Integrin-organization of growth factor signaling. Engagement of integrins with the ECM allows permissive growth factor (GF)
signaling to the MAPK cascade. Regulation of this cascade can occur at the level of the receptor, Raf or MEK. Integrins tether
the actin cytoskeleton, leading to the formation of cortical actin and stress fiber structures. Direct integrin engagement favors
GTP-loading of Cdc42. Cdc42 may, through the formation of cortical actin associated structures or activation of PAK, impinge
either at the level of Raf or MEK. Integrin engagement also leads to tyrosine phosphorylation of FAK and the formation of
possible adaptor/scaffolding complexes containing p130(Cas), paxillin (Pax) and Src. p130(Cas) is capable of adhesion-
dependent binding to 14-3-3, creating a link between the tyrosine phosphorylation of FAK and the activation of Raf. Alternatively,
signals downstream of integrins may impinge upon a MAPK scaffold protein, such as MP-1.

The nonfluorescent precursor (NPE-HCCC2, top) becomes fluorescent when activated by UV light, and fluorescent HCCC2
diffuses from 'cell 1' to 'cell 2'; the change in fluorescence in each cell can be used to quantify the gap junction permeability. A
gap junction composed of connexin channel subunits is shown with its hypothesized linkage to a SOC via the scaffolding
protein zonula occludens-1 (ZO-1); other proteins are dynamically linked to gap junctions (including c-src, serine-threonine
kinases, tubulin, catenins, caveolin-1 and scaffolding proteins), forming a molecular complex termed a 'nexus'. Studies with the
LAMP method suggest that Ca2+ entry through the SOC increases Ca2+ concentration to very high levels in close proximity to
the gap junction, thereby directly or indirectly closing the intercellular channels.
