DO’s 1. Visually inspect the card for any evidence of drying, cracking of gel or bubbles entrapped in gel and if found so discard the card. 2. Centrifuge unused cards for 1 cycle (10 mins) in the MatrixTM Card Centrifuge before use. 3. Cards should always be stored upright in the racks in an air- conditioned environment between 4 - 25°C. 4. Diluent-2(LISS) should always be stored at 2 - 8°C. 5. Allow MatrixTM Diluent 2 to reach room temperature before use. 6. Whenever test cells are required, preferably use only MatrixTM Erygen Cell panels, if not available prepare cells by conventional technique and make suspension in MatrixTM Diluent 2 (LISS). 7. Cell suspensions to be made in MatrixTM Diluent 2 (LISS) only. 8. Prepare cell suspensions as per protocols provided. Too high or too low cell suspensions can cause aberrant results. 9. In Coomb’s test always pipette cells before serum/plasma. 10. In Coomb’s test, cells should always be added into the reaction chamber at an angle of 45° to create an air gap between reagent and cells. Follow detailed procedure as per the pack insert for each product. 11. Serum/plasma should always be added at an angle of 90° from the top through the walls of reaction chamber. 12. In Blood Grouping and Phenotyping Cards avoid pipette tip touching the reaction chamber. If pipette tip touches the reaction chamber change the tip. 13. To open the foil seal of cards pull the foil backwards gently to avoid splashing of reagents or use MatrixTM ContaVoid. 14. Samples must be centrifuged at 1500g for 10 minutes to avoid fibrin residue which might interfere with the reaction pattern. 15. In order to visually read the test results reading the card against white back ground is recommended.

DON’Ts
1. Do not place cards horizontally (lying down) on any surface.
Cards are to be kept upright in racks provided. 2. Do not open the foil seal of cards unless they are to be used immediately. 3. Do not use cards in which the gel shows any sign of drying, cracking or bubbles entrapped in the gel. 4. Do not store cards near any direct source of heat – on top of refrigerators, incubators, etc. 5. Do not use haemolysed samples for testing. 6. Do not use Normal Saline to prepare cell suspension. 7. Do not touch the tip of the pipette with the serum/plasma or test cells on the walls of reaction chamber to prevent any carryover or contamination. 8. Do not change the incubation or centrifugation time.
