A. Study Background
One of the biggest challenges in Indonesia as a maritime country is the high activity of
industries included oil production and transportation in marine water that triggers the production of
petrolium waste pollution. Based on research ,conducted by Eriyanto in 2012, stated that Indonesia's
oil production reached nearly 860,000 barrels of oil per day and estimated about 5 thousand to 10
thousand barrels are polluting the coastal area or roughly equivalent up to 1,590 cubic meters area. A
number of petroleum waste pollution contribute to the increasing serious disease and the decreasing
biodiversity in marine.
So far, the government's efforts to tackle pollution in marine and coastal area focuse on the
technical aspects only, such as application of government regulations that require industry to
neutralize the waste before discharging into the water and each oil companies are required to have
equipment that can stem the oil spill to pump it back in. Government attention to the technical nature
of marine pollution need to be changed in a natural way through biological processes.
There are already many studies related to degrading petroleum pollutants through
biodegradation process, which the researchers noticed that there is a process of decomposition by
microbial activity to transform the structure of a chemical compound that changed the molecular
integrity and reduced the toxicity. However, a number of pollutions in the environment is greater than
the speed of the process of pollutant degradation in a natural way that pollutants will accumulate.
Then, the human intervention required with existing the technology to solve this case.
To be Faced this problem, I was motivated to learn more about the Environmental Molecular
Biology especially its application in using Next Generation Sequencing technology. Environmental
Molecular Biology in Hokkaido University is the program study under Graduate School of
Environmental Science which bring the student to understand excellent abilities of environmental
microbes on the molecular levels. Specific research subjects are oil-field bacteria, marine bacteria,
rhizobacteria, and extremophile bacteria and hyperthermophilic archaea. With its final goals are first
to understand the kernel of life and its limitation and second to develop biotechnology useful for
restoring healthy environment. One application of this technology is the accuracy observation of
microbe diversity that potentially degraded the petroleum pollutant. Microbial diversity information in
contaminated environmental will inform RNA database based on the molecular aspect in objective to
know the comparision about the number of microbes need with the number of petrolium pollutants
that used for reducing and eliminating the toxicity of pollutant. in addition, this technology can
identify all microbes representatively residing in the location. Besides this is being able to be applied
in the biodegradation of pollutant, this technology can be applied in other scientific fields such as in
agriculture and medicine.
With this background Above, I believe that studying the environmental molecular biology may
be one of the development of molecular biology research in Indonesia. In addition, with the provision
lesson of the basics molecular biology and microbiology, then I can learn more about the
environmental focusing in molecular biology, especially in the implementation of Next Generation
Sequencing technologies.

The Graduate School of Environmental Science is
cooperatively run by about 140 faculty members belonging
to the Faculty of Environmental Earth Science, the Faculty
of Fishery Science, the Institute of Low Temperature
Science, the Research Institute for Electronic Science, the
Institute for Catalysis, the Field Science Center for
Northern Biosphere, which are developing environmentrelated science and/or technology. The mission of the
Graduate School of Environmental Science and the Faculty
of Environmental Earth Science is to assist students in
studying earth science, biology, and material science, and
using what they learn in these fields to research global
environmental issues and determine how to resolve them.
The field of education and research covers various
phenomena, issues, and perspectives which relate to the
environment on an extensive spatiotemporal scale. In
temporal terms, these issues range from those spanning
over a scale of 100,000 years such as paleoenvironmental
changes, to those being faced at present in the near future.
In spatial terms, these issues occur on scales that range
from municipal to global scale and include topics such as
natural variability of the environment, changes caused by
human influence, and the technologies for environmental
remediation and protection. Within this scope, student
research has progressed in various forms including marine
observations, survey and observation in the field both
domestically and internationally, laboratory experiments,
simulations using supercomputers, and data analysis.
Future occupational opportunities are diverse and do not
always have an obvious connection with environmental
sciences.

E. Post Study Plan


After completing the study, I hope the knowledge That I have been learned by the course in the
Master of Science focusing in Environmental Molecular Biology can be applied as a form of
dedicating contribution to the development of bioremediation method using molecular biology
approach. The future goals that want to be achieved upon completion of the study are as follows,
1. Conducting an independent research related to marine pollution in Tanjung Perak Port
through technology Next Generation Sequencing (NGS)
2. Cooperating a research related to marine pollution at the Port and Oil Drilling in Indonesia
through technology Next Generation Sequencing (NGS)
3. Compiling the recommendations based on the results of the research as policy proposals
offering applicative program to reduce pollution of the marine environment.
4. Spreading the knowledge to the others

To achieve these future goals, then the following steps are compiled,
1. Applying to be a lecturer and researcher, as well as joining the association of Environmental
Microbiology at the University who already have the study program in East Java, such as
Brawijaya University, Institute of Technology Sepuluh November and Airlangga University
and promote to be pioneer of Next Generation Sequencing technologies, or promote to be a
pioneer in Trunojoyo University, Madura.
2. Cooperating with Research Institution under the government hold such as the National
Institute of Sciences Indonesia to conduct investigations on marine pollution through
technology Next Generation Sequencing, and promote to be pioneer of this technology.
3. To be a practitioner and engage in social activities related to how to solve the pollution in the
marine waters
4. Continuing the study to a higher level such as the doctoral program, in order to improve
scientific competency in the field of environmental molecular biology.
