La Société JOUAN garantit que cet appareil ne présente aucun défaut de matériau ni de montage au moment de
la sortie d’usine et remplacera ou réparera cet appareil s’il s’est avéré défectueux en utilisation normale ou au
cours d’une opération d’entretien pendant UN AN à partir de la date de livraison.
Notre obligation dans le cadre de cette garantie se limite à la réparation de l’appareil défectueux ou d’une pièce
quelconque de cet appareil lorsqu’elle est renvoyée, en port payé, à un atelier service autorisé ou à nos services
de SAINT-HERBLAIN.
Cette garantie sera considérée comme caduque si l’appareil a été faussé, mal entretenu ou négligé, mal utilisé
ou détérioré accidentellement.
Il n’existe aucune autre garantie explicite en dehors des clauses ci-dessus.

1 UTILISATION ET FONCTION
L’incubateur à CO2 est un appareil destiné à la culture cellulaire. Ses fonctions essentielles sont de
maintenir :
- le niveau de stérilité de la chambre,
- une température constante,
- une atmosphère enrichie en CO2,
- une atmosphère avec une humidité relative élevée.

La connection pour l’alimentation en CO2 et pour le contact d’alarme à distances est située à l’arrière du
compartiment technique.

2 PROCEDURE D’INSTALLATION
Compte-tenu du poids de l’appareil, son levage et son transport doivent être réalisés par un moyen
mécanisé (diable, transpalette) conforme à la réglementation en vigueur et manipulé par du personnel
ayant suivi la formation requise.
L’appareil devra être soutenu par sa partie inférieure. S’il doit être transporté sans sa palette, dans
un escalier par exemple, s’adresser à des professionnels de la manutention.
ATTENTION :
1. Ne pas soulever l’incubateur en le tenant par la porte extérieure !
2. Ne pas déplacer l’incubateur lorsque la jaquette est remplie d’eau.
Cet appareil est conçu pour être sûr dans les conditions de fonctionnement suivantes :
- Utilisation en intérieur
- Température : 18° à 30°C
- Humidité relative maximale de 80% pour les températures allant jusqu’à 22°C.
- Altitude maximale : 2000 m
Les performances maximales sont assurées dans la plage de température suivante : 20°C à 25°C.

A
Installer l’IGO150 à sa place définitive avant de remplir la jaquette d’eau pour limiter son poids pendant
son transport. Il peut être installé selon l’une des trois configurations suivantes :
1. Posé sur un plan de travail : le plan doit pouvoir supporter correctement le poids de l’appareil qui, une
fois rempli d’eau pèse 170 kg. La profondeur du plan doit être de 70 cm minimum pour ne pas gêner
l’alimentation en gaz.
2. Sur un socle.
3. Gerbé sur un autre incubateur.
Lorsque l’incubateur est posé sur le sol, utiliser la clé mixte 17/19 livrée avec l’appareil pour mettre
l’incubateur à niveau.

ATTENTION :
1. Vous dever utiliser des bouteilles de CO2 non-équipées de tube plongeur afin d’injecter du CO2
gazeux dans la chambre. Ce CO2 devra, de préférence, être de grade médical.
2. La bouteille de CO2 doit être équipée d’un détendeur et si possible d’un manomètre.
3. Ne pas exposer l’incubateur à une pression d’entrée supérieure à 3 bar, sous peine d’endommager le
système de mesure de CO2.
4. L’appareil est livré avec deux systèmes de raccordement à la source de CO2. Pour des raisons de
sécurité ne pas dépasser la pression maximale spécifiée.
Si la pression d’alimentation est comprise entre 0.8 bar et 1 bar maxi, vous pouvez utiliser le kit de
connexion basse pression : insérer l’embout cannelé et l’olive sur le filetage extérieur de l’incubateur.
Serrer l’écrou et et monter le tuyau sur la partie cannelée.
P = 1 bar maxi
Si la pression d’alimentation est comprise entre 1 bar et 3 bars, vous devez utiliser le kit de connexion
haute pression : insérer le tube rigide avec l’olive dans le filetage extérieur de l’incubateur et serrer
l’écrou.

REMARQUE : Un inverseur de bouteille automatique peut être inséré entre 2 bouteilles de CO2 permettant
d’assurer une plus grande autonomie en CO2.
ATTENTION : une fois les connextions fluidiques terminées, verifier qu’il n’y a pas de fuite au niveau des
raccords en effectuant un test à l’eau savonneuse au niveau des raccords ou en vérifiant qu’il n’y a pas de
chute de pression au niveau des manomètres quelques minutes après le branchement.
2.7. DECONTAMINATION
L’incubateur livré n’est pas stérile. Il convient de nettoyer la chambre et ses accessoires avec de l’alcool à
60 ou 70° avant l’utilisation. Voir le chapitre 6.1 pour le nettoyage et la décontamination de la chambre.
Retirer les plateaux et les echelles qui sont installés dans la chambre.
Avant de brancher l’incubateur, vérifier que le réseau secteur correspond bien à ses caractéristiques.
Pour cela le réseau secteur doit obligatoirement comporter une prise terre de bonne qualité.

L’énergie dissipée par l’agitation du milieu de culture provoque une élévation locale de température.
Nous conseillons dans ce cas, de réduire la température de consigne afin de tenir compte de ce
phénomène.
Le câble d’alimentation de l’agitateur est introduit dans la chambre à travers le passage prévu entre la
chambre et le compartiment technique. Le passage doit être obturé autour du câble sous peine de créer
une fuite permanente.

3 SPECIFICATIONS
Tension secteur :
En sortie d’usine, l’incubateur est équipé pour fonctionner soit en 120V - 60 Hz, soit en 230V - 50 Hz.
Consommation :
Max. 800VA (nominale 170W pour un fonctionnement à 37°C).
Indice de protection :
- IP2 : Appareil protégé contre les pénétrations de corps solide de plus de 12,5 mm de diamètre et contre
les chutes verticales de gouttes d’eau.

4.3.2 TELEALARME
La prise située en face arrière, permet à l’utilisateur de connecter un dispositif de surveillance (lampe,
alarme sonore, etc…).
Tension de commutation maximale : ≈ 230 V ; 6 A maximum.
L’apparition d’un défaut provoque :
- La fermeture d’un contact de relais, qui est connecté entre les broches 2 et 3 du connecteur.
- L’ouverture d’un contact de relais, qui est connecté entre les broches 2 et 1 du connecteur.
Il y a alarme dans les conditions suivantes :
- Incubateur hors tension, lorsqu’il est déconnecté du secteur,
- Panne alimentation interne (interrupteur général disjoncté ou panne d’alimentation électronique),
- Carte électronique défectueuse,
- Détection par le microprocesseur d’une défaillance des circuits électroniques (mesure température,
mesure de CO2, conversion analogique digitale, valeur des alimentations),
- Lorsque la valeur de CO2 dans la chambre est hors de l’intervalle programmé,
- Lorsque la valeur de température dans la chambre est hors de l’intervalle programmé,

Attention aux tensions électriques présentes dans le caisson de contrôle de l’incubateur CO2.
Ne jamais retirer le capot de ce caisson sans avoir débranché l’incubateur au préalable.
Le CO2 étant un gaz toxique fermer l’arrivée de CO2 à l’incubateur, au niveau du détendeur externe de
pression :
- Avant toute intervention de longue durée supposant l’ouverture de la chambre (nettoyage,
décontamination …).
- Avant toute intervention sur la fluidique, aussi bien dans la partie située entre la bouteille de CO2 et
l’incubateur qu’entre l’entrée de CO2 dans l’appareil et la chambre.
En cas de défaut sur le système de contrôle du chauffage, l’élévation de température des parties
accessibles n’excède pas +50°C.

6.1. NETTOYAGE
Essuyer puis nettoyer les plateaux chaque fois qu’un liquide a été renversé dessus (milieu de culture,
sang…).
Une fois par mois (ou tous les 2 - 3 mois suivant l’activité du laboratoire) :
- Plateaux, bac humidificateur et échelles : démonter (cf figure ci-dessous), nettoyer puis autoclaver
(121°C, 20 mn) ou stériliser dans un four (30 mn à 180°c ou 1 H à 170°C).
- Chambre : nettoyer les parois de la chambre avec un désinfectant de surface (alcool à 70°,
glutaraldéhyde basique à 2%) en prenant soin d’éliminer les croûtes qui ont pu se former.
Attention : ne pas utiliser de produits chlorés (eau de Javel).
- Filtres microbiologiques : les contrôler régulièrement. Les changer dès qu’ils commencent à jaunir et à
s’humidifier, ne pas oublier le filtre situé à l’arrière de l’appareil.

Périodiquement et au moins tous les six mois, contrôler l’état des filtres 0,22 mm. Si la face inférieure du
filtre est grisâtre, changer le filtre. (référence du filtre : 86000302)
La dérive du système de mesure du CO2 est extrêmement faible. Il est cependant conseillé de faire
vérifier la concordance entre la valeur affichée et le taux réel de CO2 dans la chambre grâce à un
analyseur chimique. Analyseur Fyrite, référence : 41320808.
Un orifice de prise d’échantillon est situé sur la vitre de la chambre, protégé par la porte extérieure qu’il
faut ouvrir entièrement pour le dégager.
ATTENTION : Ne pas omettre de replacer le bouchon de l’orifice sous peine de provoquer une fuite
permanente.
Le changement des fusibles ne doit pas être effectué par l’opérateur mais par un technicien agréé qui fera
un diagnostic avant de décider de leur remplacement.

GUARANTEE TERMS
JOUAN guarantees that this unit is free from defects in materials and workmanship when it leaves the factory, and
undertakes to replace or repair the unit if it proves defective in normal use or during servicing for a period of
ONE YEAR from the delivery.
Our liability under this guarantee is limited to repairing the defective unit or any part of the unit providing it is
sent, carriage paid, to an authorised service centre or the SAINT-HERBLAIN office.
This guarantee is invalidated if the unit is incorrectly used, poorly serviced or neglected, mis-used or
accidentally damaged.
There is no explicit guarantee other than as stated above.

The CO2 incubator is designed to reproduce, as closely as possible, the environmental conditions of living
cells. The essential functions of the incubator are to maintain:
- The sterility of the chamber
- A constant temperature
- An atmosphere with a fixed level of CO2
- An atmosphere with a high Relative Humidity.

2 INSTALLATION PROCEDURE
Because of the weight of the incubator, all lifting and transport must be carried out using proper handling
equipment (e.g.: fork lift or hand truck), that complies with current regulations. Only trained, skilled
technicians should install a CO2 Incubator.
The machine must be supported underneath. If it has to be transported without its pallet, for example
on staircase, professional handling assistance is required.
CAUTION :
1. Do not lift or hold the incubator by holding the outer door.
2. Do not move the incubator when the water jacket is filled.
Because of the weight and size of the incubator two people are needed to unpack. The weight of the empty
incubator is 70 kg.
1) Cut the 2 plastic belts (A).
2) Remove the cardboard cover (B).
3) Remove the cardboard stoppers at the 4 corners (C).
4) Remove the cardboard wrapping (D).
5) Take the incubator by the bottom and put it on the ground.
6) Open the incubator.
7) Remove the wedges used to immobilize the shelves.
8) Remove the accessory box.

2.4. INSTALLATION
Place the IGO 150 where it will be used before filling the water jacket. The IGO 150 can be installed in 3
configurations :
1. If placed on a bench, make sure the bench can withstand properly the weight of the instrument that is
about 170 kg when the jacket is filled with water. The depth of the bench must be at least 70 cm to allow
access to the CO2 inlet. See Chapter 5.2 for floor strength.
2. On a castor stand.
3. Stacked on another incubator.
When the incubator is installed directly on the floor, use the 17/19 key delivered with the instrument to
level the CO2 incubator.
1. Open the fill port of the chamber by removing the cap located on the left front top part of the chamber.
2. Insert a tube or a funnel less than 17 mm in diameter in the access port, leaving a gap for air to
evacuate from the chamber.
3. Fill in the water jacket with 91 liters of deionized or distilled water. Fill up the jacket up to 7 to 8 cm from
the access port.
CAUTION : do not use any chlorinated product or chlorinated water in the jacket
4. Put the cap back in place after filling to avoid contamination and to limit water evaporation.

CAUTIONS :
1. You must use CO2 tanks that are not fitted with a diving tube to inject CO2 gas into the chamber.
2. The CO2 tank must be fitted with a pressure regulator, preferably the tank should be fitted with a
pressure gauge.
3. For safety reasons, the tubing used to connect the CO2 tank to the incubators must be able to stand a
pressure of 10 bars. The connections are designed to stand this pressure except for the barbed nipple
which is designed for a low-pressure connection (a pressure lower than 1 bar).
4. The unit is delivered with 2 types of CO2 connection. For safety reasons, the inlet pressure used must
not exceed th specified value.
The pressure is between 0.8 bars and 1 bar : you can use the low-pressure assembly device : insert the
barbed nipple + ferrule into the thread outlet of the incubator, tighten with the nut and mount the tubing
on the fixed nipple.
P = 1 bar maximum
The pressure is between 1 bar and 3 bars : you have to use the high pressure assembling device :
insert the rigid tubing with the ferrule into the thread outlet of the incubator and tighten the nut.
P = 3 bars maximum

REMARK : An automatic pneumatic cylinder switch can be inserted between 2 CO2 tanks to insure a
continuous supply of CO2.
CAUTION: once the connections are made, check that the tubing does not leak and cannot escape from
the fittings, by applying a gas pressure while the unit is switched off.
The incubator is not sterile when delivered. Before operating the incubator you must clean and disinfect
the incubator. See chapter 6.1. for cleaning and decontamination.
Remember that in order to maintain the electrical safety standards related to protection of operators
against indirect contact, the supply of power to the instrument must be properly fused. A supply fitted with
a circuit breaker of the correct rating complies with this requirement.
1. Set the shelf supports in place by adjusting them against the left and the right wall of the incubator
2. The plastic screws must be at the top of the chamber.
3. The rubber pads must be at the bottom of the chamber.
4. Tighten the screws with a light torque.
5. Install the number of shelves required for your culture.
6. Put the water pan in the bottom of the chamber and fill it with about 3 liters of water.
7. Start the incubator by switching the main switch to the “on” position.
8. Open the CO2 supply valve.
When the incubator is switched on, the software version, the type of jacket, and the type of sensor
module are displayed :
- h0.5 means : water jacket, software version V0.5
- d0.5 means : dry jacket, software version V0.5
- U means ULC/6MM11C module
- A means ANDROS module
9. Program the various setpoints (see Chapter 4.2.)
The optimal operating conditions are attained 24 hours after turning on the incubator.

The temperature control system of the incubator is designed to work when the content of the incubator
does not generate any heat (the heat produced by the culture is negligible).
This temperature control system is very sensitive to heat to allow a good temperature control.
If you place an instrument inside the chamber that produces heat, you may have to adjust the temperature
setpoint of the incubator to get the media at the right temperature.
The access port for the cable is located at the upper right part of the chamber. The port opens into the
electronic compartment, another port is provided in the back of the electronic compartment to enable
access to the outside of the electronic compartment.

The incubator is designed to work either at 120V - 60 Hz or 230V - 50 Hz.

Maximum 800 VA (average 170 W functionning at 37°C in an ambiant temperature of 22°C).

Never perform maintenance that requires the electronic compartment to be opened, unless the incubator
is unplugged from the electrical power supply. Be aware of the hazard of electrical shock inside the
electronic compartment of the incubator. Never remove the metallic cover of the electronic compartment
without first disconnecting the incubator from the power outlet.
Before installing the CO2 incubator check to make sure the working floor space can support :
- 431 kg/m2 for a single chamber jacket filled up with water
- 454 kg/m2 for a single chamber filled with water placed on its castor stand
- 907 kg/m2 for 2 stacked incubators filled with water.
The CO2 is a toxic gas so please close the CO2 supply to the incubator, by closing the CO2 tank valve and
the pressure regulator valve in the following situation:
- When the chamber door will be open for an extended period of time (during cleaning or
decontamination).
- When servicing the gas control parts or systems, (connections with the CO2 tank and the incubator or
systems in the technical compartment of the unit).
In case of failure of the temperature control system, the temperature increase of the accessible parts will
not exceed 50°C.
Make sure to use only ultra pure water in the water pan. The use of tap water would rapidly deteriorate the
measuring system for CO2 due to the formation of calcium deposits.

6.1. CLEANING
Wipe and clean the shelves or the entire chamber carefully each time they become contaminated with
anything (medium, blood, etc.).
Monthly or every 2-3 months depending on the activity of the laboratory :
- Shelves, shelf supports, humidity pan : remove, clean and either autoclave (20 minutes at 121°C) or
sterilize in an oven (30 minutes at 180°C or 1 hour at 170°C)
- Shelf support removal : unscrew the plastic screws in the upper part of the brackets. Pull out the brackets.
Do not autoclave the rubber pads or the plastic screws.
- Culture chamber : clean the walls with a surface disinfectant (70% alcohol, 2% glutaraldehyde). Be
careful to remove any encrusted spots of dried medium. Avoid chlorinated solutions which might
damage the interior of the incubator.
- Microbiological filter : check the color regularly. Change them as soon as they become gray or slightly
wet. Do not forget to change the filter located at the back of the instrument.

6.2. FILTERS
Periodically, and at least every 6 months check the HEPA filters. If one of the sides is dirty, change the
filter.
The drift of the IR photometer is virtually impossible. However we recommend an annual check of the CO2
calibration of the sensor by using a FYRITE analyzer.
The manual checking port is located on the front panel. It is accessible only after opening the main door.
CAUTION : the manual checking port is blanked with a small cap. Don’t forget to put it back after use to
avoid a permanent leak of CO2.
6.5 FUSES
If a fuse fails in the electrical system an electrical fault has developed somewhere in the incubator. For this
reason, the operator should not change fuses. If a fuse fails, a qualified technician, electrician or a qualified
engineer should be used to diagnose and correct the fault before replacing the fuses.
