A Practical training course on Molecular Diagnostics and bioinformatic tools Objective: The science of diagnostics gets revolutionized with the development of Molecular biology tools as it provides high level of sensitivity and specificity to the diagnostics. These tools can detect various pathogenic organisms like bacteria, viruses, etc. through the presence of DNA, RNA or protein sequences in clinical samples. Nowadays commercial kits are available to detect pathogens directly from clinical samples and make it possible to obtain results in just a few minutes or hour, but most of these kits are exogenous in origin and thus very much expensive. Therefore, it is important that highly specific diagnostic tests should be developed indigenously based on the Indian strains of pathogens. Hence, the present practical training course is designed to provide hands on training on basic molecular biology techniques used for diagnosis of pathogens in blood, semen, clinical tissues, faecal samples and characterization of pathogens and breeds of animals. For better understanding of data of molecular biology, it is essential to have the basic knowledge of bioinformatic tools; keeping these points in view this training programme will also include hands on training on bioinformatics tools. Course contents: Preparation of reagents, buffer and media. Extraction and purification of nucleic acid of viral agents from various sources like blood, semen, clinical tissues, faecal samples, etc. Electrophoretic separation of nucleic acid and protein. RNA-Polyacrylamide gel electrophoresis for segmented viruses. Template preparation for Polymerase chain reaction (PCR) Amplification of gene of interest using conventional PCR Synthesis of cDNA and RT-PCR based amplification of viral genes. DNA markers like RAPD,RFLP etc. Molecular cloning of genes in suitable vectors Sequence alignment tools e.g. BLAST Designing of PCR Primer Analysis of nucleotide sequences.
