Vitrification or ultra-rapid cooling is a revolutionary cryopreservation technique that achieves

Cryotop is the special vitrification container consisting of a fine, thin film strip attached
to a hard plastic handle for the minimum volume cooling to realize highest cooling &
warming rates resulted in 99% post -thaw survival. And the hard plastic cover is
attached to protect the oocytes and embryos on strip from not only physical damage
but also virus contamination during storage in LN2. The Cryotop method is simple,
reliable, universal, safety and easy for anyone.
The Cryotop method is now being used in more than 40 countries (700 IVF centers),
and has applied to more than 100,000 clinical cases for oocytes/embryos were
cryopreserved for these
7 years with unbelievable excellent results. The Cryotop method is the most
trustworthy cryopreservation method for oocytes and embryos proved by the best
survival rates, and created huge number of safety birth results. The Cryotop method
was developed by Kuwayama Ph.D. in Advanced Medical Research Institute of Kato
Ladies Clinic, the largest IVF center in the world.

The Cryotop Method
Oocytes and embryos that are to be vitrified by the Cryotop Method are immersed
sequentially into several, specially-formulated Cryoprotectant Solutions. These are
referred to as No. 0 – Basic Solution; No. 1 – Equilibration Solution; No. 2 –
Vitrification Solution.
With the use of very fine pipettes that have an internal diameter suitable for human
applications, oocytes or embryos are carefully pipetted into each well. It is important
that
the internal diameter of the pipette used to handle oocytes and embryos be kept as
small as possible.
The reason for this is to minimize the volume f the various solutions used to vitrify
oocytes or embryos. The smaller the volume of the Vitrification Solution used in the
final step of
the process the higher the cooling and warming rate.
The use of very high cooling and warming rates achieved with the Cryotop Method is
one of the principal advantages of this method, and is the reason for the high
functional survival of oocytes and embryos.
Oocytes are gently equilibrated with three successive steps little by little over a
period of 15 minutes. This prepares the oocytes to be exposed to the Vitrification
Solution. In the case of embryos, they will reach equilibrium in 12 minutes for
pronuclear stage and cleavage cell,
15 minutes for morula stage stage and blastocyst. They are then ready to be
exposed to the Vitrification Solution and to be vitrified.
The oocytes or embryos are then aspirated in as small a volume as possible of the
Equilibration Solution and placed onto the top surface of the first well of Vitrification
Solution. Within less than 1 minute of exposure of the oocytes or embryos to the
Vitrification Solution, they are aspirated in as small a volume as possible of the
Vitrification Solution and placed onto the top surface of the second well of Vitrification
Solution. Within less than
30 seconds of exposure of the oocytes or embryos to the Vitrification Solution,
they are aspirated into the pipette and deposited onto the end of the Cryotop itself
close to the black mark of the plastic end. Excess Vitrification Solution is then
aspirated away being very careful not to dislodge the oocytes or embryos.
The Cryotop is immediately submerged into LN2 in the styrofoam box and swirled to
vitrify the oocytes or embryos. Finally, the protective cap is carefully inserted onto the
Cryotop and twisted to assure that it has been tightly fitted onto it. The specimen is
then ready to be placed into cryovials and onto labeled aluminum canes for storage
in LN2.
The final step of the Cryotop Method is the liquefaction and recovery of the oocytes
or embryos. This is sometimes referred to erroneously as “thawing.” In Cryobiology,
the word “thawing” refers to the melting of ice. But since the Cryotop Method is a
vitrification procedure, it is preferable to refer to liquefaction (melting) of the vitrified
specimen.
After the appropriate specimen has been identified and removed from the cane and
from
the cryovial in which it was stored, the individual Cryotop is immediately and rapidly
transferred from LN2 into the Recovery Solution. The vitrified sample will melt
instantaneously.
The oocytes or embryos are then sequentially pipette through three steps to rinse
them.
They are then ready to be fertilized (in the case of oocytes) or to be incubated or
transferred (in the case of embryos).

freezing method, it yield dramatically higher survival and developmental rates for both sensitive
oocytes and all stages of embryos: zygote, cleavage stage and blastocysts.
The Cryotop Method
The use of very high cooling and warming rates achieved with the Cryotop method is one of the
principal advantages of this method, and is the reason for the high functional survival of oocytes and
embryos.
The Cryotop method is the most trustworthy cryopreservation method for oocytes and embryo that
is simple, reliable, safe and easy to use. Developed by Kuwayama Ph.D. in Advanced Medical
Research Institute at the Kato Ladies Clinic, the largest IVF center in the world, it is now being used
in more than 40 countries (700 IVF centers) world over with excellent oocyte/embryo survival rates
and a high percentage of safe deliveries.
