Gas chromatography (GC) protocol Shimadzu GC-2010 Page 1 of 6

GC setup protocol:

1. Start PC (PC08-107) and select program ‘GCsolution’

2. Select tab‘Operation’

and click on ‘Analysis 1’ (e.g. not ‘Offline Editor’ or ‘Postrun’).

Login name is required (User ID=Admin, Password=blank), click ‘OK’.

The program ‘Labsolutions GC Real Time Analysis’ is started and a connection to the GC is
established.
3. Notice the orange ‘Not Ready’ sign in the left corner. To visualize the current machine
settings click ‘view’, ‘Instrument Monitor’.
4. To run a program click ‘file’, ‘open method file’ and select a file (*.gcm). For example
select: D:\General protocols\universal_method_aromatics_8 min.gcm.

5. The CG-program is selected by clicking on ‘Download’ or by selecting ‘Acquisition’,

‘download Instrument Parameters’.

6. Turn on the flame by clicking on ‘ON’-button in the Instrument Monitor panel.

7. Check if all CG-parameters are set correctly (all light on GC are green and no yellow
labels in the Instrument Monitor panel.

8. IN CASE of ERROR, select on the GC-machine: ‘DIAG’, ‘STANDARD

DIAGNOSIS’, ‘ENTER’. A diagnostic analysis will follow. Usually on of the gas
flasks are empty (CAR1=helium, Ask Martin). A help guide is selected by clicking
‘Configuration and Maintenance’, ‘Maintenance Guide’. Select ‘GC-2010’ for help on
e.g. septum replacements.

9. Multiple sample runs are setup by clicking on ‘Batch Processing’, ‘Batch Table
Wizard’. Make sure that ‘data file’ is saved to the correct directory by clicking on file
name and selecting right folder.
10. Batch runs are started by pressing ‘START’ and saving the batch-file.
11. Anaerobic samples (100 ul, SGE-syringe, Microlance 25G needle) are taken and
injected (Luer Lock Needle) in the sample injection port, and pressing ‘START’ on the
GC machine.

12. Retention times, Curve Area, and Curve Height of each run are stored during each run
op the PC.
13. The data is inspected using the ‘Postrun’ program that can be selected in the
‘Operation’ tab of the GCsolution software (see point 2)

.
14. After you have finished your analyses: select ‘file’, ‘close batch file, and close the
batch file tab.

15. Set the CG in standby-mode by clicking ‘file’, ‘open method file’; d:\general
protocols\standby. Press the ’Download’ button. The GC machine then goes to standby
modus. Do not switch off the GC!
16. Close all software in the PC and switch it off.
