Exposures to particulate matter and gaseous air pollutants have been associated with respiratory tract inflammation, disruption of the nasal respiratory
and olfactory barriers, systemic inflammation, production of mediators of inflammation capable of reaching the brain and systemic circulation of
particulate matter. Mexico City (MC) residents are exposed to significant amounts of ozone, particulate matter and associated lipopolysaccharides.
MC dogs exhibit brain inflammation and an acceleration of Alzheimer’s-like pathology, suggesting that the brain is adversely affected by air pollutants.
MC children, adolescents and adults have a significant upregulation of cyclooxygenase-2 (COX2) and interleukin-1β (IL-1β) in olfactory bulb and frontal
cortex, as well as neuronal and astrocytic accumulation of the 42 amino acid form of β-amyloid peptide (Aβ42), including diffuse amyloid plaques in
frontal cortex.
The pathogenesis of Alzheimer’s disease (AD) is characterized by brain inflammation and the accumulation of Aβ42, which precede the appearance of
neuritic plaques and neurofibrillary tangles, the pathological hallmarks of AD. Our findings of nasal barrier disruption, systemic inflammation, and the
upregulation of COX2 and IL-1β expression and Aβ42 accumulation in brain suggests that sustained exposures to significant concentrations of air
pollutants such as particulate matter could be a risk factor for AD and other neurodegenerative diseases.

Il turnover degli elementi, cioè il tempo di permanenza di un elemento in un comparto è molto variabile.
Di norma è: estremamente lento nella litosfera (tempi geologici), molto lento anche nell'idrosfera,

IN 1958, Dr. Roy Hertz described the “steroid cycle,” anticipating what we now call endocrine
disrupter research, as follows: “. . . we have to consider that the introduction of . . . [hormones
into cattle feed lots] leads to the exposure . . . of individuals who might otherwise not ever
in their lives come in contact with such materials . . . . This is not a theoretical consideration
because we . . . now have encountered two families, each with two children, who presented with
simultaneously developing gynecomastia attributable to the accidental contamination of
vitamin capsules by estrogens during manufacture. If such estrogens can, by stray handling, get
into such pharmaceutical preparations, can they not very readily get where they are not wanted
on the farm? There is one additional consideration in this regard . . . . The fecal excretion of
these materials . . . will be dropped on the soil and . . . over generations there will be
constant replenishment of the soil surface with steroidal substances of this kind.
This in turn has its effect potentially on surface water-supply contamination and also potentially on
the vegetable content of steroids in crops raised on such soil . . . . I think that we are now
actually setting up a steroid cycle in our environment, and we have to give very serious
consideration to its implications for our subsequent development and growth and possibly
reproductive functions“
Molecular nitrogen (N2) is the most abundant element in the air we
breathe. Yet, large quantities of this nitrogen are not available to
organisms. Why is this the case? One reason molecular nitrogen is in
short supply is because it takes a large amount of energy to break
the triple bond that holds the two nitrogen elements together.
Nitrogen fixation is accomplished by nitrifying bacteria in soil and
water. These organisms are capable of breaking this bond and
converting the nitrogen into ammonia (NH3) and nitrate (NO3).
The resulting nitrogen is termed fixed nitrogen. Such a scenario allows
nitrifying bacteria living in conjunction with certain plants to
convert atmospheric nitrogen into a form that plants can use which
is ultimately passed up the food chain. When organisms break down
proteins during respiration, they release their nitrogen as ammonia
(fish), urea (mammals), or uric acid (birds, reptiles and
insects). Decomposers obtain nourishment from these products by
converting them back into ammonia, which plants can then use again,
completing the cycle. Finally, denitrification, accomplished again by
bacteria, converts nitrate back into nitrogen gas
(POPs)

Twenty years ago, HIV was three logs further down the scale, whereas polio was three logs higher. Fifty
years ago, malaria was finally eradicated from Europe, where it had formerly been widespread, including in
England (Shakespeare’s ‘ague’)… and, despite concern over multi-resistance to antibiotics the situation is
considerably better than in the era before… Thanks to effective immunization policies of the WHO, smallpox
was eradicated in 1977; polio and measles viruses, which have no animal reservoir, may soon be
eliminated in the same way (?)
The 1997 outbreak of avian influenza in Hong Kong occurred in mixed markets, where live chickens, quail
and ducks were stacked together in close quarters with humans… The increasing predilection for meat of exotic
species has exacerbated the risk of exposure to infections not previously encountered: this situation probably
triggered the SARS epidemic. Although we are still not sure of the natural reservoir species of SARS coronavirus,
the live markets and restaurants in Guangzhong sold small carnivores, and several species of civet cat, racoon
dog and ferret badger captured in China, Laos, Vietnam ,Thailand.. In Africa..21 human deaths owing to Ebola
virus infection ensued from the butchering of a single chimpanzee... HIV has crossed from chimpanzees to
humans

Overall, H1 isolates collected over the last 75 years from the U.S. pig population exhibit
considerable variability in pathogenicity. There appears to be an increase in genetic and
antigenic diversity coincident with the emergence of the swine triple reassortant H3N2 in
1998

reassortant viruses,
containing genes of avian,
human, and swine
influenza virus origin.
From when it was first isolated in 1930 until 1998, the classical H1N1 (cH1N1) subtype of SIV was essentially the only SIV
lineage circulating in the US swine population. Genetically and antigenically, the cH1N1 SIV and the human influenza
virus implicated in the 1918 Spanish Flu pandemic are similar.
Beginning in 1998, however, clinical disease caused by H3N2 subtypes was recognized in a few states and
soon spread throughout the entire country. Described as reassortant viruses, the original H3N2 strain had
three genes from a human H3N2 influenza virus that circulated in the human population during 1995, as
well as five genes from the cH1N1 SIV. The initial outbreak of influenza attributed to H3N2 occurred during
August of 1998 in a North Carolina pig farm
Appearing at approximately the same time in Minnesota, Iowa, and Texas were additional H3N2 isolates
that had triple reassortants (Figure 3), with genetic material from an avian influenza virus (two genes),
from human (three genes) and swine (three genes) viruses as well.
The emergence of these H3N2 viruses is widely regarded as a remarkable occurrence, as it was the first
time reported that the genome of an influenza virus consisted of gene segments from three different viral
lineages
L’H1N1 è veramente più pericoloso degli altri virus influenzali?
• L’allarme iniziale, tra gli esperti, era legato al fatto che si tratta di un virus
chimerico che, sul piano genetico, presenta sequenze umane, aviarie e
suine, e che non aveva ancora infettato l’uomo.
• Non si tratta, in realtà, di un virus del tutto nuovo,tanto meno di un virus
“uscito da un laboratorio” come qualcuno ha detto. Già nel 2008 erano
stati segnalati nel maiale (e sono da allora monitorati) ceppi simili.
• Anche il fatto che colpisca in un periodo climatico inusuale e che in alcuni
(rari) casi avrebbe colpito più duramente soggetti giovani e sani deve
essere preso in considerazione. Ma rimane il fatto che su oltre 250mila
casi ufficialmente registrati (che con ogni probabilità non rappresentano
il numero totale) i decessi sono stati in tutto 2800, per la maggior parte
nelle Americhe e tra soggetti a rischio.

Il primo caso umano è stato registrato in Messico, ufficialmente il 18
marzo. E’ vero che il virus circolava già da mesi?
• H1N1 circola tra gli umani da oltre novant’anni, dai tempi della Spagnola,
ed è il più diffuso nel nord del pianeta.
• Fu isolato nel 1933 ed è riapparso sulla scena più volte, tanto che da
decenni il comune vaccino influenzale lo contiene.
• Scomparve dal 1957 al 1976, ma continuò probabilmente a circolare tra i
maiali e tornò a infettare l’uomo nel 1977 in circostanze piuttosto
misteriose.
• Poi subì mutazioni e riarrangiamenti, dando origine a tutti i principali
sottotipi oggi circolanti e a ceppi ibridi sul piano genetico, come quello
messicano, che contiene appunto sequenze di origine suina, aviaria e
umana.
E’ questo mix a renderlo nuovo e a far temere che possa essere un nemico
impegnativo per il sistema immunitario?
• I virus diventano pericolosi nella misura in cui cambiano profilo antigenico e
non vengono riconosciuti dal nostro sistema immunitario. Ma soprattutto
nel caso in cui acquisiscono la capacità di innescare una risposta
infiammatoria sistemica, la cosiddetta tempesta di citochine (un
meccanismo descritto per molti virus totalmente differenti, come il Corona-
Sars, il virus della Dengue e molti altri virus emorragici).
• Questo virus non sembra avere tali capacità, che hanno reso micidiali il virus
della Spagnola e l’H5N1 della cosiddetta aviaria che dal 1997 ha ucciso
alcune centinaia di persone.
• Nè abbiamo al momento strumenti (e neppure modelli teorici) per
prevedere quale tipo di mutazione o di ricombinazione possa favorire la
trasformazione di questo o altri sottotipi in uno pericoloso pandemico: vari
ricercatori hanno cercato e in parte trovato mutazioni della sequenza che
possono determinare un aumento della virulenza, ma è sempre più chiaro
che è piuttosto l’assetto complessivo, “sistemico” del genoma virale quello
che conta.
Si tratta quindi di trasformazioni del tutto imprevedibili?
• E’ un dato di fatto che molti virus pericolosi - per intenderci quelli in grado
di innescare la tempesta di citochine o altre gravi reazioni immunologiche
sistemiche – colpiscono spesso soggetti giovani e forti, a differenza dei
comuni virus influenzali che sono pericolosi essenzialmente per soggetti
anziani e debilitati.
• Ma è veramente troppo presto per dire che questo virus possa dare
problemi in tal senso.
• Anche affermare che il virus non può dare problemi non è però del tutto
corretto: non dobbiamo dimenticare che nel 1918 la prima ondata
epidemica non sembrò allarmante, ma poi arrivò la seconda …
Una volta che hanno infettato una persona, come si comportano?
• I virus influenzali per penetrare nelle cellule degli organismi ospiti utilizzano
una particolare proteina, l’emoagglutinina, che muta leggermente nel
tempo ed è contrassegnata con una H e con un numero, che designa i vari
sottotipi: H1N1, H2N2, H3N2..
• Finora solo i sottotipi dotati di emoagglutinina H1, H2, H3 infettano
facilmente l’uomo, perché si adattano bene al recettore sulle cellule delle
nostre vie aeree, vi si replicano con facilità, impegnano le nostre difese: è
questa “battaglia” (di solito per noi rapidamente vittoriosa) che si traduce
nei sintomi - febbre, spossatezza, dolori muscolari…- tipici della comune
influenza.
• Se l’uomo entra in contatto con un sottotipo “nuovo” dotato di una HA
diversa da quelle note alle nostre cellule (è il caso del temibile H5N1) o non
succede nulla, perché il virus non riesce a entrare nel nostro organismo o
o può scatenarsi una gravissima reazione immuno-infiammatoria nel
polmone e/o in circolo: questa è la famosa e temuta tempesta di citochine
..
Un organismo esposto a ripetute infezioni sviluppa difese più forti ?
• Diciamo che questa sarebbe forse una “regola aurea”, che dovremmo
prendere in considerazione in molte occasioni.
• Ma per farlo bisognerebbe non essere condizionati né da allarmi e timori
eccessivi, né dalle molte analisi affrettate e semplicistiche di quanti
accusano le multinazionali,
le istituzioni sanitarie internazionali e nazionali di avere
inventato tutto o addirittura di aver programmato la pandemia per motivi
di lucro o peggio..
• Personalmente sono abbastanza convinto che ci sia stato un eccesso di
“zelo” e che sarebbe più saggio attendere gli sviluppi di una situazione
che al momento non appare particolarmente allarmante e sulla quale
non abbiamo grandi possibilità di incidere, senza rischiare.
• Un errore abbastanza grave, cui si può ancora porre rimedio, è stato
quello di gridare troppo presto “al lupo, al lupo..”. E questo non solo e non
tanto per il panico che si è creato, almeno in alcuni casi, ma per quello che
potrebbe succedere in futuro.
• Se le nostre conoscenze sui virus influenzali non sono del tutto errate, una
“vera” pandemia influenzale (dovuta alla rapida diffusione planetaria di
un sottotipo virale che associ alla contagiosità la virulenza) ha buone
probabilità di verificarsi in un futuro non troppo lontano. Non sappiamo se
sarà questo o un altro H1N1 a mutare o piuttosto un sottotipo
relativamente “nuovo” per l’uomo (magari un H5 o un H7 o ancora un
H9..): il rischio forse in assoluto più grande sarebbe se, sulla scorta di un
ottimismo del tutto ingiustificato (perché fondato sul superamento di
epidemie planetarie tutt’altro che incontenibili), milioni o miliardi di
esseri umani non credessero più alla minaccia. Allora sì, sarebbero guai.
