Research experience:

PLiMAP Technology: Peripherally-associated membrane proteins (MAPs) function in a variety of cellular
pathways ranging from cell signaling, host immunity, host-pathogen interaction and vesicular transport.
Identification of MAPs from a complex mix of proteins and functional characterization necessitate
methods that report on binding to membranes of a defined lipid composition. We have developed a
photoactivatable fluorescent phospholipid as a generic reporter of peripheral protein-membrane
interactions. When incorporated into liposomes and exposed to UV, proteins bound to liposomes
become crosslinked with the fluorescent lipid and can be readily detected and quantitated by in-gel
fluorescence analysis. This modification obviates the requirement for high-speed centrifugation
common to most assays that detect liposome-bound proteins. We refer to this assay as Proximity-based
Labeling of Membrane-Associated Proteins (PLiMAP). We are extending this methodology to enrich
membrane associated proteins and identify novel membrane remodeling proteins.

Thesis title: “Studies on the effect of copper nanoparticles and copper complex on DNA molecules,
bacteria and cancer cells: implications in antibacterial and anticancer therapy.”
Metal nanoparticles by virtue of its higher surface to volume ratio and Surface Plasmon Resonance (SPR)
exhibits contrasting physical properties to bulk metal. We have identified a novel in vitro DNA
degradation potential of copper nanoparticles (CuNPs) mediated by singlet oxygen, and its ability to
induce apoptosis in cancer cell lines. CuNPs also showed a superior bactericidal activity over ionic
copper by inducing in vivo nucleic acid degradation, reactive oxygen species generation, and higher
copper transport into the bacterial cells.

Contributed to the development of Insulin Glargine (BASALOGTM) from laboratory to industrial scale,
which is the first long acting insulin developed and marketed by Biocon Limited.
