1. Utiliser les pattes (disponibles dans le tube et noter le nombre)

Resuspendre le culot sec avec 100 µl d’H2O milli Q, délicatement en grattant bien les bords 15. laisser une nuit à 4°C 16. remélanger à la pipette l’ADN 17. Digestion RNAse A à raison de 1µg/ml et 2h à 37°C

To quantify the amount of DNA, readings should be taken at wavelengths of 260 nm
and 280 nm. The reading at 260nm allows calculation of the concentration of nucleic acids in the sample. An O.D. of 1 corresponds to approximately 50 µg/ml for double-stranded DNA. The ratio between the readings at 260 nm and 280 nm (O.D.260/O.D.280) provides an estimate of purity of the nucleic acid. Pure preparations of DNA have ratios equal or superior to 1.8 (Maniatis et al., 1989) Before readings, the samples were diluted 1/50 (QSP 1 ml) and read in a CE 2021 – CECIL spectrophotometer.
