[Summer 2019] - Holiday Plan

I will stay in our university during this summer vacation to carry out my
experiment.
1. Landfill leachate treatment (my thesis)
The ECMR effluent will be undergone the BAF treatment with the media of
ceramsite and GAC. If the BAF effluent would not meet the Chines standard
discharge due to the high COD concentration after ECMR process even though the
biodegradability index is suitable for biological treatment, the ECMR effluent will
be treated by UF process in order to reduce the COD concentration from 800
mg/L down to below 500 mg/L
BAF process
- Start-up phase
The seed or bacteria are fed into the BAF reactor with the flow rate of 1 L/h in
order to promote the microbial accommodation, the air flow rate was 4 L/min.
This step may last in 3 – 4 weeks until the system reaches the steady state phase.
- Steady state phase
In the steady state phase, 0.2 L of sludge and 0.5 L of ECMR effluent were mixed
and added into the BAF reactor, and then aerated for 3 days to growth biofilm.
Afterwards, the reactor was started with continuous feeding.
The hydraulic retention time is adjusted at 12 h, air blower is 4 L/min to maintain
the DO in the effluent at 3 – 5 mg/L
The backwash process is weekly carried out with 5 min scouring by air followed by
10 min of simultaneously scouring by air and water and finished by blowing air.
The COD, BOD5 in the effluent are tested to examine the performance of BAF
process.
2. E. Coli removal by ECMR process
2.1. Prepare the 1 µm carbon membrane
The carbon membrane with different pore size are fabricating to obtain the 1 µm
pore size membrane. The main component affecting the pore size of carbon
membrane is carbon methyl cellulose (CMC), different concentration of CMC
were used to fabricate membrane. After that, the pore size of each membrane is
determined.
2.2. Identify the mechanism of E. Coli removal in the electro-oxidation
process as well as comparing the effect of OH radical and chlorinated
compounds
- Preparation of free-chloride solution containing E. Coli
The E. Coli suspension is made by inoculating the E. Coli bacteria into the broth
solution containing 5 g/L of NaCl, 10 g/L of beef extract, 5 g/L of peptone, the pH
solution of 7.4 and incubated for 24 h at 37oC. After that, the solution is
centrifuged at 6000 rpm for 10 min, the supernatant is taken out and the biomass
is used to prepare the E. Coli solution.
- Treatment of E. Coli with and without chloride.
