ACTIVITY # 1
PARTS AND FUNCTIONS OF THE COMPOUND MICROSCOPE

OBJECTIVES:
1. To identify the parts and functions of the light compound microscope
2. To know how to handle and use the microscope properly.

MATERIALS:
→ A compound microscope, lens or tissue paper

PROCEDURE:
1. Your teacher is going to demonstrate before the class the proper ways of handling the
microscope from the time you get it from the counter until you place it down on the lab
table. She will also point out the various parts of the microscope and how they are
supposed to be manipulated.

2. After having been shown all the parts of the compound microscope, you are now
expected to be able to point them out in your own microscope.

3. Identify first the larger parts of the microscope which are the ARM, BASE, BODY TUBE
and the STAGE.

4. Where is the eyepiece or ocular?_________________________________________

To which of the four parts in #3 is it attached?_______________________________
Clean it with a lens or tissue paper.

5. Look for the objectives.

What do you call the round object that holds the objectives?____________________
To which of the four parts in #3 are the objectives attached?____________________
6. Which is longer, the high power objective (HPO) or the lower power objective (LPO)?
___________________________________________________________________
Look for the magnification numbers of the objectives.
What is the magnification number of the LPO?______________________________
The HPO?___________________________________________________________
Put the LPO directly in line with the ocular.

7. Look for the mirror. Wipe both sides of the mirror with lens or tissue paper.
Compare the two sides of the mirror.
Are both sides of the mirror plain?_________________________________________
If not, what kind of mirror is one side?______________________________________

8. Try to move the mirror in all directions. Towards what direction can it face?
___________________________________________________________________

9. With both eyes open, look into the ocular with your left eye. At the same time, adjust
the mirror so that the field of view is brightest. PRACTICE LOOKING THROUGH THE
MICROSCOPE WITH BOTH EYES OPEN.

10. Where is the diaphragm?

To which of the four parts in #3 is attached?_________________________________
Look into the ocular again.
Alternately close and open diaphragm.
What is the effect on the field of view?
___________________________________________________________________

11. Where is the coarse adjustment knob? ____________________________________

Slowly move it clockwise and counterclockwise alternately. What happens?
___________________________________________________________
Which part of the microscope does it move? ________________________________

12. Where is the fine adjustment knob? ______________________________________

Move it clockwise and counterclockwise. When can you see the movement better,
when you turn the fine adjustment or the coarse adjustment?
___________________________________________________________________
13. Find the inclination joint. Loosen it. Incline the microscope slightly toward you and
tighten the joint. Now sits on your stool and look into your microscope while sitted.
When is it more convenient to look into the microscope, when it is inclined or when it
is upright?
___________________________________________________________________
14. Return the microscope to its upright position. Tighten the inclination joint. Return the
mirror to an upright position, side facing you. See to it that the LPO is I line with the
ocular and is one centimeter above the stage. Point the stage clips toward you.

WITH THESE, YOU ARE NOW READY TO RETURN THE MICROSCOPE TO THE
COUNTER.
