Experimental determination of pH of solutions

I. Introduction

The pH of different solutions can be determined between the two methods;

Electrometric method and colorimetric method. Determination of the pH of them are

essential for it must be at certain levels in order to work properly especially the

substances inside our body. It tells us the level of pH measuring the nature how acidic,

basic or balanced pH (neutral) which the nature of the solution would determine its use.

Color possess quite different measurement problems from other physical quantities

such as pressure or temperature, because it is not a physical object

but visual phenomenon in which there can be a considerable difference between

what we think and what is actually present (Othmer,K., 1964).

Buffers are also important substances which are capable of stabilizing pH in an

aqueous solution (Aqua health products Inc., 2019). They are used for studying the pH

level of different solutions to maintain stability for each. So the buffer prepared on

previous experiment will be applied in this experiment to determine each pH.

II. Objectives

1. To determine the pH of the following samples using the pH meter dependent

upon the set of trial indicators.

A. Determination of pH level using the Electrometric method and;

B. Determination of pH level through color change using the Colorimetric

method.
III. Methodology

Apparatuses and materials:

 3 droppers  Juice (Zest-O)

 2 spot plates  Tap water

 3 test tubes/vials  Urine

 Sprite

Procedure:

A. PREPARATION OF COLORIMETRIC BUFFER STANDARD SOLUTIONS

Prepare the colorimetric buffer standard solution assigned to you by pipetting

4.0 mL of the buffer into a vial/test tube and adding two drops of the indicator for

this pH. Label the vial with the pH and the initials of the indicator and submit to the

instructor.

B. COLORIMETRIC DETERMINATION OF pH

Determine colorimetrically the pH of 0.01 N HCl, 0.02 N Hac, 2% gelatin

solution, and 0.02 M CaCO3.

Determine the approximate pH range of the samples by using either a universal

indicator (eg. Bogen indicator) or a set of trial indicators. This is done by touching

the indicator’s dropper to a cavity of your spot plate then adding a drop of the

solution. If Bogen indicator is being used, the colors below will indicate the

approximate pH:

Be guided by the following flow diagram in order to determine the pH range of your

sample.

Knowing the approximate pH range, make the final pH determination by pipetting

4.0 mL of the solution into a vial adding 2 drops of the indicator used in the preparation

of the colorimetric buffer standards covering this pH range. Compare with the buffer

standards containing this same indicator. Use a white background. The pH of solution is

the pH of buffer standard that matches closely in color and intensity.

For solution that are slightly colored or turbid like gelatin solution, place a vial or

distilled water behind the gelatin with the indicator and a vial with gelatin behind the

buffer standard during the color comparison.


C. ELECTROMETRIC DETERMINATION OF pH

Determine the pH of the following samples using the pH meter: Sprite; Juice(Zest-

O); Tap water and; Urine.
