La Société JOUAN garantit que cet appareil ne présente aucun défaut de matériau ni de montage au moment de
la sortie d’usine et remplacera ou réparera cet appareil s’il s’est avéré défectueux en utilisation normale ou au
cours d’une opération d’entretien pendant UN AN à partir de la date de livraison.
Notre obligation dans le cadre de cette garantie se limite à la réparation de l’appareil défectueux ou d’une pièce
quelconque de cet appareil lorsqu’elle est renvoyée, en port payé, à un atelier service autorisé ou à nos services
de SAINT-HERBLAIN.
Cette garantie sera considérée comme caduque si l’appareil a été faussé, mal entretenu ou négligé, mal utilisé
ou détérioré accidentellement.
Il n’existe aucune autre garantie explicite en dehors des clauses ci-dessus.

1 UTILISATION ET FONCTION
1.1. DESCRIPTION
Le centrifugeur KR4i sert à séparer, grâce à la force centrifuge des particules de densité ou de taille
différentes en suspension dans un fluide, par rotation autour d’un axe.
Une gamme de rotors s’adapte sur l’arbre d’entraînement en fonction des récipients contenant les
échantillons, de la capacité et de la performance recherchée. La vitesse maximum (et par conséquent la
force centrifuge maximum), est fonction du rotor utilisé.
Un système de commande permet de contrôler les vitesses, les pentes d’accélération et de freinage, la
durée des cycles et la température.
La réfrigération est assurée par un groupe frigorifique chargé en fluide sans CFC. Le fluide réfrigérant
circule autour de la chambre pour obtenir une efficacité maximale.
Par sa taille et ses performances, le KR4i convient aux applications suivantes :
- Fractionnement du sang
- Séparation industrielle
- Biochimie
- Radio immunologie

Les particules en suspension dans l’échantillon sont séparées par la force centrifuge relative (FCR)
appliquée à cet échantillon. La force exercée est proportionnelle au carré de la vitesse de rotation et à la
distance séparant l’échantillon de l’axe de rotation.
Les formules suivantes permettent un rapide calcul des paramètres de centrifugation et des conséquences
de la modification de l’un d’eux.
Remarque : la valeur du “rayon” programmée peut correspondre à un certain niveau dans le tube (cas
des séparations sur gradient) et non forcément au rayon maxi (fond du tube). Les rayons maxi sont
indiqués dans le tableau Spécifications Générales au chapitre 3.

2 PROCEDURE D’INSTALLATION
Compte-tenu du poids de l’appareil, son levage et son transport doivent être réalisés par un moyen
mécanisé (diable, transpalette) conforme à la réglementation en vigueur et manipulé par du personnel
ayant suivi la formation requise.
L’appareil devra être soutenu par sa partie inférieure. S’il doit être transporté sans sa palette, dans un
escalier par exemple, s’adresser à des professionnels de la manutention.
Cet appareil est conçu pour être sûr dans les conditions de fonctionnement suivantes :
- Utilisation en intérieur
- Température : 5° à 40°C
- Humidité relative maximale de 80% pour les températures allant jusqu’à 22°C.
- Altitude maximale : 2000 m
Les performances maximales sont assurées dans la plage de température ambiante suivante : 15°C à
25°C.
L'appareil doit être installé dans un endroit propre non poussiéreux et non corrosif.
Prévoir un espace de 20 cm à l'arrière de l'appareil et un espace de 10 cm sur les côtés pour assurer une
ventilation suffisante.
Les centrifugeurs sont de préférence, à placer dans des pièces spécifiquement allouées, ne comportant
aucun autre matériel de travail.
Les murs ou cloisons situés à proximité des centrifugeurs (moins de 1 m), doivent être de nature à stopper
le déplacement d’un centrifugeur en cas d’accident majeur (sont à proscrire, notamment, les cloisons
amovibles, les cloisons de bois aggloméré à simple épaisseur sans renforts, les cloisons de placoplâtre
non renforcées).
Le sol doit être plan et d’une résistance suffisante pour supporter l’appareil (346 kg) et pour ne pas
générer ou entretenir de vibrations. Ces vibrations peuvent être la cause de variations dans les résultats
de centrifugation. Voir aussi chapitre 2.5 pour l’emplacement des patins.
Il n’y a pas de prescription minimum obligatoire quantifiée pour définir les caractéristiques du sol, car il y
a beaucoup de paramètres techniques à considérer, tels que, notamment :
- Constitution de la ou des couches constituant le sol,
- Ancrage dans les murs,
- Structures de support du sol,
- Charges déjà appliquées au sol considéré et leur emplacement,
- Charges déjà appliquées aux structures de support.
La disposition des appareils doit permettre des interventions de maintenance régulières ; autrement dit, il
est recommandé de pouvoir déplacer les appareils avec le plus de facilité possible.

ATTENTION : Avant d’installer l’appareil lire attentivement ce qui suit. Pendant la manutention ne jamais
glisser la main sous l’appareil.
- Retirer les agrafes à l'aide d'un tournevis.
- Démonter les 2 tirefonds (A) à l'aide de la clé de 19 située dans la boîte d’accessoires.
- Démonter le tirefond (B).
- En utilisant comme levier le tasseau (C) :
. Retirer le chevron et la rampe de déchargement (D) en faisant levier à l’arrière.
. Retirer le chevron et la rampe de déchargement (E) en faisant levier sur le côté droit à l’avant.
- S'assurer que les roulettes sont bien en face des rampes (F) et que celles-ci sont bien perpendiculaires
à la palette.
2 personnes sont nécessaires pour descendre l'appareil : 1 à l'avant du centrifugeur pour guider et freiner
l'appareil dans sa descente, 1 à l’arrière pour amener le centrifugeur sur les rampes.

Le circuit électrique doit être conforme aux réglementations en vigueur. Le KR4i est délivré sans prise.
Son branchement doit être réalisé par un technicien agréé suivant le code couleur européen :
- Bleu : neutre, marron : phase, jaune/vert : terre
Suivant le modèle, l’alimentation nécessaire est :
- 230 V ±10 % 50 Hz 25 A max.
- 208 / 230 V 60 Hz 25 A max.
Nous rappelons que, pour respecter les normes de sécurité électrique relatives à la protection des
personnes contre les contacts indirects, l’alimentation de l’appareil doit être assurée par une prise de
courant munie d’un dispositif de protection assurant la coupure automatique de l’alimentation en cas de
défaut d’isolement. Une alimentation équipée d’un disjoncteur différentiel de dimension convenable
satisfait à cette exigence.
La prise alimentant l’appareil doit être immédiatement identifiable et à portée de main de
l’opérateur.

Le KR4i est équipé d’un verrouillage à 2 points qui interdit le démarrage tant que le couvercle n’est pas
complètement fermé et empêche l’ouverture de celui-ci jusqu’à ce que le rotor soit arrêté.
Après mise sous tension de l’appareil, attendre l’affichage de l’écran de veille (vitesse / durée /
température), puis tirer vers l’avant la poignée de déverouillage située sur le côté droit de l’appareil : le
couvercle est déverouillée et s’ouvre automatiquement.
En cas de coupure d'électricité ou de panne du centrifugeur, le dispositif de déverrouillage du couvercle
du centrifugeur interdit son ouverture.

2.7. INSPECTION
Avant installation, le rotor doit être soigneusement examiné afin de s’assurer de son parfait état de
propreté et de toute absence de corrosion.
La corrosion chimique ou la corrosion sous contrainte peuvent entraîner l’apparition de microfissures et à
terme, l’éclatement du rotor et occasionner de sérieux dégâts au centrifugeur lui-même.
Les particules collées au fond des alvéoles peuvent casser les tubes lors de la centrifugation, le rotor n’est
plus équilibré et se trouve contaminé par l’échantillon répandu.
L’orifice central du rotor, le clip au centre du rotor et l’axe moteur doivent être maintenus propres et en
parfait état. Ces différentes parties doivent être essuyées avant chaque utilisation.
Se reporter au chapitre 6 : Risques, Précautions, Limitations d’usage
Cette opération doit être effectuée par du personnel qualifié et autorisé. Ne jamais tenir le
rotor par son carenage.

2.8.2. ROTORS CARÉNÉS RP4 - RC6 - RP6, SANS COUVERCLE / ROTOR SANS CARÉNAGE RHK4B
Le colis du rotor contient :
. le rotor,
. un tube de graisse pour les tourillons,
. une notice d'instruction et d'entretien.
- Vérifier la propreté de l'axe moteur et du moyeu du rotor.
- Graisser très légèrement l'axe moteur.
- Repérer la position de la clavette sur l'axe et la rainure sur le moyeu.
- En tenant le rotor par 2 branches opposées, l'amener à la verticale de l'axe moteur et l'introduire
doucement sur l'axe. Le rotor glisse librement sur l'axe.
- Mettre en place la rondelle et la vis de fixation : la serrer dans le sens horaire (couple de serrage 8 m.N)
ATTENTION : Ne jamais serrer le rotor sans la rondelle de fixation.
- Graisser très légèrement les tourillons.

S’assurer que les accessoires sont bien en place et équilibrés.
- Vérifier que le rotor est bien serré sur l’axe.
- Vérifier que les éléments d’accrochage du couvercle de rotor sont bien serrés.
- Vérifier qu’il n’y a pas d’eau ou de liquide dans le fond du carénage.
- Vérifier que les nacelles sont toutes en place et qu’elles pivotent librement.
- Vérifier qu’il n’y a pas de panier transfusion vide dans les nacelles.
Le déséquilibre entre les charges symétriquement opposées est amplifié par la centrifugation. Il est donc
important d'équilibrer les charges et de placer les charges identiques symétriquement par rapport à
l'axe.
ATTENTION : N'utiliser que des récipients (tubes, poches, flacons) connus pour résister à la centrifugation.
Bien que le KR4i admette un déséquilibre de plus de 50 grammes, un déséquilibre de plus de
15 grammes entraine des vibrations qui peuvent provoquer une remise en suspension. L'utilisation
d'anciens accessoires sur le KR4i ne doit se faire qu'après consultation d'un représentant JOUAN.
ATTENTION : un déséquilibre excessif est susceptible d’endommager sérieusement le rotor et le
centrifugeur.

4 PRINCIPE DE FONCTIONNEMENT
4.1. ENTRAINEMENT
Le KR4i est dôté d’un moteur asynchrone triphasé contrôlé en tension et en fréquence. La vitesse de
rotation de l’ensemble rotor peut ainsi être asservie sur la consigne de vitesse demandée par l’utilisateur.
4.2. ACCELERATION
Une selection parmi 10 pentes d'accélération permet d'ajuster sa puissance en fonction de la fragilité des
échantillons ou de leur contenant.
Ainsi, la préparation de plasma riche en plaquettes requiert une accélération faible. En transfusion, il est
conseillé d'utiliser une pente d'accélération moyenne ou faible de façon à permettre aux poches de
"s'installer" au fond des nacelles sans éclater sous l'effet d'un effort trop brutal.
La table ci-dessous donne les durées d'accélération indicatives en fonction du type de rotor utilisé, jusqu'à
la vitesse nominale.

On dira, par exemple, que telle séparation s’effectue en centrifugeant l’échantillon dans telle machine (ce
qui définit l’accessoire, donc le rayon de centrifugation) pendant 10 minutes (durée) à 4000 x g (FCR).
Si le centrifugeur atteignait instantanément sa vitesse de consigne et freinait de même, et si la vitesse
demandée était effectivement atteinte et constante, la reproductibilité des résultats serait garantie d’une
centrifugation à l’autre.

Dans ces conditions, on comprend qu’il est équivalent de centrifuger un échantillon à 4000 g pendant
10 minutes que de centrifuger à 2000 g pendant 20 minutes (ou 8000 g pendant 5 minutes).
Autrement dit, les centrifugations ont le même effet lorsque le produit : DUREE X ACCELERATION est
constant. Ce produit correspond à la surface de la courbe.

L'intégrale de cette courbe se calcule en additionnant les surfaces élémentaires obtenues en effectuant le
produit (Intervalle de temps élémentaire x accélération appliquée pendant cet intervalle de temps).
Les courbes de deux centrifugations successives pour lesquelles ont été appliquées la même vitesse et la
même durée ne sont pas identiques. En effet, suivant la masse des échantillons, la température, l'humidité
de l'air de la cuve, les variations de la tension secteur et l'usure du moteur, la pente d'accélération peut
varier ainsi que la vitesse correspondant au plateau.
Lorsque les durées des phases de plateau sont très faibles ou très grandes, les fluctuations précédemment
décrites peuvent engendrer des variations de la valeur de l'intégrale de l'ordre de 5 à 10%. La qualité des
séparations obtenues en est parfois fortement altérée, comme, dans le domaine de la transfusion sanguine,
dans le cas de la préparation de plasma riche en plaquettes.
Le module de commande du centrifugeur KR4i permet de s'affranchir des différentes fluctuations
possibles. En effet, au lieu de programmer une durée de centrifugation, il est possible de fixer la valeur
souhaitée de l'intégrale pour une vitesse ou une accélération donnée.
Lorsque cette valeur sera atteinte, la phase de freinage du centrifugeur sera automatiquement
commandée.
Le calculateur du centrifugeur KR4i fournit les valeurs de l'intégrale de centrifugation au cours du temps,
même dans le cas où le programme sélectionné fonctionne avec une consigne de durée. Ainsi, la mise au
point des méthodes de centrifugation et la determination des valeurs d'intégrales à introduire s'en trouvent
largement simplifiées.
Le travail avec intégrale permet une meilleure reproductibilité des manipulations.

Les centrifugeurs KR4i sont équipés d'un système de réfrigération qui permet de maintenir les échantillons
à une température inférieure à la température ambiante. La mesure effectuée dans la cuve est numérisée
et le contrôle de la température est géré par le microprocesseur qui commande le groupe frigorifique.
L'opérateur peut programmer une compensation de température de façon à ce que la température finale
de l'échantillon soit obtenue avec plus de précision. Elle se programme en fonction de la différence entre
la température de l'échantillon obtenue pendant le cycle et la température programmée.
Suivant le modèle choisi, le condenseur du groupe frigorifique du KR4i est refroidi soit par air, soit par
eau.

5 INSTRUCTIONS D’UTILISATION
Les informations données ci-dessous sont succinctes. Veuillez vous référer au manuel spécifique
d’utilisation des centrifugeurs en transfusion sanguine (réf. 89001173).
Les paniers sont des produits consommables qui doivent être remplacés après quelques centaines de
cycles suivant leur état.
Les paniers à cloison centrale doivent être chargés symétriquement de part et d'autre de la cloison. La
dissymétrie ne doit pas excéder 100 grs sous peine de déformation irréversible du panier. On peut
utiliser pour compléter un chargement des poches de transfert remplies d'eau.
Les paniers souples sont toujours disponibles aux utilisateurs actuels. Il est conseillé aux nouveaux
utilisateurs de travailler avec les paniers semi-rigides qui sont plus facile à manipuler.
Paniers à cloison
Placer une poche dans chaque compartiment.
Les deux poches doivent avoir à peu près la même masse de façon à ce que le centre de gravité de
l’ensemble se situe au niveau de la cloison centrale. Ceci assure un plan de vol horizontal qui garantit une
centrifugation correcte des poches. Lorsque le nombre de poches à centrifuger est insuffisant pour
charger tous les compartiments, compléter avec des poches factices remplies d’eau.
Frapper le panier sur une paillasse pour bien enfoncer les poches et s’assurer qu’elles touchent le fond du
panier.

APPLI par un mot de passe si nécessaire.
STOP Enclenche le freinage du rotor. C’est une touche d’arrêt manuel qui annule la fin
du programme en cours.
NOTE : Lorsque l’appareil est sous tension, l’économiseur de l’écran est activé automatiquement après
5 minutes de non utilisation. L’écran rétroéclairé s’eteint mais l’affichage reste visible. L’écran se réallume
lorsqu’on appuie sur n’importe quelle touche du clavier.
5.3. UTILISATION
5.3.1. INTRODUCTION
Le système de programmation VIDEOset allie simplicité et puissance. VIDEOset inclut une visualisation
des paramètres sur écran graphique et une aide à la programmation.

Si le programme ne convient pas, le rappel d’un programme mémorisé s’effectue de la façon suivante :
soit sélectionner le numéro directement du pavé numérique, soit appuyer sur DETAIL
APPLI # = tous les
paramètres du programme actif sont affichés.

Puis appuyer sur APPLI # pour faire défiler les différents programmes mémorisés. Maintenir la touche
enfoncée pour faire défiler plus rapidement les programmes.

Remarque : Si le rayon n’a pas été defini par la sélection du rotor (et éventuellement par la modification de
son rayon) l’écran détaillé n’affichera pas le nombre de g (valeur 0).
Il est impossible d’appeler directement un programme ou composant à l’aide du pavé numérique.

Rentrer directement la valeur souhaitée à l’aide du pavé numérique. Note : ceci n’est possible que si un
rotor a été sélectionné, le nombre de g étant proportionnel au rayon de centrifugation (distance séparant
l’axe de centrifugation de la base du tube : voir chapitre 1.2).
Pour sélectionner un rotor, appuyer sur la touche ROTOR et utiliser la touche !pour déplacer le curseur
sur la liste des rotors. Si le rotor est utilisé avec des adaptateurs, appuyer sur la touche “RAYON” et entrer
le rayon à l’aide des touches - et + (cf tableau accessoires, 3.4). Valider avec la touche ENTREE. Lors de la
modification du rayon, le nombre de g correspondant est affiché automatiquement.
Pour modifier les pentes d’accélération et de freinage, appuyer sur la touche ACC ou DEC et utiliser les
touches - et + pour sélectionner les pentes désirées. Les profils d’accélération et de freinage sont affichés
sur l’écran graphique.
Appuyer sur ENTREE pour retourner à l’écran des paramètres liés à la vitesse.
Une fois le rotor, le rayon et les pentes d’accélération et de freinage programmés l’opérateur peut
sélectionner la fonction MAX. Cette touche entre automatiquement la valeur maximale de la vitesse (et du
nombre de g correspondant au rayon programmé) pour le rotor choisi.
Appuyer sur ENTREE pour valider toutes les modifications apportées aux paramètres liés à la vitesse et
revenir à l’écran simple de programmation (qui peut indiquer un contrôle par FCR).
Appuyer deux fois sur ECHAP pour retourner à l’écran durée / intégrale. Si le programme actif vient d’être
utilisé, VIDEOset affiche la valeur de l’intégrale calculée lors de ce cycle de centrifugation.
L’appui sur ENTREE permet de retourner à l’écran simple de programmation : l’intégrale de centrifugation
remplace la durée pour une reproduction parfaite des résultats d’un cycle à l’autre (cf 4.4).
Note : ceci n’est possible que si un rotor a été sélectionné, l’intégrale étant lié à la valeur du nombre de ‘g’
qui est proportionnel au rayon de centrifugation du rotor.
Ou bien pour sélectionner la durée au lieu de l’intégrale, déplacer la flèche clignotante à l’aide de la
touche en face de la durée et entrer une valeur ou choisir INFINI pour une utilisation en continu et
appuyer sur ENTREE pour valider et retourner à l’écran simple de programmation.
FIN
Pour utiliser les paramètres modifiés sans les sauvegarder appuyer sur MODIF .
Pendant un cycle contrôlé par l’intégrateur, la valeur de consigne de la durée sera indiquée comme
REPROductible. La valeur de l’intégrale est alors décrémentée. Le temps correspondant est incrémenté.

En cours de centrifugation
A partir de l’écran de rotation de base ou de l’écran de rotation détaillé appuyer sur MODIF. Les trois
paramètres principaux (vitesse ou nombre de g, température, durée ou intégrale) sont affichés. Déplacer
le curseur à l’aide de la touche et entrer la valeur souhaitée à l’aide du pavé numérique.
FIN
Appuyer sur MODIF pour revenir à l’écran d’origine. Les valeurs modifiées en cours de centrifugation ne
sont pas mémorisées à la fin du cycle.
NOTE : Il n’est pas possible de modifier la valeur de l’intégrale pendant un cycle.

5.3.9. CONFIGURATION
Le KR4i peut être configuré selon les besoins de l’opérateur. Sélectionner CONFIG sur l’écran de veille.
A) Programmation de la langue
“LANGUAGE SETTING” est affiché en inverse vidéo. Appuyer sur ENTREE.
Choisir 1 pour l’ANGLAIS et 2 pour le FRANÇAIS.
L’écran de veille sera affiché dans la langue sélectionnée.
C) Aide à la maintenance
Un code spécial donne accès à cet écran réservé aux techniciens de service après-vente.

+ Si nécessaire, saisir le code d'identification Opérateur à l'aide du clavier. Valider en appuyant sur
"ENTREE" ou Annuler en appuyant sur "ECHAP".
+ Pour identifier les paniers à centrifuger, utiliser le lecteur code à barres du centrifugeur. Saisir avec
celui-ci le code d'identification du premier support et valider en appuyant sur "ENTREE" et ainsi de suite
jusqu'au dernier support (6 au maximum sur version V3.00).

5.3.11. SECURITES
En cours de centrifugation, des anomalies de fonctionnement peuvent survenir. L’écran affiche en clair la
nature du dysfonctionnement.
IMBALANCE : Des vibrations importantes risquant d’endommager l’appareil, ses accessoires, ou les
échantillons ont déclenché la sécurité. La cause en est l’équilibrage incorrect des portoirs. Dès l’arrêt de
l’appareil, ouvrir le couvercle, rééquilibrer la charge par rapport à l’axe d’entraînement du rotor et, pour
les rotors swing-out, par rapport à l’axe de pivotement des nacelles et démarrer une nouvelle
centrifugation.
S’assurer que les tourillons des nacelles sont bien graissés. Ajouter un peu de graisse (flacon fournis avec
l’appareil) après avoir nettoyé l’ancienne graisse avec un papier absorbant. NOTE : Ne jamais utiliser des
nacelles provenant de jeux différents.
Si le message réapparaît, faire intervenir un technicien.
LID UNLOCKED : le couvercle n’est pas correctement fermé. L’appareil vous demandera de le fermer.
TACHOMETER FAILURE : le système de contrôle détecte un défaut du système de mesure de vitesse. Par
exemple des valeurs de vitesse très différentes lors de deux mesures consécutives, ou en cas de
microcoupure secteur. La vitesse sera mesurée immédiatement après rétablissement de l’alimentation.
Après arrêt du moteur, une minuterie est enclenchée “TIME OUT”, n’autorisant le déverrouillage du
couvercle qu’après un délai de sécurité.
MOTOR OVERTEMPERATURE : Une utilisation dans des conditions incorrectes ou une défaillance du
moteur ont permis à la température du moteur d’atteindre le seuil de sécurité, le cycle de centrifugation
est automatiquement arrêté.
CHAMBER OVERTEMPERATURE : Une utilisation dans des conditions incorrectes ou une panne de la
réfrigération a permis à la température de la cuve de dépasser 45°C. Le cycle de centrifugation est
automatiquement arrêté.
D’autres sécurités intrinsèques ne font pas l’objet de messages mais garantissent l’usage du centrifugeur :
- sécurité de survitesse
- limitation de courant
- blindage mécanique.

Corrosion Chimique
Elle est dûe à la présence de produits chimiques ioniques à la surface du rotor, susceptibles de réagir
avec les matériaux constituants du rotor. L’aluminium est un métal qui réagit facilement avec les solutions
ioniques. L’anodisation est une couche obtenue par traitement de l’alliage qui améliore sa résistance en
surface.

La présence prolongée de produits ioniques sur le rotor finit par attaquer l’anodisation (décolorations
visibles le plus souvent), mettant à nu l’alliage d’aluminium qui se corrode à son tour, provoquant, au
premier stade, des traces de piquage sur le métal, puis, en cas d’excès de produit ionique, des sels
d’aluminium vont apparaître, blanchâtres, donnant au métal, un aspect “rongé”.
Les produits chimiques à l’origine de ce phénomène de corrosion ne proviennent pas forcément d’un
écoulement accidentel dû à un tube brisé par exemple. Il peut s’agir :
- De vapeurs chimiques présentes dans le laboratoire qui se dissolvent dans l’humidité résiduelle, dans
l’eau de condensation (centrifugeurs réfrigérés) présente dans le fond des alvéoles.
- De liquides corrosifs provenant de tubes non bouchés trop remplis : le liquide déborde en cours de
centrifugation
- De portoirs, adaptateurs, racks, flacons dont l’extérieur a été souillé par un produit chimique ou mal
rincés après décontamination (eau de Javel).
Note : si les produits sont très corrosifs, un simple essuyage est insuffisant : les traces résiduelles se
dissolvent peu à peu dans l’humidité présente dans l’alvéole ou dans l’eau de condensation (portoir à
température ambiante placé dans une nacelle réfrigérée).
ATTENTION à la présence de particules solides sous le fond des tubes, portoirs, racks ou adaptateurs.
Ces particules sont écrasées par la force centrifuge et traversent la couche protectrice des nacelles et
rotors (anodisation), créant ainsi des passages faciles.

6.2.2. NETTOYAGE
Les utilisateurs ne devront pas utiliser de méthodes de nettoyage ou de décontamination differentes
de celles recommandées sans avoir au préalable vérifié que ces méthodes ne risquent pas
d’endommager l’appareil et ses accessoires et sans avoir reçu l’accord du constructeur.
Idéalement, les rotors devraient être lavés après chaque utilisation ; une fois par semaine ou après 10
utilisations est suffisant. Toutefois, le nettoyage immédiat est indispensable si le rotor a été en contact avec
un échantillon, un solvant ou tout autre produit chimique. N’utiliser que des détergents non alcalins.
Ne pas oublier de nettoyer la partie centrale du rotor qui vient en contact avec la tête d’entraînement. Pour
les alvéoles des rotors, les inserts pour tubes et les réducteurs, utiliser une brosse à poils synthétiques afin
de ne pas endommager le matériau.
NE JAMAIS UTILISER DE BROSSE OU D’EPONGE METALLIQUES.
Rincer le rotor à l’eau claire, voire à l’eau distillée.
Essuyer entièrement, en particulier dans les endroits difficilement accessibles : les fonds d’alvéoles, les
parties taraudées ou filetées. Un séchage complet des parties peu accessibles est obtenu par un soufflage
d’air chaud, avec un sèche-cheveux par exemple.
ASSUREZ-VOUS QUE LES ALVEOLES SONT BIEN SECHES.
Pour les rotors de type swing-out, retirer la graisse des tourillons et des portées de tourillons et la
remplacer avec une faible quantité de graisse neuve. Cette opération assure le libre basculement des
nacelles : la quasi totalité des balourds est dûe à la friction des nacelles.
NE LAISSEZ JAMAIS UN ROTOR HUMIDE SUR UNE SURFACE METALLIQUE même en acier inoxydable. Il
peut se produire des réactions électrochimiques qui entraîneraient la corrosion du métal.

Quelques exemples de décontamination sont présentés ci-dessous. Les rotors doivent être entièrement
désassemblés avant tout traitement à la chaleur et après tout traitement chimique. Les joints, les tubes
doivent également être décontaminés.
Désinfection
Bactéries et virus : 10 minutes de contact avec de l’éthanol à 70°. L’autoclavage pendant 20 minutes à
120°C détruit la plupart des micro-organismes. Il peut s’ensuivre une décoloration partielle de
l’anodisation des rotors en alliage d’aluminium. Démonter le rotor : les joints, le couvercle doivent être
autoclavés séparément du rotor. Les joints déformés par la chaleur doivent être remplacés.
TOUTE PIECE QUI A ETE PORTEE A PLUS DE 130°C DOIT ETRE CHANGEE.
L’eau de Javel, diluée à 0,1 %, est efficace après immersion de 10 minutes, contre les spores, les virus et
les bactéries mais est agressive pour les alliages d’aluminium. Ne pas l’utiliser pour des rotors dont
l’anodisation a été endommagée. Un rinçage abondant à l’eau claire suivi d’un séchage complet, est
indispensable.
Le formol à 37 % est également très efficace par immersion. Cependant, comme l’eau de Javel, le Formol
est agressif pour les alliages d’aluminium. Un rinçage abondant à l’eau courante pendant au moins 5 mn,
suivi d’un séchage complet, est nécessaire. ATTENTION : LE FORMOL EST TRES IRRITANT POUR LES
MUQUEUSES.
Le glutaraldéhyde à 2 %, vendu sous des noms de marques telles que CIDEX et GLUTAREX, suppose
l’immersion totale pendant 10 mn pour obtenir la stérilisation complète. Rincer abondamment puis sécher.
ATTENTION : LE GLUTARALDEHYDE PEUT S’ACCUMULER DANS LES RESERVES ADIPEUSES DE
L’ORGANISME.
Le phénol et ses dérivés sont à proscrire.

Substances dangereuses :
Il est recommandé à l’utilisateur de prendre toutes les précautions nécessaires lors de la centrifugation de
substances connues pour leur toxicité, radioactivité ou lorsque elles sont contaminées par des micro-
organismes pathogènes (et notamment pour les matières du groupe Risque II identifié dans le “Manuel de
Sécurité Biologique pour Laboratoire” édité par l’Organisme Mondial de la Santé). Voir aussi chapitre
6.2.3.
Le phénomène :
Ce phénomène physique normal apparaît dès qu’un effort mécanique est appliqué, relâché puis répété.
Cette accumulation d’alternances effort / repos (nombre de cycles) se traduit à partir d’un nombre donné
de cycles par une baisse très rapide des caractéristiques de résistance mécanique : l’objet ne peut plus
supporter autant d’efforts qu’au début. Si l’effort est répété avec la même intensité, il y a rupture.

C’est ce phénomène de fatigue exagéré qu’on utilise lorsqu’on plie et déplie plusieurs fois un mince objet
(un morceau de fil de fer, une pièce de plastique) pour le briser.
Chaque cycle de centrifugation est un cycle de fatigue pour les nacelles.
La durée de vie :
Les nombres de cycles au bout desquels des objets identiques soumis aux mêmes efforts, se briseront
varient dans une large fourchette. Dans le cas de nacelles de centrifugeurs, plusieurs milliers de cycles
pourraient séparer les ruptures de 2 nacelles identiques soumises aux mêmes efforts. On est dans une
situation dite "probabiliste". Les conséquences d’une rupture de nacelle étant graves, il convient de situer
la limite du nombre de cycle dans la zone où le risque est considéré comme nul. Il est recommandé à
l’utilisateur de remplacer les accessoires (rotors et nacelles) ayant plus de 16 000 cycles (et 10 ans d’age).
Une attention particulière sera portée aux nacelles NHK4B limitée pour ces raisons à 9000 cycles.
Bien que l’installation de rotors et de nacelles autres que ceux décrits dans ce manuel soit interdite sur le
KR4i, il apparait important de donner l’exemple des nacelles de transfusion P6 (réf. 11178112) et P4 (réf.
11178074) dédié au KR4.22. On a considéré que chaque cycle est effectué avec une charge maximale en
poches transfusion.
L’utilisateur doit impérativement conserver l’appariement du centrifugeur avec ses
accessoires (rotors et nacelles) afin de pouvoir suivre plus aisément l’évolution du nombre de
cycle de cet ensemble (un compteur de cycles équipant chaque centrifugeur).

Remarques :
Les courbes montrent que les centrifugations "douces", programmées pour l’obtention de PRP et des
plaquettes à partir du Buffy-coat, jusqu’à 1500 g (soit environ 2200 trs/mn) n’entraînent aucun effet de
fatigue. Donc le nombre de cycles effectués basse vitesse n’est pas à considérer pour l’appréciation de la
durée de vie des nacelles.
Des tensions électriques élevées existent derrière les panneaux du KR4i. Aucune opération ne requiert
l’intervention de l’utilisateur à l’intérieur de l’appareil. Seuls les techniciens habilités sont autorisés à
intervenir dans les règles de l’art. Ne pas retirer ces panneaux avant de débrancher le centrifugeur de la
prise de courant.

- Ne jamais faire fonctionner un rotor ni ses accessoires à une vitesse de rotation supérieure au maximum
indiqué dans le chapitre 3.
- Ne pas essayer de supprimer l'effet du système de verrouillage du couvercle.
- Utiliser uniquement des accessoires conçus pour le KR4i.
- Equilibrer correctement les charges (voir 2.9 et 5.1) et ne pas dépasser le niveau maximum indiqué sur
le rotor ou les nacelles.
Par l’effet de ventilation qu’il crée dans sa chambre, un centrifugeur est un générateur d’aérosols. Dans le
cas de centrifugation de produits présentant un risque biologique, il vous appartient d’une part de
prendre les dispositions matérielles qui préviennent ou réduisent ce risque et d’autre part de former les
utilisateurs à l’usage de cet appareil et de ses accessoires dans le contexte de leurs applications.
Les informations ci-dessous méritent d’être divulguées aux utilisateurs, même occasionnels :
- Un centrifugeur ventilé (sans réfrigération interne) expulse en permanence dans la pièce l’air qui a
traversé la chambre tandis qu’un centrifugeur réfrigéré, voire thermostaté (chambre froide) possède
une chambre relativement étanche. Les risques de diffusion d’aérosols sont donc minimisés dans un
centrifugeur réfrigéré et importants dans un centrifugeur ventilé.
- Des aérosols se produisent naturellement avec des tubes ou flacons non bouchés. Ils se produisent
également lorsqu’une déformation voire une casse provoquent la fuite lente ou brutale de l’échantillon.
Nous attirons votre attention sur le cas des tubes utilisés dans des rotors angulaires : si, en statique, le
tube étant en position verticale, le niveau du liquide n’atteint pas le bouchon, il peut l’atteindre au cours
de la centrifugation et fuir. Il faut donc suivre les recommandations du fournisseur des tubes en ce qui
concerne le niveau de remplissage et l’étanchéité du tube.

Dès qu’un risque biologique est connu ou soupçonné, les échantillons sont à placer dans des conteneurs
fermés. Si de tels conteneurs ne sont pas disponibles, il faut utiliser les accessoires étanches tels que
nacelles à couvercle étanche ou rotor angulaire à couvercle étanche. Les nacelles N6BMT (11178140)
sont certifiées étanches (CAMR - Porton Down). Les nacelles N6B (11178138) sont étanches si elles sont
équipées d’un couvercle 11178216.
- Durée d’un aérosol : lorsqu’un aérosol est créé dans une chambre de centrifugeur il peut persister en
suspension dans l’air pendant 3 à 5 minutes après l’arrêt du rotor. La manoeuvre d’ouverture du
couvercle provoque elle-même la dispersion de l’aérosol dans l’environnement.
- Lorsqu’un risque élevé d’aérosol ou de casse est décelé, il faut manipuler les accessoires, même s’ils
sont apparemment étanches avec des gants et les ouvrir dans un environnement assurant la protection
de l’opérateur (hotte, boîte à gants, masque ...).

Il n'y a pas de nettoyage quotidien à faire, sauf en cas de casse accidentelle de verre ou de projections
importantes de produit dans la chambre du rotor ou dans le carénage.
Si l'utilisateur souhaite nettoyer la face avant du module de commande, il faut :
- mettre hors tension le centrifugeur,
- nettoyer à l'aide d'une éponge humide imbibée d'un peu de produit savonneux et rincer avec une
éponge humide. Sécher avec un chiffon sec et non pelucheux et en cas de nécessité de désinfection,
utiliser de l'alcool à 70°.

- Vérifier l’aspect des accessoires utilisés (rotors, nacelles, portoirs, couvercles étanches) - se reporter au
chapitre 6.
- Vérifier la continuité de la mise à la terre de protection (pour toute intervention à l’intérieur du
centrifugeur).
Le changement des fusibles ne doit pas être effectué par l’opérateur mais par un technicien agréé qui fera
un diagnostic avant de décider de leur remplacement.

1.1. DESCRIPTION
The KR4i centrifuge is designed to separate substances of different density by centrifugal force.
Different rotors can be fitted to the drive in order to match the sample containers and the required
performance. The maximum speed (and, by consequence, the resultant centrifugal force) is a function of
the rotor in use.
A control system permits the speed, acceleration and braking rates, run time and temperature to be
monitored and controlled.
The refrigeration is assured by a refrigerant group charged with a CFC-free fluid. The refrigerant fluid
circulates around the bowl to achieve maximum efficiency.
The KR4i is mainly dedicated to the following application fields :
- Blood separation processing
- Large volume industrial processing
- Biochemistry
- Radio immuno assay

Suspended solids are separated by the Relative Centrifugal Force (RCF) applied to the sample. The
effective force increases with the square of the speed of rotation and the distance from the axis of rotation.
The following formulae permit the calculation of primary parameters and of transformations relating to
changes in primary parameters.
NOTE : The value introduced for the radius can be adjusted to allow for position within the tube such as at
a boundary. Maximum radii are quoted in the specifications tables (chapter 3).

2 INSTALLATION PROCEDURE
Due to the weight of the machine, all lifting and transporting must be carried out using proper handling
equipment (eg : fork lift trolley) that complies with current regulations, and by people having undergone
the necessary training.
The machine must be supported from underneath. If it has to be transported without its pallet, for
example on a staircase, professional handling assistance is required.
- Indoor use.
- Temperature : 5° to 40°C.
- Maximum relative humidity of 80% for temperatures up to 22°C.
- Maximum altitude : 2000 m.
Maximum performance is assured across the following ambient temperature range : 15°C to 25°C.
Place the centrifuge and its accessories in a clean, non-corrosive environment to avoid damage.
Install the unit leaving at least 20 cm (8 in.) clearance between its back and any obstruction, to allow
subsequent placement of a vessel to collect condensates. Likewise, leave approximately 10 cm (4 in.)
clearance between two adjacent machines.
Centrifuges should be placed preferably in a room specifically intended for their use and therefore
containing no other apparatus.
Walls and partitions that are within a distance of 1 metre should be strong enough to stop the centrifuge
should there be a major accident (mobile partitions, single-width chipboard and plasterboard partitions
without reinforcement are not strong enough).
The floor must be completely flat and strong enough to support the weight (346 kg) of the machine without
creating or maintaining any vibrations. Vibrations may be caused by variations during centrifugation. See
also chapter 2.5 for positioning the damping feet.
There are no set minimum specifications on the type of floor required as there are many technical factors
to be taken into account, such as :
- Composition of the different layers of the floor,
- How the floor is joined to the wall,
- Structures supporting the floor,
- Positions and weights of loads already on the floor,
- Loads already existing on support structures.
The machines must be positioned so that they can be moved easily to enable regular access for
maintenance.

WARNING : Read the following instructions carefully before installation. During manipulation, never put
your hand underneath the machine.
- Remove staples with a screwdriver.
- Remove both anchor bolts (A) using the 19 mm spanner/wrench supplied in the tool box.
- Remove the front anchor bolt (B).
- Using the prop (C) as a lever :
. Remove the wooden beam and portable ramp (D) by lifting the machine at the back.
. Remove the wooden beam and portable ramp (E) by lifting the machine at the right hand side, towards
the front.
- Ensure that the ramps (F) are perpendicular to the pallet and in line with the wheels.
2 people are needed to unload the machine : 1 person at the front of the centrifuge to guide and brake the
machine as it comes down from the pallet, a second person behind to guide the machine onto the ramps.

The electrical supply circuit must conform to currently applicable regulations. The KR4i centrifuge is
delivered without an electrical plug ; this must be connected by a qualified engineer according to the
following conductor colour code :
- Blue : neutral, brown : phase, yellow/green : ground
Depending on the equipment part number, the electrical supply should be :
- 230 V ±10 % 50 Hz 25 A max.
- 208 / 230 V 60 Hz 25 A max.
Remember that in order to respect the electrical safety standards related to protection of operators against
indirect contact, the supply of power to the instrument must be via a power socket fitted with a protection
device ensuring automatic cut-off in the case of an insulation fault. A supply fitted with a cicuit breaker of
the correct rating complies with this requirement. It is also recommended that different machines be
connected to a circuit breaker that is situated outside the room.
It must be immediately possible to identify the plug of the machine, which must also be easy to
reach by the user.

The KR4i is equipped with a double locking safety device which stops the machine starting if the lid is not
completely closed and prevents lid opening before the end of rotation.
When the centrifuge is switched on, wait until display returns to the stand-by mode then pull forwards the
latch lever located on the right hand side of the unit : the lid is automatically unlocked and opens.
In the case of a mains power cut, opening of the lid is prevented by the lid locking safety device.

It is recommended to wait for the mains to be switched back on so that this safety device enables the lid to
be unlocked.
During rotation, the slightest leak from a sample is enough to create aerosols. Rotation of the rotor can only
be detected when the machine is switched on. Opening the lid manually if the rotor is still rotating, would
disperse the aerosols in the environment.
Rotating parts are also a risk as these could come in contact with the user or be ejected. There is
particularly high risk of injury if :
- The user tries to manually stop the rotor,
- Any object falls inside the centrifugation chamber.

2.7. INSPECTION
Before installation, the rotor should be thoroughly inspected for corrosion and cleanliness.
Chemical and stress corrosion will eventually lead to disruption of the rotor with potential severe damage
to the centrifuge.
Particles stuck inside the pockets can cause breakage of tubes and lead to major imbalance and / or loss of
sample and contamination.
Check that the central hole of the rotor and the drive spindle are clean and undamaged. Remove any
particles with a soft cloth.
Refer to chapter 6 : Hazards, Precautions and Limitations of use
Rotor installation must only be undertaken by qualified and authorized personnel. Never lift a
rotor by its windshield.

The packing contains :
. a rotor,
. grease for trunnions and shaft,
. an instruction sheet.
- Check cleanliness of the motor drive shaft and rotor hub.
- Apply a light film of grease to the motor shaft.
- Note collar position on the shaft and the groove on the hub.
- Pick up the rotor by two opposite arms and place it on the drive shaft, ensuring shaft and hub alignment.
Position the rotor gently, despite its weight. The rotor slides freely onto the drive shaft.
- Position the washer and the bolt and screw clockwise with the wrench supplied.
CAUTION : Never tighten the rotor bolt without the washer.
- Apply a light film of grease to the tip of the trunnions.

Ensure that the accessories are correctly fitted and balanced.
- Check that the rotor is fully locked onto the drive shaft.
- Check that the rotor lid fittings are screwed on tightly.
- Check that there is no water nor other liquid in the base of the windshield.
- Check that all of the buckets are in place and that they swing freely.
- Check that there are no empty transfusion liners in the buckets.
When installing tubes into each bucket insert, be certain that the samples are distributed symmetrically.
Swing-out rotors should be loaded with a full compliment of buckets to maintain correct balance and safety
for the rotor.
CAUTION : Use only centrifuge-rated glass or plastic ware, tubes, vials or bottles. Although the KR4i can
compensate for an imbalance of more than 50 grams, sample containers should be balanced to avoid any
long-term effects of vibration and assure good sample separation. If you intend to use old accessories on a
new centrifuge, please contact your JOUAN representative for advice.
WARNING : a major imbalance can seriously damage the rotor and the centrifuge.

4 OPERATING PRINCIPLES
The rotor is driven by a three phase asynchronous motor. The rotor is situated in the centre of an armour
plated centrifugation bowl. A control system ensures the correct drive.
4.2. ACCELERATION
The KR4i centrifuge allows a choice of 10 different acceleration rates, thus permitting variation of the time
for the centrifuge to attain the desired speed.
If the centrifuge is used for blood transfusion purposes, the choice of an intermediate acceleration rate
allows optimal blood bag positioning in the buckets, thus limiting the risk of bursting and formation of
wrinkles. Moreover, to carry out selective separations, such as separation of platelets, it is possible to
improve the results by adjusting the acceleration rate.
The table below shows approximate acceleration times according to rotor type and maximum speed.

For example, a given separation may be described as obtainable by centrifuging the sample in a given
machine (which determines the accessory, thus the centrifugation radius) for 10 minutes (duration) at
4000 x g (R.C.F.)
Theoretically, if the centrifuge instantly attains the set point speed and brakes instantly, and if the requested
speed attained is constant, reproducibility of results is guaranteed from one centrifugation to the next.

The integral of this curve is calculated by adding the elementary areas obtained by calculating the product
(elementary time interval x acceleration applied during this time interval).
The curve of two successive centrifugations for which the same speed and duration are applied are not
identical. Indeed, according to sample weight, temperature, atmospheric moisture in the chamber, mains
voltage fluctuations and motor wear, the acceleration rate, as well as constant speed phase, may vary.
When constant speed durations are very limited or very long, the previously described fluctuations may
give rise to 5 to 10% variations in value of the integral. The quality of the resultant separations is sometimes
considerably modified, as for example when preparing sub cellular fractions.
Integration control allows suppression of the effect of the various potential fluctuations. Indeed, rather than
programming a given duration of centrifugation, it is possible to set the desired value of the integral for a
given speed or centrifugal acceleration level (R.C.F.).
Upon attainment of that value, the centrifuge braking phase is started automatically.
The centrifuge calculator always provides values of the centrifugation integral over time. Thus, the
optimization of centrifugation and determination of integrals to be entered is considerably simplified.

The KR4i is equipped with a refrigeration system which allows the rotor chamber temperature to be
controlled at a lower level than the ambient temperature. The measured value of air temperature in the
bowl is displayed and the temperature is controlled by the microprocessor which controls the cooling unit.
The operator can use the temperature compensation setting so that the sample temperature is more
accurately controlled according to the user request by measuring the difference between the sample
temperature obtained during the run and the set value.
The cooling unit of the KR4i has either an air cooled condensor or a water cooled condensor.

It is essentiel to understand the precautions, risks and limitations of use described in chapter 6
to achieve the best performance and highest safety with the instrument and its accessories.
The information given below is succinct. Please refer to the manual specific to the use of centrifuges
in blood transfusion centres (Cat N°. 89001172).
Liners are consumable items which must be replaced after several hundred runs according to their
condition.
Liners with partition must be loaded symetrically about the partition. The load on one side must never
exceed the load on the other side by more than 100 g under risk of permanent damage to the liner. Bags
filled with water can be added if necessary to balance the load.
Supple liners are available for existing users. New users are advised to use the semi-rigid liners which are
easier to manipulate.

The mains switch is located on the lower left side of the front panel.
The control system includes :
- A dot-matrix graphic LCD screen, able to display numbers, letters, and special characters in addition to
animated graphics.
- Eight keys, surrounding 2 sides of the screen, have their functions determined according to the name
displayed on the adjacent part of the screen.
Not all keys are necessarily operational at any one time and more than one key can operate the same
function when the name of the function is displayed across a wide area of the screen next to these keys.
- In addition, a standard numeric keypad (0 to 9 and CE, clear entry) enables parameter values to be
keyed in directly when required.

Certain key functions are very common : others are found only on a dedicated screen. They operate as
follows :
+ Increases the value of the indicated parameter. Sustaining pressure on the key
causes a rapid increase.
+/- Changes the arithmetical sign of the current figure (from positive to negative or
vice versa).
- Decreases the value of the indicated parameter. Sustaining pressure on the key
causes a rapid decrease.
ABOUT Shows a graphical representation of the integral, together with values calculated
INTEGRAL from the previous run (where possible).
ACC Enables the acceleration rate to be altered between the 10 choices from
0 (slowest) to 9 (fastest).
BASIC Switches the display from the fully detailed run screen to the basic run screen
DISPLAY with 3 fundamental parameters.
BRK Enables the braking rate to be altered between the 10 choices from 0 (weakest)
to 9 (strongest).
DETAILS Gives access to those parameters associated with the indicated parameter.
DISPLAY Displays all preselected values of the program whose basic parameters are on
APPLI # screen.
END Returns to the original run screen after temporary values have been entered.
MODIF
END Returns to the stand-by screen, keeping currently selected values in volatile
PROG memory ready for use but not in permanent memory for later recall.
EXPAND Switches the display from the basic run screen to the detailed run screen which
DISPLAY displays all parameters.
MAX Directly chooses the highest possible speed or “g” force taking into account the
selected rotor and radius (cannot operate without prior rotor selection).

& SAVE has not been already saved.
MODIF(Y) Enables parameter values to be changed (Values changed during a run other
than PRE COOL will not be memorized).

NEW Gives access to the selection of new parameter values starting from a set of
APPLI default values.
PRE Starts a predetermined precooling cycle (2000 rpm, 4°C, 15 min). During this
COOL cycle all other programming facilities are operational.
RADIUS Enables the effective radius of a selected rotor to be modified in order to allow for the
use of an adaptor or to select a boundary at which a set “g” force can be applied.
ROTOR Lists the available rotors from which a selection can be made with their capacity
and performance.
SCROLL One press displays all parameter values of next used memory. Continued
APPLI # presses will display the values for all memorised applications. Also acts as
application (program) selection key.
NOTE : If the instrument remains switched on, but not used, then 5 minutes after the end of the last run the
screensaver operates. The backlight is extinguished although the display can still be read. Press any key
to light up the screen then continue with normal operation.
5.3. USE
5.3.1. INTRODUCTION
The VIDEOset graphics based control software is designed to combine power with simplicity. VIDEOset
includes on-screen guidance and intuitive programming.
The screens presented in this manual could differ slightly according to the version of the software
5.3.2. SWITCHING ON
Immediately after switching on, the screen displays the JOUAN logo, then the stand-by screen is displayed
showing the number of the last application program used and its basic parameters.

5.3.3. PRECOOLING
A keypress on CE will cause the display of the value of the actual temperature of the air in the bowl to the
left of the set value during the time that the CE key is pressed.
PRE
If the operator wishes to precool the bowl and rotor, he simply selects COOL having installed the rotor, and
the KR4i runs according to the pre-set values which are indicated on the stand-by display.
The display now indicates PRECOOL ON. The remaining precool time, real temperature and the dynamic
rotation symbol are displayed.
Uniquely, during a PRECOOL run, it is possible to access all programming facilities. The precool run can
be halted at any time using the STOP keys
Otherwise, another program can be recalled either by selecting the program number directly on the
DETAIL SCROLL
keyboard or by pressing APPLI # which first shows all details of the current program, then APPLI # (same
key) to scroll through the memorized application programs whose parameters are thus indicated on the
screen.
Sustained pressure on the key causes rapid scrolling.
STANDBY SCREEN
Once the required program is displayed, simply press a START key to run the program.

During a run, the basic screen displays the 3 basic parameters with both set and live values.
EXPAND
A keypress on DISPLAY presents a detailed screen showing all possible run parameters and the values of
BASIC
those selected, both set and live. A keypress on DISPLAY returns the basic screen.
Note : If the radius has not been defined by selecting a rotor (and possibly modifying its radius to allow for
the use of an adaptor) the screen will not display the RCF.
It is impossible to call up a program or parameter from the keyboard.
Advanced parameters
A) Speed related : RCF, acceleration, braking, rotor, radius
To program any of the speed related parameters, select DETAILS from the simple programming screen
when the flashing cursor !indicates SPEED. A screen appears with the speed related parameters and the
flashing arrowhead indicates RCF.

A value can be input via the keypad so long as a rotor has already been selected. (No selection of rotor
makes it impossible for the software to control by “g” force since this is directly linear with the radius of
the base of the sample tube from the centre of the drive axis - see 1.2)
To choose a rotor, select ROTOR and use !to move the cursor down the list of rotors to that which will be
used. If an adaptor will be used in the rotor select RADIUS and use the - & + keys to choose the effective
radius (see accessory table, 3.4.) and select ENTER. During operation with - &+ the dynamic radius arrow
moves and the related RCF is changed in real time.
If the user wishes to modify the acceleration and/or braking rates, he selects ACC or BRK and uses the - &
+ keys to choose the rate(s) indicated on the dynamic graphic screen Select ENTER to return to the speed
related parameter screen.
After selection of ROTOR (and maybe RADIUS, ACC, BRK) the operator has the option to select MAX.
Without requiring any knowledge on the part of the operator, selection of MAX causes the instrument to
program the top speed of the rotor and the related RCF allowing for actual radius.
After all the required parameter changes for this screen have been made, select ENTER to save the
changes and return to the simple programming screen (which could indicate control by RCF).
Select ESC/ESC to return to the time / integral screen. If this application has just been used, VIDEOset will
indicate the recently totalled integral value.
Selection of ENTER will return the user to the simple programming screen but with control by integral
instead of time, ensuring the highest possible run-to-run reproducibility (see 4.4)
Note : this will only be possible if a rotor has already been selected, the integral value being linked to the
‘g’ force which is calculated from the centrifugation radius of the rotor.
Alternatively, use to indicate time and enter a value or press HOLD for operation with no time limit.
Selection of ENTER makes a return to the simple programming screen.

To use the modified parameter changes without permanently storing them select PROG .
During a run controlled by the integrator, the preset value for time will indicate REPROducible, the integral
value will count down and the timer will count up the corresponding time.

B) During a run
From the basic or detailed run display, select MODIFY. The three main control parameters (speed or RCF/
temperature/time or integral) will be displayed together with the current set values. Use to move the
cursor ! to the parameter to be modified and enter the value via the keypad.

Select MODIF to return to the original run screen. Values modified during a run cannot be stored (such a
randomly modified run would be impossible to reproduce) but remain in the volatile memory until the end
of the run.
NOTE : It is not possible to modify the integral value during a run.

5.3.9. CONFIGURATION
The KR4i can be configured to operate in set ways. To set up the instrument, select CONFIG from the
stand-by screen.
A) Language programming
Having selected CONFIG, the screen highlights LANGUAGE SELECTION then select ENTER.
Select 1 for ENGLISH or 2 for FRENCH.
The stand-by screen will be displayed in the selected language.
Having selected CONFIG use to highlight STAND-BY REFRIGERATION ON/OFF then select ENTER.
Choose 1 for active refrigeration during stand-by and 2 for refrigeration OFF during stand-by.

If necessary, enter the Operator Code via the keyboard (the ENTER or ESC to abort).
After the identification of the liners by the PC, identify the liners to be centrifuged by the KR4i using the bar
code reader on the centrifuge. Read the ID of the first support then press ENTER. Continue up to the last
support (a maximum of 6 for BB-CQCS V3.0).

5.3.11. SAFETY
At any time when the instrument is able to detect and inform the operator of a fault condition the screen will
display a special message from among the following.
IMBALANCE : Important vibrations which can damage the accessories, the unit and the samples have
been detected. The cause is the bad balancing of the inserts. As soon as the motor stops, open the
centrifuge and redistribute the samples to produce an equal weight on diametrically opposite sides of the
rotor and, for swing-out rotors, on each side of the trunnion for each bucket.
Using the grease supplied ensure that the trunnions are regularly lubricated removing the old grease first
with a clean tissue.
NOTE : Never mix buckets from different sets on the same rotor.
If the message persists, call an engineer.
LID UNLOCKED : Displayed if the lid was not correctly shut. The instrument will inform you to close it
correctly in order that it can start the requested run.
TACHOMETER FAILURE : Displayed when an unrealistic speed situation is detected.
For example, if the measured speeds in two sequential measurement steps are very different, or if a
power cut occured or if the unit was switched off momentarily during a run, in which case speed would be
detected immediately upon switch-on. If speed is readable (certainly true in the later case) its real value
will be displayed and in every case a TIME OUT safety lock will be applied, preventing the user from
opening the lid for a set period after the speed reaches zero, or at least effects this lock-out period in the
case of no detected speed.
MOTOR OVERTEMPERATURE : When using the centrifuge in bad conditions (e.g. : motor failing) a sensor
detects an overtemperature in the motor and the run is automatically stopped.
CHAMBER OVERTEMPERATURE : When using the centrifuge in bad conditions or if the refrigeration
system is failing, the chamber temperature can seriously increase depending on the speed level and the
heat can damage the samples. At 45°C the run is automatically stopped.
Other safety features are also included in the centrifuge. There is no corresponding message but these
features ensure safe use of your centrifuge :
- overspeed safety
- current limitation
- armour plating

6.1. CAUTIONS
Chemical Corrosion
This corrosion is characterised by chemical reactions due to the existence of an electrolyte liquid on the
surface of the item. If these substances are allowed to remain on the surface the corrosion will almost
certainly occur. This produces first a discoloration of the anodisation then pitting of the metal. Aluminium is
a metal which easily reacts with ionic solutions. Anodisation produces a protective layer on the surface of
the rotor.

Acidic and alkaline solutions sustaining their pH level will create problems of corrosion in aluminium
rotors. Chlorides, present in salts or even in skin contact with the rotor are among the most aggressive and
harmful substances commonly found in the laboratory.
The chemical products which are the origin of this corrosion do not necessarily originate from broken
tubes - for example they could come from :
- Chemical vapours present in the laboratory which are dissolved in the residual humidity, in condensed
water (refrigerated centrifuges) present at the base of the rotor pockets.
- Corrosive liquids originating from overfilled uncapped tubes (the liquid overflowing during
centrifugation).
- Inserts, adaptors, racks, bottles whose exterior has been soiled by a chemical product or poorly rinsed
after decontamination (with bleach, for example).
NOTE : If the products are very corrosive, simply rinsing is insufficient. Residual traces dissolve little by
little in the humidity present in the bottom of the pocket, in the condensed water (such as occurs when
putting an ambient temperature insert in a cold rotor).
BEWARE of the presence of solid particles beneath tubes, inserts, racks or adaptors. These particles are
crushed by the centrifugal force and penetrate the protective, anodised layer of buckets and rotors, thus
creating easy pathways for corrosion.
Stress Corrosion
This term relates to the phenomenom of accelerated corrosion due to the effect of centrifugation when a
corrosive chemical is in contact with the alloy. From the time when the aluminium alloy has been attacked
by chemicals, stress corrosion begins to appear. As it is on a microscopic scale it is even more dangerous
than a macroscopic scale since it is invisible.
During centrifugation, chemicals responsible for corrosion are also submitted to the tremendous “g” force
which pushes them against the alloy. This close contact facilitates the chemical reaction which occurs
much faster than in a static situation. Moreover, centrifugal force is very directional thus corrosion under
stress creates, with a very small amount of corrosive product, straight microscopic fissures. Each
centrifugation run makes the chemical migrate further and further.
A fissure, although it is microscopic, is a cut in the metal, breaking the cohesion of the material. As one
weak link in a chain allows the chain to break, so the microfissures break the chain of resistance of the
accessory to centrifugal force.
As accessories are designed with high levels of safety, rupture does not occur as soon as the first
microfissures are produced.
Depending upon the location of the fissure, disruption may occur before it reaches the external surface of
the accessory. The fissure creates a weakness which makes the accessory less and less resistant to
mechanical fatigue.
The corrosion by a small amount of corrosive product does not disrupt the accessory but makes it
mechanically weaker and weaker until it disrupts due to both centrifugal effort and number of cycles.
Because stress corrosion is largely invisible, it is essential that rotors are scrutinised regularly paying
particular attention to susceptible parts such as the base of the pockets, the outer edges and the base of
the rotor. If fissures are suspected, it is forbidden to use the rotor until it has been examined by a
specialist.

6.2.2. CLEANING
Users must not use cleaning or decontamination methods different from those recommended without
first having verified that these methods do not risk damaging the instrument or its accessories, and
obtained prior authorization from the manufacturer.
Ideally, rotors should be washed after every use but at least weekly in warm water containing a few drops
of mild non alkaline detergent (domestic washing up liquid is ideal) and EVERY TIME AFTER A SPILLAGE
HAS OCCURRED.
Do not forget to wash the core of the rotor which comes into contact with the drive spindle.
Each pocket of the rotor must be washed thoroughly using a small nylon brush.
DO NOT USE METAL WIRE BRUSHES.
Once the rotor is clean, rinse it in running water, preferably distilled.
Dry the rotor with a soft absorbant non-woven cloth or tissue. Drying may be finished off with warm air jet
(e.g. a hair dryer).
MAKE CERTAIN THAT THE POCKETS ARE WELL DRIED.
For swing-out rotors, be sure to clean the grease from the trunnions and replace it with a small amount of
fresh grease. This will ensure that the buckets swing freely.
The vast majority of apparent imbalance problems arise from the failure of the user to clean and grease
the trunnions and not from instrument error.
Pay particular attention that no deposit remains in the bottom of the pocket because the pressure of a flask
or tube from above during centrifugation will significantly increase the chances of corrosion.
NEVER LEAVE A DAMP ROTOR ON A METAL SURFACE, PARTICULARLY STAINLESS STEEL because an
electrochemical reaction could take place with the aluminium or magnesium in the rotor.

Examples of commonly used techniques are outlined below. The information is given as a guide only. It is
the responsibility of the owner to use the most suitable procedure. The rotor should always be completely
disassembled before being subjected to heat and after external chemical cleaning. Seals, tubes and plastic
components should be decontaminated.
Disinfection
Alcohol (70% ethanol or isopropanol) applied for 10 minutes is ideal for bacteria and viruses. Autoclave
rotors for 20 minutes at 120°C to destroy micro-organisms. Rotor lids must be disassembled from rotor
bodies. ‘O’ rings, autoclaved separately, should be replaced when deformed.
ANY PART WHICH HAS BEEN SUBJECTED TO TEMPERATURES ABOVE 130°C MUST BE DISCARDED.
Hypochlorite “bleach” used at 0.1% concentration with 10 minutes immersion is effective against bacteria,
spores and viruses but, as an oxidising agent, is corrosive to metal alloys and must be thoroughly rinsed
off and dried. It should never be used if there is surface damage to the rotor. Note that the black colour on
the surface of the rotor will be gradually washed out if the rotor is regularly autoclaved or bleached. This
does not necessarily denote a degradation in the anodization.
Formalin (37% formaldehyde in water) in contact for 10 minutes has a similar effect to chlorine bleaches.
Rotors should be thoroughly rinsed under running water for 5 minutes to remove all traces of formalin then
dried completely. NOTE : FORMALDEHYDE IS TOXIC.
Gluteraldehyde 2%, sold under many brand names such as Cidex and Glutarex, requires total immersion
for 10 minutes to ensure sterility. Thorough rinsing and drying is essential to protect users.
CAUTION : GLUTERALDEHYDE BUILDS UP TO A TOXIC LEVEL IN THE FATTY TISSUES OF THE BODY.
Phenols are very corrosive and should never be used.
Radioactive decontamination
We recommend that all radioactive contamination be referred to your Radioactivity Safety Officer. Rotors
may be decontaminated by a mixture of equal volumes of :
a) Distilled water,
b) SDS diluted to 10%,
c) Ethanol diluted to 70%.
The rotor should then be rinsed with ethanol followed by distilled water and then dried completely. JOUAN
makes no claims as to the effectiveness of proprietary brands of decontaminating solutions.

The IEC 1010-2-020 standard relative to laboratory centrifuges recommends the user :
- To mark out a clearance envelope 30 cm around the centrifuge or establish special procedures for the
non entry of all persons or all dangerous materials into this space during the operation of the centrifuge.
- To provide an emergency stop switch enabling the mains power to be cut in the case of a malfunction.
This switch should be placed at a safe distance from the centrifuge, preferably in a different room from
that in which the centrifuge is situated.
Dangerous substances:
It is recommended tat the user takes all the necessary precautions when centrifuging substances known
for their toxicity or radioactivity or when they are contaminated by microbiological pathogens (notably
material of Risk Group II as identified in the “Biological Safety Manual for Laboratories” edited by the
World Health Organisation). See also chapter 6.2.3.
The phenomenon
This normal physical phenomenon appears when a mechanical force is applied and relaxed repeatedly.
The accumulation of alternate application and releasing of force (as in the number of runs) translates after a
given number of runs into a very rapid reduction in mechanical strength. The item cans no longer resist
the same force as at the start of its life. If the force is applied repetitively with same intensity, it will rupture.

It is this phenomenon of excessive fatigue which is used when repeatedly bending and unbending a piece
of wire or plastic in order to break-it.
Each centrifugation run is a fatigue run for the buckets.
Bucket lifetime
The number of runs after which identical items break having been submitted to the same stresses varies
enormously. For centrifuge buckets, several thousand runs could separate the rupture of 2 identical
buckets submitted to the same stresses. We are dealing with a "probability" situation. As the consequences
of a rupture are serious, it is necessary to limit the number of runs to the zone where the risk is considered
to be zero. It is recommended to replace accessories (rotors and buckets) after 16 000 runs (and
10 years). An exception to this is the NHK4B bucket which needs to be replaced after 9 000 runs.
Although the use of any rotors or buckets, other than those described in this manual, is not allowed on the
KR4i, it is important to study the example of the P6 (Cat. N° 11178112) and P4 (Cat. N° 11178074)
transfusion buckets for the KR4.22. For these calculations it has been considered that each run has been
carried out with a full load of blood bags.
It is absolutely essential that any one rotor and buckets are used uniquely with the same
centrifuge in order to be able to track the evolution of the number of runs of the complete
system (a run counter is fitted with each centrifuge).

Remarks :
The graphs show that "soft spin" runs such as for obtaining PRP and for platelets from the buffy coat (up to
1500 x g or about 2200 rpm) do not incur any fatigue. So the number of runs carried out at low speed is not
counted in the total for the calculation of the lifetime of the buckets.
6.5.3 .CORROSION
The above graphs have been established from tests carried out on samples of metal in perfect condition
and over a period of many hours.
As the useable lifetime of the buckets is several years, it is important to take into account the wear due to
corrosion, to which the buckets may have been submitted, independently of the fatigue phenomenon.
High voltage is present behind the panels of the KR4i. It is not necessary for the user to intervene inside the
machine. Only qualified techniciens are authorized to do this. Do not remove these panels unless the
centrifuge has been first disconnected from the power outlet.

- Never spin a rotor or its accessories at more than the maximum speed, indicated in chapter 3.
- Do not attempt to override the lid interlock assembly.
- Use only accessories designed for use in the KR4i.
- Load the rotor only in the recommended way (see 2.9 and 5.1) and do not spin higher than the speed
indicated on the rotor and accessories. As the centrifuge starts to spin, an improperly loaded rotor could
cause enough force to damage the drive shaft and the rotor chamber.
By the action of the turbulence created in the bowl a centrifuge is an aerosol generator. In the case of
products presenting a biological risk you must take suitable precautions to prevent or reduce this risk and
also train operators in the use of this instrument and its accessories in the context of their applications.
The following information should be given to all users including those persons using the equipment
infrequently :
- A ventilated centrifuge (without built-in refrigeration) permanently exhausts into the room air which has
passed through its bowl while a refrigerated (or thermostated) centrifuge contains a relatively sealed
bowl. Thus the risks from the spread of aerosols are minimised in a refrigerated centrifuge and
significant in a ventilated centrifuge.
- Aerosols are produced naturally when using uncapped tubes or bottles. They are also produced when a
container deforms or breaks provoking a smaller or greater sample leak. We draw your attention to the
use of tubes used in angle rotors. Even if the liquid level does not touch the cap when in a static state, it
could do so during centrifugation and leak. You should therefore follow the instructions of the tube
supplier concerning the fill level and sealing of the tube.

Once a biological risk is known or suspected, samples should be placed in a sealed container. Should
such containers not be available, sealed accessories should be used such as buckets or angle rotors
with sealed lids. N6BMT buckets (11178140) are certified by CAMR - Porton Down for their containment
efficiency. N6B buckets (11178138) are sealed if fitted with lid (11178216).
- Duration of aerosol presence : when aerosols are created in a centrifuge bowl they persist in the air for a
period of 3-5 minutes after the rotor stops moving. Indeed, the action of opening the lid itself provokes
the dispersion of aerosols in the environment.
- When an elevated risk of aerosols or of breakage is perceived the accessories should be handled using
gloves and opened in an environment ensuring the protection of the operator (safety cabinet, glove
chamber, wearing a mask ...), even if they are apparently sealed.

Daily cleaning is not required, except in the event of glass breakage or a large amount of spillage in the
rotor chamber or fairing.
If you wish to clean the control panel :
- Turn off the centrifuge,
- Clean with a moist sponge and a soap product and rinse with a moist sponge. Dry with a lint-free cloth.
To disinfect, if required, use 70% alcohol.
a) In the event of spillage between the chamber and the windshield, clean the rotor chamber with a
detergent and water only.
b) The rotors and buckets should be washed with warm water (at < 50°C) and a mild soap or detergent
(for example : a household detergent for handwashing).
Rinse the rotor thoroughly with (preferably distilled) water. Dry the rotor or bucket, and preferably with
hot compressed air store upside down.
c) The KR4i centrifuge is equipped with a drainage hose, attached to the rotor chamber to enable rinsing
the chamber when required.
To rinse and drain, operate the unit at 3000 rpm, at 20°C temperature for 1 minute.
The following preventive maintenance procedures are required for the KR4i centrifuge :
- Ensure that the positioning instructions, as described in chapter 2.5, are always carried out
(horizontability, anti-skid pads, castor positioning).
- Check the condition of all accessories (rotors, buckets, inserts, sealed lids) - see chapter 6.
- Check earth continuity protection (when undertaking any intervention inside the machine).
