1. Students are reminded that practical work is the foundation on which the study
of sciences stands and every student is required to give the most earnest and
careful attention to it.
2. The laboratory work is arranged to accompany as closely as possible the
principles of theory discussed in the lecture classes.
3. Students must observe strict silence in the laboratory.
4. Eating, chewing, drinking, smoking or application of cosmetics is expressly
forbidden in laboratory areas. Storage of food items and outdoor clothing is also
not permitted.
5. No outdoor clothes are to be brought into laboratory areas.
6. Laboratory coats (properly fastened) must be worn at all times in all laboratory
areas including instrument rooms.
7. Place all extra clothing, unnecessary books, backpacks and purses in an
appropriate place. The laboratory area must be kept free of articles not in use.
8. Always keep the containers of alcohol away from open flames.
9. All cuts and abrasions should be covered with a waterproof dressing before
beginning any laboratory work.
10. Disposable protective gloves should generally be worn as necessary (and
changed regularly!) When working at the bench- particularly when handling
chemicals, or any biological material likely to contain micro-organisms.
11. No one is admitted to the lab room 10 minutes after the lab begins.
12. Each student must have his/her own lab record book/Journal.
13. The pages of the notebook/Journal must be numbered in pen all the way to the
end. This is to ensure that none are removed during the year.
14. Lab reports in the notebook are due at the end of the lab period.
15. Anyone caught plagiarizing work in the laboratory notebook, from a current or
past student's notebook, will be liable to punishment.

16. Students are responsible for the apparatus which are placed for their use on the
work bench.
17. If any article is stolen by the student, he/she will be suspended from the
laboratory.
18. Students breaking articles or apparatus will be required to replace it.
If the articles are broken on account of carelessness, they will have
to pay a fine in addition.
19. Cost of breakages should be paid within seven days.
20. When they finish their work they must leave the apparatus in the same position
in which they were placed originally.
21. On completion of experimental work, the workstation and all equipment must
be thoroughly cleaned.
22. All laboratory personnel should wash their hands at regular intervals and always
immediately when contamination is suspected. Hands should always be
thoroughly washed and rinsed on completion of all experimental work and
always before leaving the laboratory.
23. All solid and filter paper waste should be in the dustbin.
24. Lone working or carrying out experimental work in departmental research
laboratories outside normal working hours is not permitted other than by prior
arrangement.
25. Before leaving the lab be sure all electrical devices are unplugged from the wall.
26. Any faults to items of laboratory equipment should immediately be reported to
On Duty staff so that appropriate action can be taken. Prompt notification can
then be given to other potential users and if necessary the equipment in question
can be removed from service until repairs are carried out.
27. Report all accidents and injuries immediately to the instructor.
28. The certificate for the practical course required by the University regulations
will not be granted unless the student's progress has been satisfactory. This will
be judged not only by the practical work done in the laboratory, but also by the
neat, systematic, regular writing of the accounts in the home record book.

Students of all the F.Y, S.Y, T.Y and final year should follow the following rules
strictly:
1. Students should reach respective practical lab at least 5 minutes before scheduled
practical time.
2. Students should enter the laboratory after putting up their apron.
3. Students should come ready with all requirements for particular practical.
4. Students should listen all the instructions and practical to perform in depth and
should ask for any of the difficulty to concerned teacher.
5. Students should handle all inflammable solvents with care and should never take
them near flame.
6. Students should handle acids and alkalis in fuming cupboard only.
7. Students should keep all the reagent and solvents at proper place after using them.
8. Students should take care that while performing experiments they do not cause
any harm to property of college.
9. Students should not use mobile phone inside laboratory and should work in
discipline.
10. Students should report both positive as well as negative results to concerned
teacher and get it signed on that particular day only.
11. After due permission from concerned teacher student should leave practical lab
silently.
12. Students should remove their apron after leaving respective practical laboratory.

2. Good quality of disposable plastic gloves must be used profusely while handling
both corrosive and poisonous chemicals.
3. Use close fitting safety spectacle or vision covering the entire face for protection
in the event of chemical splashing or spraying of corrosive or toxic hot liquid or
gases.
7. All experiments involving toxic solvents and reagent should be carried out in an
efficient fume-cupboard.

2. Water should never be employed to extinguish fire produced from organic solvents.
6. Fire. Personal safety is most important. If a person’s clothing catches on fire, he/she
needs help. Prevent him/her from running. If he/she is close enough, put him/her under
safety.

Dragandroff Reagent
Boil 14gm of Sodium Iodide with 5.2 gm basic Bismuth Carbonate in 50ml
glacial acetic acid for a few minutes. Allow it to stand overnight and filter off the
precipitate of Sodium Acetate crystals. To 40ml of the red brown filtrate add 160ml
of glacial acetic acid and 1ml water. Store it in bottle. When needed add 20ml of
Acetic acid to 10ml of stock solution and add up to 100ml water.
Fehling Solution A
Dissolve 34.66gm of Copper Sulphate in distilled water and make volume
up to 500ml.
Fehling Solution B
Dissolve 173gm of Potassium Sodium Tartarate and 50gm of Sodium
Hydroxide in distilled water and make volume up to 500ml.
Hagers reagent
A saturated solution of picric acid is used for detection of alkaloids.
Iodine Sol.
Dissolve 2gm Iodine and 3gm Potassium Iodide in 100ml water.
Mayer’s reagent
Dissolve 1.Mercuric chloride in 60ml distilled water. (a)

Procedure: -
1. Place the microscope on a stable place on a bench of suitable height free from
vibrations.
2. Provide illumination from any of the three sources, i.e., built-in lamp, external
lamp or sunlight.
3. Direct the path of light to pass through diaphragm, condenser, and the hole of the
stage with maximum intensity while setting the mirror. Use the concave side of
the mirror if there is no condenser and use the planeside if there is a condenser;
keep the condenser at the top position.
5. Revolve the nosepiece and align the low power objective 10X to examine the
object on the slide [objective must click into place].
6. Look through the eyepiece and not into it. Adjust the illumination to improve the
contrast. In case of low power, the illumination is cut down to minimum by
reducing the aperture size.
7. Put one hand on the focusing knob [coarse or fine as the case may be], and other

on the screw to move the stage. Use the course adjustment knob for bringing the
object to the focus and get a sharp focus with the fine adjustment knob. The lower
power objective is commonly used to screen the field of the view. Bring the
object of interest in the centre.
8. Switch to high power [40X] and increase the illumination as needed. While
changing the objective by rotating the nose piece make sure that the objective
clicks into place. Repeat the process of focusing as described earlier by using the
coarse adjustment knob followed by fine adjustment knob. Normally modern
microscopes being parfocal, once the object is focused under any objective, it
stays in focus for all objectives and no further adjustments are necessary.
9. After screening under low power and examining under high power, oil-immersion
objective is used for obtaining greater details of the object.
10. Place a small drop of oil on the slide, over the path of the light. Increase the
illumination to maximum. Turn the nosepiece and set the oil immersion objective
in its place.
11. Use fine adjustment knob to get the object in focus. If this fails, use coarse
adjustment to focus the slide.
12. With the use of fine adjustment, bring the object sharply into focus. It is often
necessary at times to raise or lower the condenser so that the optimum
illumination is secured. This is termed ‘critical illumination’.
13. Finally when the observation is finished, the oil adhering the slide is removed by
means of xylene and wiping it clean by means of lens paper.
1. Carry the microscope by holding its arm or limb with one hand and the other hand
under the foot. Never swing the microscope while carrying it nor turn it upside
down.
3. Never touch any optical surface with anything other than lens tissue.
4. At the end of the experiment, gently clean all immersion oil off the lenses. Do not

use any organic solvents [eg. ethanol, ether, xylene or toluene] to remove the oil
on a regular basis. This may dissolve the cement holding the lens in the socket.
5. Dust and fungus are the worst enemies of microscope. Fungus growing on lenses
and scratches caused by dust particles on the lens surface can ruin the microscope.
Hence, when not in use store the microscope away from the dust and humid
environment.

1. Pour the water inside the water bath so that the level is one half inches above the
heater.
2. Connect the plug to the electric supply and switch on the mains.
3. Set the temp. knob as per the required temp.
4. After the required period is over cut off the electricity supply.
5. Remove the water from it and wipe dry.
Precaution:
1. Keep the water bath clean and replace the water frequently when it appears dirty.
2. Take care that material is placed in the water bath should never invert under any
condition.
3. Check the water level before starting the instrument and never allow the
instrument to dry up.

by position
10. To weigh the compound further press again power key and continue in similar
manner.
