Minyak Atsiri = Essential Oil
LOCALIZATION
Synthesis & accumulation of essential oils are generally
associated with the presence of specialized
histological structures, often located on or near the
surface of the plant:
- Oil cells of Zingiberaceae
- Glandular trichomes of Lamiaceae
- Secretory cavities of Myrtaceae or Rutaceae
- Secretory canals of Apiaceae or Astereraceae
(Compositeae)
VOLATILE OIL COMPOSITION
• Mixtures of HC’s and oxygenated compounds
derived from these HC’s.
– Oil of turpentine – mainly HC’s
– Oil of Clove – mainly oxygenated compounds
• EXCEPTION: Oils derived from glycosides (e.g. bitter
almond oil & mustard oil).
• Oxygenated compounds – responsible for the
odour/smell of the oil. They are slightly water
soluble – Rose water & Orange Water; more
alcohol soluble.
• Most volatile oils are terpenoid. Some are aromatic
(benzene) derivatives mixed with terpenes.
• Some compounds are aromatic, but terpenoid in
origin (e.g. Thymol – Thyme)
CHEMICAL COMPOSITION
Volatile oils are divided into 2 main classes based on
their biosynthetic origin
Terpenes, or terpenoids,
are the largest group of
secondary products
(metabolites).
They are all formed from
acetyl CoA or glycolytic
intermediates.
CLASSIFICATION OF TERPENES
All terpenes are formed
from 5-C elements

PROPERTIES OF VOLATILE OILS
• Almost entirely volatile without
decomposition.
• Density: Most are less than 1g/ml.
– 2 are heavier – Oil of Cinnamon and Clove
oil.
• Soluble in ether, chloroform & alcohol.
• Slightly soluble in water: give it a
characteristic odour & taste.
• Leaves a temporary translucent stain on
paper which disappears as the oil
volatilizes.
• Most are colourless. Oxidize on exposure
to air and resinify colour becomes
darker (odour changes slightly).
• All are characteristic odours.
• Most are optically active.
PRODUCTION OF ESSENTIAL OILS
Essential oils may be produced
i. By steam
- Simple steam distillation
- Saturated steam distillation
- Hydrodiffusion
i. By expression
ii. Other Methods
Advantages:
• Limits the alteration of the constituents of the oil
• It shortens the duration of the treatment
• It conserves energy
• It can also be conducted on on-line in automated set ups.
c. HYDRODIFFUSION
i. SOLVENT EXTRACTION
Extraction is generally preceded by a process of:
bruising the fresh, wilted or semi-desiccated
organs, chopping herbaceous drugs, pounding
roots & rhizomes or turning wood into chips or
shavings.
The procedure is conducted in specialized facilities e.g.
Soxhlet-type extractor.
SOLVENT EXTRACTION
The solvent selection is influenced by technical & economical factors
ADVANTAGES:
- obtain extracts which are very close in composition
to the natural product.
- It is possible to adjust the selectivity & viscosity, etc
by fine tuning the temperature & pressure
- All result in the increase of popularity of this type of
method
EXTRACTION BY SUPERCRITICAL GASSES
USES
Initially developed to decaffeinate coffees, prepare hops
extracts or to remove nicotine from tobacco, the method
is now used to
- Prepare spice extracts (ginger, paprika, celery)
- Specific flavours (black tea, oak wood smoke)
- Plant oils
- To produce specified types of a certain product, e.g.
thujoneless wormwood oil.

rectify the oils obtained.

STEAM DISTILLATION
• Steam Distillation is the most popular method used to
extract and isolate essential oils from plants for use in
natural products. This happens when the steam vaporizes
the plant material’s volatile compounds, which eventually
go through a condensation and collection process.
• Distilasi Uap adalah metode yang paling populer
digunakan untuk mengekstraksi dan mengisolasi minyak
esensial dari tanaman untuk digunakan dalam produk
alami. Ini terjadi ketika uap menguapkan senyawa volatil
bahan tanaman, yang akhirnya melalui proses kondensasi
dan pengumpulan
STEAM DISTILLATION PROCESS
1. Wadah besar yang disebut Still, yang biasanya terbuat dari stainless
steel, mengandung bahan tanaman yang ditambahkan uap ke dalamnya.
2. Melalui saluran masuk, uap dialirkan melalui bahan tanaman yang
mengandung minyak yang diinginkan, melepaskan senyawa volatil
tanaman dan mengubahnya menjadi uap.
3. Senyawa tanaman yang diuapkan bergerak ke labu kondensasi atau
Kondensor. Di sini, dua pipa terpisah memungkinkan air panas keluar dan
air dingin masuk ke Kondensor. Ini membuat uap dingin kembali ke
bentuk cair.
4. Produk sampingan cairan aromatik turun dari Kondensor dan terkumpul
di dalam wadah di bawahnya, yang disebut Pemisah. Karena air dan
minyak tidak bercampur, minyak esensial mengapung di atas air. Dari
sini, ia disedot. (Beberapa minyak atsiri lebih berat daripada air, seperti
minyak atsiri cengkeh, sehingga mereka ditemukan di bagian bawah
Pemisah.)
STEAM DISTILLATION
Hidrodistilasi
• Untuk mengisolasi minyak esensial dengan hidrodistilasi, bahan tanaman
minyak atsiri dikemas dalam diam dan jumlah air yang cukup ditambahkan
dan dididihkan; alternatifnya, uap hidup dialirkan ke wadah besar (still).
Karena pengaruh air panas dan uap, minyak atsiri dibebaskan dari kelenjar
minyak di jaringan tanaman. Campuran uap air dan minyak dikondensasi
oleh pendinginan tidak langsung dengan air. Dari kondensor, distilat
mengalir ke separator, di mana minyak atsiri terpisah secara otomatis dari
air destilat.
• Mekanisme Penyulingan
Hidrodistilasi bahan tanaman melibatkan proses fisikokimia utama berikut:
– Hidrodifusi
– Hidrolisis
– Dekomposis oleh panas
Proses Hidrodifusi
• Difusi minyak atisiri dan air panas melalui membran tanaman dikenal
sebagai hidrodifusi.
• Dalam distilasi uap, uap sebenarnya tidak menembus membran sel kering.
Oleh karena itu, bahan tanaman kering hanya dapat dikeringkan dengan
uap kering hanya jika semua minyak atsiri telah dibebaskan dari sel-sel
pembawa minyak dengan penyaringan bahan tanaman secara menyeluruh
terlebih dahulu.
• Tetapi, ketika bahan tanaman direndam dengan air, pertukaran uap dalam
jaringan didasarkan pada permeabilitasnya saat dalam kondisi bengkak.
Membran sel tanaman hampir tidak tembus terhadap minyak atsiri. Oleh
karena itu, dalam proses yang sebenarnya, pada suhu air mendidih, bagian
dari minyak atsiri larut dalam air yang ada di dalam kelenjar, dan larutan
minyak-air ini meresap, melalui osmosis, selaput yang bengkak dan
akhirnya mencapai permukaan luar, di mana minyak diuapkan dengan
melewatkan uap
Proses Hidrodifusi
• Aspek lain dari hidrodifusi adalah kecepatan penguapan minyak
tidak dipengaruhi oleh volatilitas komponen minyak, tetapi oleh
tingkat kelarutannya dalam air.
• Oleh karena itu, konstituen yang mendidihkan tinggi tetapi lebih
larut dalam air dari minyak dalam jaringan tanaman menyaring
sebelum konstituen yang mendidih tetapi kurang larut dalam air.
• Karena laju hidrodifusi lambat, distilasi bahan yang tidak
terkontaminasi membutuhkan waktu lebih lama daripada bahan
yang dihaluskan
Hidrolisis
• Hidrolisis dalam konteks ini didefinisikan sebagai reaksi kimia antara air
dan konstituen tertentu dari minyak atsiri.
• Ester adalah konstituen dari minyak atsiri dan, di hadapan air, terutama
pada suhu tinggi, mereka cenderung bereaksi dengan air untuk
membentuk asam dan alkohol. Namun, reaksi tidak lengkap di kedua arah
dan hubungan antara konsentrasi molal berbagai konstituen pada
