Animales de estudio
Juveniles machos y hembras de trucha arcoíris (O. mykiss) con un rango de peso de 150 a 250 g,
serán criados desde la etapa de huevo en el laboratorio de producción acuícola de la FES-I UNAM.
La dieta de los peces se realizará con una dieta comercial y una experimental.
Los organismos serán anestesiados y se obtendrá sangre periférica de la vena caudal de los
organismos mediante punción con vacutainer®. Se utilizarán tubos con heparina sódica para la
posterior obtención de linfocitos e igualmente se usarán tubos para la obtención de suero sanguíneo
(centrifugado los tubos a 2,500 rpm durante 10 minutos). El contenido de proteína del suero
sanguíneo se determinará mediante la técnica de Lowry con un kit comercial (Sigma Diagnostics,
MO, EUA).
Se extraerá el RNA total de macrófagos y linfocitos con Trizol de acuerdo a Camilot (2008). Finalizada
la extracción el RNA extraído se cuantificará en un espectrofotómetro y la integridad del mismo se
visualizará en una electroforesis a 310nm en un equipo fotodocumentador.
El RNA obtenido se usará como templado para sintetizar la secuencia de cDNA correspondiente a la
secuencia del gen de la caspasa-4 y -5 (Se diseñarán oligonucleótidos específicos para la caspasa-
4 y -5). Finalmente, para visualizar los productos de cada reacción se realizarán electroforesis en
geles de agarosa al 1.2% teñidos con EtBr en TBE al 0.5x. Los geles se observarán en un equipo
fotodocumentador a 310nm.
La PCR en tiempo real se realizará por triplicado, utilizando como plantilla el cDNA sintetizado a
partir del RNA obtenido, se usarán distintos juegos de oligonucleótidos específicos para la caspasa-
4 y -5. Las reacciones se realizarán en un sistema de PCR en tiempo real StepOnePlus ™ (Applied
Biosystems Inc., Foster City, CA, EE. UU.). Las condiciones de ciclado serán 95°C (20 s) seguidas
de 50 ciclos de 95°C (3 s) y 60°C de (30 s).
