Magnifying lens is fixed on its appropriate plate & marker is connected to either right or left side of the instrument. 4.1.2 Switch ON the mains. 4.1.3 Switch on the power button. The internal light will be flashed ON. PROCEDURE : OPERATION Ensure that the instrument is clean & free from dust Adjust the spirit level provided on the top of the balance for precise weighing. Clean the weighing pan before and after weighing. Ensure that the setting knobs are in the zero position. Switch ON the mains. For checking position move the knob, on the left side panel at bottom of the instrument, turn half anti clockwise direction, and when turn on the clockwise direction gives the weighing position. 4.1.7 First turn the operating knob in the checking position. The light source will be on and check that the zero zero of the vernier scale and projection scale are coinciding with one another. 4.1.8 If it does not coincide, adjust with the adjusting knob provided on the right side panel of the instrument. 4.1.9 First check in the approximate weight in its checking position, if it within the desired range then bring same knob first in the rest position and then turn the knob on its weighing position. During weighing close the weighing cabinet for more accuracy. 4.1.10 Note down the accurate weight and put the operating knob in its rest position and set all weight adjustment knobs at zero position before switching off the instrument. 4.1.11 The balance has the capacity to produce weight up to 200gm.

PROCEDURE : OPERATION Clean the working table,top & side glass with IPA 70% before & after use. Clean the tubelight with clean dry sterile duster. Check the level of the instrument base. The base should possess uniform level with no ups & downs. 4.1.4 Check Air Flow pressure manometer indicate within acceptance limit [12 to 17 mm for Microroom, 10 to 15 mm for Sterility Room] 4.1.5 Start the laminar flow to allow the flow of sterile air. 4.1.6 Continue the work after half an hour of starting the laminar flow. 4.1.7 Switch on the tubelights of the laminar flow bench. 4.1.8 Bring only required sterile utensils & glass wares to the LAF bench. 4.1.9 Start gas burners. 4.1.10 Carry out microbiological work near the flame of gas burner. 4.1.11 Do not do any writing work in the Laminar Flow. 4.1.12 Immediately remove plates, glass ware & media from the area. 4.1.13 Clean the working area.Spray the disinfectant solution. 4.1.14 Switch OFF the tube lights of LAF bench.Switch OFF the instrument when not in use.

PROCEDURE : OPERATION Ensure that the autoclave is clean & free form dust & placed in a safe area. Fill the autoclave with D.M. water till the heater is immersed fully in the water. Load the article to be sterilized in the basket. Shut the lid by help of lifter & close the autoclave. Tight the screws diagonally. Switch ON the mains. Open the steam release valve & release the steam for 5 min., to avoid vacuum in the chamber. Close the valve. 4.1.8 Continue heating till the pressure gauge shows 15lbs/In2 pressure & temperature 121C srC constant. OBJECTIVE : To lay down a procedure for the operation of B.O.D Incubator SCOPE : This procedure is applicable for operation of B.O.D Incubator ID No : PPL/INST/BOD/043 Make : Spectrum Location : Microbiology Department RESPONSIBILITY : HOD shall be overall responsible for the operation of B.O.D Incubator PROCEDURE : OPERATION Switch on the mains switch and B.O.D. Red main switch. Press black button and adjust required temperature with help of Reset and zero knob. Release the black button, actual temperature will display. Check the temperature on digital display, should match with temperature on thermometer, which is kept inside the incubator. For cooling time compressor will on after 90 sec. period Heating and cooling will be indicated by Red & Green colour lamp respectively, provided on front panel. Check the actual temperature after sometime. Temperature record should be maintained daily. Temperature deviation accepted upto +/- 1C.

4.0 PROCEDURE : 4.1 OPERATION 4.1.1 Switch ON the mains. 4.1.2 The green indication light will be ON. 4.1.3 There is three-way switch for the selection of temperature range. 4.1.4 The requied temperature (e.g.105C) can be set by a thermostatically controlled knob, by rotating it in clockwise or anti-clockwise direction. 4.1.5 Note : When temperature is increased, a red indication light will be ON & then will be OFF automatically. 4.1.6 Inside the oven, there are two trays. Put any object that requires drying , on this tray & close the door of the oven. 4.1.7 A small hole is provided on the top of the instrument, to place thermometer or any other temperature measuring device. 4.1.8 A vapour or fumes releasing valve is also provided on the top or side of the instrument. 4.1.9 Switch OFF the instrument & mains, when not in use for long period. 5.0 ABBREVIATION  SOP : Standard Operating Procedure 6.0 REFERENCE Operating Manual
