate that


surrounds the mold from each side is also originated from plastic. The meatloaf
cooker unit is designed with a hole at the back in order to let the melted fats from
the meatloaf to be flown away into a small drainage system at the bottom plate of
the cooker unit. When the meatloaf emulsion sample is being put into the mold, its
initial thickness and length is recorded using a vernier calliper. Then, the mold is
being sandwiched together by the electrode plates. After the life wire and neutral
wire is then connected to the transformer, power supply is turned on and initial
voltage of 40V is set manually by using the multimeter. The voltage capacity is up
to 240V only. At the same time, the initial temperature used is recorded using the
thermocouple by inserting it into a small hole at the front of the mold ensuring it
to have a touch with the meatloaf emulsion. Multimeter A must be set to read the
current until desired voltage is obtained thus start button can be switched on. The
time gap for temperature recording is 10 minutes until a constant or a stable
reading of resistivity is obtained therefore the cooking process can be stopped.
The meatloaf emulsion is taken out from the mold and the length together with the
thickness is measured again using vernier calliper. Next, the cooked meatloaf is
cut to have a look of its internal texture. The same steps are repeated again using
voltages of 50V and 60V. All three cooked meatloaves are being analyzed for its
color using lovibond tintometer, determination of fat, protein and moisture content
as well as tested for its microbial content. The electrical conductivity observed is
recorded.
4.3 Proximate Analysis
4.3.1 Determination of Fat Content
All three samples are prepared in duplicate form. Weight of empty solvent beaker
used was weighed first followed by 2grams (0.01) of pre dried sample prepared
into a pre dried extraction thimble together with filter paper and covered up with a
cotton wool. The thimbles prepared is then being transferred in a soxhlet
extraction machine (Soxhlet Extractor Darmstadt, Germany). Solvent beaker is
added with 125 ml of petroleum ether. After 2 hours and 30 minutes the extraction
should be completed. However, 10 minutes before the extraction is completed,

oven is pre-heated at 100C. Solvent beaker with extracted fat in it is taken out
using towel and moved into the oven to be dried for 30 minutes. Then, it is being
cooled using a dessicator and weighed. The formula used is

Determination of Protein Content


All of the samples include the blank sample is prepared in duplicate form and they
should be homogeneous. Three steps are involved in the determination of protein
content which are digestion, distillation and titration. Digestion started with 2
grams of meatloaf sample is put into the digestion tube and filter paper is used to
weight the sample. Empty digestion tube for blank is prepared. Each tube is added
with 2 kjeltabs of Cu 3.5 first and then 12ml of H 2SO4 added afterwards. Tube is
shaken to ensure that the sample is wetted together with the acid. Digestion is
done using the Kjedahl digester. The exhaust system is attached to digestion tube
in rack and the water aspirator is set to full effect. Rack is loaded with exhaust into
a preheated digestion block with temperature of 420C. After that, START button
is pressed and digestion is done continously until all samples are clear with blue or
green solution (approximately 1 hour 15 minutes of time taken). Then, the rack of
tubes is removed with exhaust remained in the tray. Cooling down process is done
for 10 to 20 minutes. Next step is distillation which started with addition of 80ml
of deionized water into each cooled tube carefully. 25ml of 4% boric acid as
receiver solution is added which also containing the indicator methyl blue and
methyl red to a conical flask. They are placed into the distillation unit and
platform is raised to ensure that the distillation outlet is submerged in the receiver
solution. Digestion tubes are connected to the distillation unit and safety door is
closed. Then, analyze button is pressed. 50 ml of 40% NaOH is dispensed into the
tube. Receiver solution in the distillation flask has light green colour indicating
the presence of an alkali-ammonia. Final step is the titration. The distillate with
standardized HCl (approximately 0.1000N or 0.2000N) is titrated until there is

changes of colour in the distillate; from light green to light grey colour. Thus,
volume of acid consumed in the titration method is noted as Vsample.

4.3.3

Determination of Moisture Content


Using the oven-drying method, approximately 5grams of the meatloaf sample is
put in to a moisture pan and being placed into the oven. It is left to dry for 21
hours and a half. Next, the moisture pan is taken out from the oven and being
cooled down for 20 minutes before the weight is recorded. The process of heating
and weighing the sample is repeated until successive weight has no difference of
more than one milligram. The formula used to calculate the weight loss is:

4.4 Determination of CFU/g


4.4.1 Preparation of Food Homogenate
Each 5 grams from uncooked meatloaf sample, cooked meatloaf at 40V, 50V and
60V are weighed. Then, the samples are mixed into a stomacher bag with the
addition of 45ml of peptone water. The food sample is blended in a stomacher at a
speed of 15000 rpm for 2 minutes.
4.4.2

Serial Dilution
1ml of blended stock is pipette into 9ml of diluent that contain peptone water. The
mixture is then being shaken generously to ensure that it is homogeneously mixed.
After that, a subsequent transfer of same volume is done into a second tube
containing a sterile, 9ml diluent from the previous first tube. First tube labelled
with concentration of 10-1 of cells and 10-2 for the second tube. The process of
serial dilution is continued until the concentration of 10-4 is obtained.

4.4.3

Plating Process
0.1 ml of bacteria from each dilution is pipette onto an agar plate at the center.
Then, L-shaped glass rod or hockey stick rod is steriled by dipping it into a beaker
of ethanol and passed through a flame to burn off the alcohol. The agar surface in
the plate is spreaded evenly using the sterile glass rod. Then, the same sterilization
procedure is done again with the L-shaped glass rod. Procedure is repeated in
order to inoculate the remaining plates. Next, all of the plates are sealed and
labelled according to its number of dilution.

4.4.4

Incubation
Sealed plates are incubated in incubator at temperature of 30C to 35C for 48
hours. After two days, the colonies appeared on the plates are counted.

4.4.5

Colony Counting
The number of bacteria between 30 to 300 is counted and data obtained is
tabulated in table as below:

4.5 Sensory Analysis


The cooked meatloaf is used to be evaluated in a sensory laboratory. 10 panellists
chosen will be served with warm cooked meatloaf. Before the sensory evaluation
started, a short briefing session regarding the cooking method and correct step to
evaluate the sample is done. Sensory characteristic involved in the evaluation
includes the meatloaf colour, tenderness, juiciness, taste and overall acceptance.
Results obtained is presented in ANOVA.

7a. Specific risk assessment for procedure and precautions to be


taken
Before any experiment is conducted, one should be prepared in terms of every hazard available and
must be familiar enough with the procedures of handling each equipments to be used in order to
avoid any unwanted incidents happened. If all precautionary steps as stated in Table 5a and Table 5b
are followed in details, there would be low risk of contact with hazardous chemicals and any hazard
can be overcomed.
Every work done in laboratory must be conducted with complete PPE being worn, any work with
hazardous chemicals must be done carefully and in well-ventilated area to avoid chemical spillage
that may lead to bad incidents.
Every electrical equipment must only be switched on after every components have been connected
and the surrounding is not in wet condition. All of the cables should be checked thoroughly to
ensure correct connection of wires and to detect any damages.
Disposal of hazardous chemical must be done according to Table 6 and any glassware used must be
handled with care.
